    Notes: Abstract Unlike the α and β polypeptides of the core light-harvesting complex (LH1) of Rhodobacter (Rb.) sphaeroides, the α and β polypeptides of the peripheral light-harvesting complex (LH2) of this organism will not form a subunit complex by in vitro reconstitution with bacteriochlorophyll. Guided by prior experiments with the LH1 β polypeptides of Rb. sphaeroides and Rhodospirillum rubrum, which defined a set of interactions required to stabilize the subunit complex, a series of mutations to the Rb. sphaeroides LH2 β polypeptide was prepared and studied to determine the minimal changes necessary to enable it to form a subunit-type complex. Three mutants were prepared: Arg at position −10 was changed to Asn (numbering is from the conserved His residue which is known to be coordinated to bacteriochlorophyll); Arg at position −10 and Thr at position +7 were changed to Asn and Arg, respectively; and Arg at position −10 was changed to Trp and the C-terminus from +4 to +10 was replaced with the amino acids found at the corresponding positions in the LH1 β polypeptide of Rb. sphaeroides. Only this last multiple mutant polypeptide formed subunit-type complexes in vitro. Thus, the importance of the C-terminal region, which encompasses conserved residues at positions +4, +6 and +7, is confirmed. Two mutants of the LH1 β polypeptide of Rb. sphaeroides were also constructed to further evaluate the interactions stabilizing the subunit complex and those necessary for oligomerization of subunits to form LH1 complexes. In one of these mutants, Trp at position −10 was changed to Arg, as found in LH2 at this position, and in the other His at position −18 was changed to Val. The results from these mutants allow us to conclude that the residue at the −10 position is unimportant in subunit formation or oligomerization, while the strictly conserved His at −18 is not required for subunit formation but is very important in oligomerization of subunits to form LH1.

    Notes: Summary A convenient miniassay for aflatoxin has been developed for cultures ofAspergillus flavus andA. parasiticus grown for 3–10 days in 10 ml of a coconut extract medium. The sensitivity of the assay, as measured by photofluorometry (365 nm maximum excitation; 445 nm maximum emission), is of the order of 0.01 μM (3.12 ng/ml) for aflatoxin B1 dissolved in aqueous iodine (0.26 mM). High performance liquid chromatography, monitored by fluorometric analysis of both an aflatoxin B1 standard and selected culture filtrates, confirmed the sensitivity of the assay and indicated specificity for iodine-enhanced fluorescence of aflatoxin in the coconut extract medium. Thin layer chromatography further confirmed the aflatoxin titers and the specificity for enhancement of aflatoxins B1 and G1 in culture filtrates.

    Notes: Summary Background: The body's nucleotide pool derives from three potential sources: de novo synthesis, salvage of preformednucleosides/bases or the diet. The relative contributions of these pathways of assimilation are poorly understood in vivo. Dietary nucleotides have been suggested to have beneficial effects an the development and repair of the gastrointestinal tract. Tissues with a rapid turnover, such as the gut and the immune system cells, may utilise preformed nucleotides (coming from the diet), in situation in which there is a high demand of nucleotides for nucleic acid synthesis. Therefore, nucleotides could be onsidered as conditionally essential nutrients. Aim of the work and methods: -Development of a method to measure synthesis de novo of RNA-purine nucleotides in Caco-2 cells, relying an the incorporation of 14C-glycine into the purine ring of the nucleotide. To establish the fractional synthesis rate of RNA purine nucleotides in Caco-2 cells, grown in culture medium containing different concentrations of glutamine, in the presence or abscence of added nucleotides. To investigate the degree to which tissue ribonucleosides are derived from the culture medium or from de novo synthesis in the presence of different concentrations of glutamine, using undifferentiated Caco-2 cells, stressed or not by the addition of IL-1β to the medium. Results and conclusions:The presence of high levels of glutamine in the culture medium is essential for cell proliferation (estimated by measurement of the fractional synthesis rate of purine nucleotides) and the presence of nucleotides cannot replace the glutaminedependence of Caco-2 cell proliferation. The incorporation or exogenous purine nucleotides into RNA of Caco-2 cells is rather limited, and it becomes important when cells are stressed by glutamine deprivation. Stress by addition of interleukin-1β resulted in the maintenance or the increase in de novo synthesised RNA-purine nucleotides, even in the presence of exogenous nucleotides. However, the addition of interleukin-1β to the culture medium led to an enhanced salvage of preformed pyrimidine nucleotides for nucleic acid synthesis when glutamine was present in the medium at a concentration of 0.5 mmol/L.

    Notes: Abstract: Currently, designers are faced with searching through a ‘sea’ of on-line knowledge to support their decision making activities. This paper describes WebCADET, which is a reimplementation of the stand-alone CADET – a Knowledge-Based System (KBS) for product design evaluation. WebCADET aims to provide effective and efficient support for designers during their searches for design knowledge. WebCADET uses the ‘AI as text’ approach, where KBSs can be seen as a medium to facilitate the communication of design knowledge between designers. The development of WebCADET to include practical support via World Wide Web-based functionality, which illustrates the potential of the ‘AI as text’ approach, is described in the paper.

    Notes: Abstract This paper describes the fundamental modeling approaches adopted for crack nucleation and propagation in a software system that is specifically designed to simulate problems with evolutionary geometry. Only the topological and geometrical aspects of crack modeling, and how these aspects affect the database representation in the system, are addressed in the present discussion. The following are the innovative features of the present crack modeling approach: (a) crack simulation is done with a true geometric representation of the structure, via solid modeling; (b) crack modeling relies on the sophisticated, topology-based data structure of this system to support linkage to the solid model, fast interaction and accurate representation of evolving flaw shapes; (c) the system provides the ability to specify flaws of arbitrary shape (including non-planar flaws), size and orientation at arbitrary locations in the geometric model; (d) the flaw is specified at the desired location in the actual structure geometry, rather than at a location in the mesh; (e) the system uses all its automatic and local remeshing capabilities for the simulation of flaw initiation and growth.

    Notes: Abstract Lasers are an important tool in the field of muon physics. A new application of lasers, namely producing polarized muonic atoms, is the subject of a new program at LAMPF. One technique already demonstrated is stopping unpolarized muons in a laser polarized3He target. A more promising idea is to polarize neutral muonic helium by collisions with laser polrized Rb vapor. These methods for producing polarized muonic helium will be useful for measuring the spin dependence of nuclear muon capture and for determining the induced pseudoscalar coupling.

    Notes: Abstract Biological macromolecules have unique rheological properties that distinguish them from common synthetic polymers. Among these, fibrin has been studied extensively to understand the basic mechanisms of viscoelasticity as well as molecular mechanisms of coagulation disorders. One aspect of fibrin gel rheology that is not observed in most polymeric systems is strain hardening: an increase in shear modulus at strain amplitudes above 10%. Fibrin clots and plasma clots devoid of platelets exhibit shear moduli at strains of approximately 50% that are as much as 20 times the moduli at small strains. The strain hardening of fibrin gels was eliminated by the addition of platelets, which caused a large increase in shear storage modulus in the low strain linear viscoelastic limit. The reduction in strain hardening may result from fibrin strand retraction which occurs when platelets become activated. This interpretation is in agreement with recent theoretical treatments of semi-flexible polymer network viscoelasticity.
