    Description / Table of Contents: Zusammenfassung Zur Untersuchung möglicher Struktur-Wirkungs-Beziehungen in der Reihe der verbrückten Thalidomid-Analoga wurden 36 offenkettige und verkürzte Abkömmlinge der Testsubstanz „K 2004” (2) synthetisiert. Die pharmakologische Untersuchung der hergestellten Verbindungen ergab in keinem Falle eine signifikante sedative Aktivität. Dies legt den Schluß nahe, daß die intakte Glutarimid-Struktur bei1 und2 für das Zustandekommen der schlafinduzierenden Wirkung unentbehrlich ist.
    Notes: Abstract For the evaluation of possible structure-activity relationships in the series of bridged thalidomide analogues 36 openchain and shortended congeners of the test substance “K 2004” (2) have been synthesized. The pharmacological evaluation of these compounds revealed in no case any significant sedative activity. This leads to the conclusion that the intact glutarimide structure must be essential for the sleep-inducing activity of1 and2.

    Notes: Abstract The cystic fibrosis transmembrane conductance regulator (CFTR) gene of 110 cystic fibrosis (CF) patients from the south-west of Germany was screened for 12 different mutations. This analysis resulted in an identification of 79% of all CF mutations and a complete genotype in 66% of the families. The most common mutation found was ΔF508 (67%). Another 5 mutations accounted for a further 12.5% (4% G542X; 3% R553X; 3% N1303K; 2% 1717-1 G→A; 0.5% G551D) whereas 6 mutations (R117H, A455E, ΔI507, S549I, S549N, and R1162X) were not found. Fifty-four (49%) patients were AF508 homozygotes and 18 (16.5%) were compound heterozygotes for ΔF508 and one of the rarer mutations. These frequencies differ slightly from those found in the north of Germany and considerably from those reported from the south of Europe, which seems to be consistent with a north to south decline of the relative abundance of ΔF508. Two patients, age 6 and 25 years, were compound heterozygotes for G542X and N1303K. The clinical features of the 6 year old were characterised by severe gastrointestinal and as yet only mild pulmonary complications whereas the 25 year old manifested severe pulmonary and gastrointestinal symptoms indicating that the N1303K mutation of the C-terminal CFTR nucleotide binding fold significantly impairs protein function in both the pancreas and the lungs.

    Notes: Abstract The geographic distribution and origin of CFTR mutations in Germany was evaluated in 658 three-generation families with cystic fibrosis (CF). Fifty different mutations were detected on 1305 parental CF chromosomes from 22 European countries and overseas. The major mutation ΔF508 was identified on 71.5% of all CF chromosomes, followed by R553X (1.8%), N1303K (1.3%), G542X (1.1%), G551D (0.8%) and R347P (0.8%). According to the grandparents’ birthplace, 74% of CF chromosomes had their origin in Germany; the ΔF508 percentage was 77%, 75%, 70% and 62% in northern, southern, western and eastern Germany, respectively. Ten or more mutant alleles in the investigated CF gene pool originated from Austria, the Czech Republic, Poland, Russia, Turkey and the Ukraine. This widespread geographic origin of CFTR mutations in today’s Germany reflects the many demographic changes and migrations in Central Europe during the 20th century.

    Notes: Abstract A protein closely resembling the purple membrane protein pre-exists in the cell membrane of H. halobium prior to the appearance of functional bacteriorhodopsin. It is associated with a differentiated membranous structure which has been isolated on a sucrose gradient and appears to be a precursor of the purple membrane. The identity of the precursor protein as a form of the purple membrane protein was established in different ways: (1) The cell proteins were labelled in vivo with 14C-proline during dark aerobic growth, the label was “chased”, and the cells transferred to the illuminated near-anaerobic conditions under which purple membrane is optimally synthesised (induction conditions). Cell lysates were fractionated on sucrose gradients at different times after induction. Label first found in the precursor fraction appeared within 24 h in the purple membrane fraction. (2) SDS-urea-acrylamide gel electrophoresis of the purple membrane protein and the precursor showed only one protein band whose migration coincided with that of the purple membrane band. (3) The amino-acid analysis of the purified precursor was very similar to that of the purple membrane. The absorption spectrum of the precursor showed little of the characteristic absorption of bacteriorhodopsin at 570 nm. A major band appears at 412 nm, the exact nature of which is not known. The difference spectrum (reduced versus oxidised) of a purified fraction showed only traces of cytochrome. Thin-layer chromatography of an acetone-soluble lipid extract indicated the presence of retinal and Β-carotene. Cells grown in the presence of nicotine did not develop purple membrane after induction: the species absorbing at 412 nm was much less abundant than in non-inhibited cells, but a new fraction was present with a sharp peak at 345 nm consisting mainly of lycopene.

    Notes: Abstract Pronase treatment of aqueous suspensions of purple membrane fragments from H. halobium leads to the cleavage of bacteriorhodopsin. The protein fragments remaining in the membrane after treatment with relatively small concentrations of enzyme (2% w/w) in normal daylight range in molecular weight from 20,000-21,000 daltons, indicating that cleavage occurs mainly near the extremities of the protein chain. At higher enzyme concentrations the relative amounts of protein fragments having smaller molecular weight increase. Generally, the relative loss of retinal chromophore is larger than that of protein and thus the retinal binding site seems to be located near one of the chain ends that is cleaved off by enzyme. Irradiation with white light during the time of proteolysis (at both low and high enzyme concentrations) results in extensive cleavage, so that under certain conditions no high molecular weight components can be detected in SDS-polyacrylamide gels. It, therefore, appears that parts of the bacteriorhodopsin chain become more exposed to enzyme digestion when the purple membrane is illuminated. Enzyme treated aqueous purple membrane fragment suspensions still show photocycle activity. The main consequence of proteolysis is a pronounced appearance of biphasicity in the decay of M412 and the regeneration of bR570. Simultaneously the yield of O660 is reduced. As with untreated purple membrane, the correlation between the rates of decay of M412 and regeneration of bR570 is greatest when the yield of O660 is lowest.

    Notes: Abstract We have demonstrated the feasibility of determining isotope ratios in very small samples of osmium by deuteron activation in the Livermore Tandem Van de Graaff accelerator. The method involves determination of cross-section ratios by using several osmium isotope mixtures carefully prepared from electromagnetically-enriched osmium isotopes. The accuracy of this method was checked by a determination of the isotope ratios in naturally-occurring osmium. Our analysis demonstrated that an osmium specimen removed from a sample of commercial grade perrhenic acid contained only 0.4% natural osmium, the rest being radiogenic187Os.

    Notes: Abstract We study restrictions for the standard model coming from triviality. The answer depends on the embedding scale Λ where new physics appears. We obtain upper bounds for the Higgs mass and for the top mass. Contrary to some papers we find larger ranges of allowed masses for plausible embedding scales. The upper bound for the Higgs mass decreases with Λ. The maximum Higgs mass appears to be ≃1 TeV at Λ≃1 TeV.

    Notes: Abstract A methodology for regional application of forest simulation models has been developed as part of an assessment of possible climate change impacts in the Federal state of Brandenburg (Germany). Here we report on the application of a forest gap model to analyse the impacts of climate change on species composition and productivity of natural and managed forests in Brandenburg using a statistical method for the development of climate scenarios. The forest model was linked to a GIS that includes soil and groundwater table maps, as well as gridded climate data with a resolution of 10 × 10 km and simulated a steady-state species composition which was classified into forest types based on the biomass distribution between species. Different climate scenarios were used to assess the sensitivity of species composition to climate change. The simulated forest distribution patterns for current climate were compared with a map of Potential Natural Vegetation (PNV) of Brandenburg. In order to analyse the possible consequences of climate change on forest management, we used forest inventory data to initialize the model with representative forest stands. Simulation experiments with two different management strategies indicated how forest management could respond to the projected impacts of climate change. The combination of regional analysis of natural forest dynamics under climate change with simulation experiments for managed forests outlines possible trends for the forest resources. The implications of the results are discussed, emphasizing the regional differences in environmental risks and the adaptation potentials of forestry in Brandenburg.
