    Notes: A plasmid stabilization system, active in high copy-number plasmids, was cloned from the large resident plasmid, pSLT, of Salmonella typhimurium. The ytl2 gene, together with a 249-bp region (termed incR) downstream of the gene, imparted 〉104-fold stability to a pBR322-based plasmid. The ytl2-incR region was then used to stabilize a recombinant plasmid carrying the human epidermal growth factor gene (with the Escherichia coli K-12 ompA signal sequence), behind the lacUV5 promoter. In shake flask tests to optimize expression of human epidermal growth factor, loss of recombinant plasmid was 〈1% when growth (both before and after induction with isopropyl-β-d-galactopyranoside) took place even in the absence of antibiotic selection, and the specific activity of secreted human epidermal growth factor was ca 20 μg per 108 cells at harvest, compared to a figure of ca 3 μg per 108 cells when a comparable plasmid, but devoid of the ytl2-incR region, was employed, as outgrowth of plasmid-free cells after induction severely compromised the specific activity of the secreted product.

    Description / Table of Contents: Resume Deux modèles de description de la rupture par fissuration des matériaux à base de ciment sont ici étudiés: celui de la ‘bande de fissuration’ et celui de la ‘fissuration fictive’. Les deux types d’analyse sont très proches mais on montre cependant que le modèle de ‘fissure fictive’ peut se ramener à une méthode de superposition de trois facteurs d’intensité de contrainte dont l’utilisation requiert la connaissance d’expressions analytiques. Cette méthode se révèle donner des résultats très similaires à ceux obtenus par éléments finis avec la ‘fissure fictive’ et présente l’avantagè de pouvoir être programmée sur un ordinateur de type P.C. On démontre la capacité de prédiction du modèle de fissuration fictive par comparaison de la courbe expérimentale charge/flèche d’une petite éprouvette de mortier entaillée avec la courbe théorique calculée à partir des données obtenues sur des poutres plus grandes. On suggère que la méthode d’essai RILEM, peur la détermination de l’énergie de rupture, soit aussi utilisée afin d’établir une relation bilinéaire contrainte-déplacement pour la phase de radoucissement qui apparaît dans la zone de fissuration.
    Notes: Abstract The crack band and the fictitious crack line models of the fracture process zone in cementitious materials are discussed. The two methods are quite similar. However, the fictitious crack model is shown to lend itself to a simple K-superposition method which makes use of known analytical expressions. This method is shown to give very similar results to a finite-element analysis of the fictitious crack model and has the advantage that it can be programmed for a personal computer. The predictive capability of the fictitious crack model is demonstrated by comparison of the experimental load-deflection curve for a small notch bend mortar specimen with a theoretical curve calculated from data obtained from larger beams. It is suggested that the RILEM test method for the determination of the fracture energy should be used also to establish a bilinear stress-displacement relationship for the strain-softening of the fracture process zone.

    Notes: Summary Treated herein is an elastica under a point load. One end of the elastica is fully restrained against translation, and elastically restrained against rotation, while the other end portion is allowed to slide over a friction support. The considered elastica problem belongs to the class of large-deflection beam problems with variable deformed arc-lengths between the supports. To solve the governing nonlinear differential equation together with the boundary conditions, the elliptic integral method has been used. The method yields closed-form solutions that are expressed in a set of transcendental equations in terms of elliptic integrals. Using an iterative scheme, pertinent bending results are computed for different values of coefficient of friction, elastic rotational spring constant and loading position, so that their effects may be examined. Also, these accurate solutions provide useful reference sources for checking the convergence, accuracy and validity of results obtained from numerical methods and software for large deflection beam analysis. It is interesting to note that this class of elastica problem may have two equilibrium states; a stable one and an unstable one.

    Description / Table of Contents: Sommario Nel lavoro viene introdotto il concetto di simmetria per la descrizione del comportamento meccanico dei materiali. Per ogni materiale esistono infinite descrizioni per il suo comportamento mecanico in cui non e' necessario che i gruppi di simmetria siano gli stessi. E' stato provato che ogni descrizione anisotropa puo' essere trasformata in una isotropa od in un'altra anisotropa il cui gruppo di simmetria sia un po' piu' ampio diquello originale. Vengono introdotti i concetti di configurazione di riferimento, stato di riferimento ed elemento di riferimento di una deserizione. Si riporta anche il methodo per calcolare l'anisotropia indotta dalla deformazione.
    Notes: Abstract In this paper the symmetry concept is introduced for descriptions of the mechanical behaviour of materials. For any material there exist infinite descriptions for its mechanical behaviour whose symmetry groups need not be the same. It has been proved that any anisotropic description can be transformed into an isotropic or another anisotropic description whose symmetry group is a little larger than the original one. The concepts of reference configuration, reference state and reference element are introduced for a deseription. The method to calculate deformation induced anisotropy is also provided.

    Notes: Abstract A copper [Cu(II)]-accumulating strain, Pseudomonas putida II-11, isolated from electroplating effluent removed a significantly high amount of Cu(II) from growth medium and buffer. A laboratory-scale fixed bed reactor with cells of P. putida II-11 immobilized in polyacrylamide gel was constructed. The adsorption of Cu(II) by the immobilized cells was pH-dependent. Maximum removal of Cu(II) by the immobilized cells was at pH 8.0. The presence of Cr(IV), Ni(II) and Zn(II) did not significantly inhibit Cu(II) uptake whereas the presence of Pb(II) reduced Cu(II) uptake by fivefold. The presence of borate, carbonate, chloride and sulphate did not significantly inhibit Cu(II) uptake. The Cu(II) removal capacity of the bioreactor with immobilized cells did not change significantly when operated at retention times greater than 3 min. More than 90% of Cu(II) adsorbed on immobilized cells could be recovered by eluting with 0.1 m HCl. The bioreactor could be used for at least five loading-elution cycles without loss of Cu(II) removal capacity. The feasibility of using this bioreactor to remove and recover Cu(II) from electroplating effluent is discussed.

    Notes: Abstract The effect of space flight on the production of the antibiotic monorden on two types of agar media, T8 and PG, by Humicola fuscoatra WC5157 was examined on board the US Space Shuttle mission STS-77 in May 1996. Paired space-flight and ground control samples were prepared using identical hardware, protocol, media, and inoculum. Inoculation occurred simultaneously for both groups 2.5 h after launch. The flight and ground samples were allowed to grow for the entire 10-day mission in a dark, thermally controlled (22 °C) environment. Post-flight HPLC analysis of the flight and ground sample extracts indicated that the production of monorden by H. fuscoatra WC5157 in the flight samples was higher than in the ground samples in both agar media. In the T8 medium, the production of monorden in the flight and ground samples was 11.6 ± 3.5 μg and 8.9 ± 1.1 μg respectively (30% increase). In the PG medium, the production of monorden in the flight and ground samples was 23.8 ± 3.3 μg and 8.2 ± 2.2 μg respectively (190% increase). The production of monorden in the flight and ground control samples was confirmed by HPLC-MS analysis.

    Notes: Abstract A new integral equation method for the analysis of the interactions between cracks and elastic inclusions embedded in a two-dimensional, linearly elastic, isotropic infinite medium subjected to in-plane force is presented. By distributing dislocations along the crack lines and forces along the matrix-inclusion interfaces, a set of coupled integral equations is obtained. The discretization procedure of the integrals involved is discussed and the relations between the stress intensity factors and the values of the dislocation functions at the respective crack tips are derived. Several sample problems are presented in order to determine the versatility and the accuracy of this approach.

    Notes: Abstract  This paper presents a three-dimensional elasticity solution to the free vibration problem of thick cylindrical shell panels of rectangular planform. The natural frequencies and corresponding mode shapes were obtained for thick cylindrical shell panels and detailed parametric investigations were carried out. Comparisons were also made with corresponding finite element simulation results. To validate the accuracy of the results as well as the stability of the present methodology, comprehensive convergence studies were also carried out. Further comparisons of present results were made with existing experimental and numerical results (classical, first-order and higher-order shell theories) available in open literature and the validity and range of applicability of the various shell theories examined.
