    Notes: Summary The relationships between growth, cephamycin production and isopenicillin N synthetase (IPNS) activity in cultures of Streptomyces clavuligerus were examined to establish conditions that optimize the yield and specific activity of the enzyme. Unexpectedly for a secondary metabolic pathway component, IPNS was synthesized preferentially during rapid growth and reached its maximum specific activity in cultures supplied with readily assimilated sources of nitrogen. The activity decreased sharply as cultures entered stationary phase. On the other hand, comparisons of growth and antibiotic production on a range of carbon and nitrogen sources as well as measurements of IPNS activity in chemostat cultures implicated “catabolite repression”, a mechanism usually associated with separation of trophophase and idiophase activities, as an important factor in controlling expression of the secondary metabolic pathway. An explanation for the timing of IPNS biosynthesis is suggested.

    Notes: Summary Canopy temperatures of wheat, barley, rape and perennial rye grass crops, grown under temperate humid climatic conditions at different irrigation regimes were measured during two growing seasons, 1986 and 1987, by determining the emission of radiation in the wavelength interval 8〈λ〈14 μm. Global radiation, net radiation, air temperature, relative humidity and wind speed were measured simultaneously. The canopy temperature of the crops either fully irrigated or under water stress fluctuated up to 6 °C within a few minutes in response to rapid changes in global radiation. At high level of global radiation (800–1000 W m−2) canopy-air temperature differences up to 8 °C were measured whereas at low level of global radiation (100–200 W m−2) canopy-air temperature differences were found to approach zero or become negative even at severe crop water stress. Canopy temperature differences between water stressed and fully irrigated crops up to 6 °C were measured under conditions of high evaporative demand whereas under conditions of low evaporative demand canopy temperature differences between water stressed and fully irrigated crops approached zero even at severe crop water stress. For each crop the lower base line, i.e. the relationship between canopy-air temperature difference and vapor pressure deficit for a fully irrigated crop, was estimated by linear regression. In most cases a poor correlation was obtained which is attributed to considerable temporal variability in global radiation and wind speed and to the narrow range of prevailing values of vapor pressure deficit. However, from the base line for rape and barley it was possible to calculate apparent values of the aerodynamic resistance and the crop resistance which were of the same order of magnitude as those found for other crops by using this method under more arid climatic conditions.

    Notes: Abstract Recombinant isopenicillin N synthase fromStreptomyces clavuligerus was produced in the form of inactive inclusion bodies inEscherichia coli. These inclusion bodies were solubilized by treatment with 5 M urea under reducing conditions. Optimization of refolding conditions to recover active isopenicillin N synthase indicated that a dialysis procedure carried out at a protein concentration of about 1.0 mg ml−1 gave maximal recovery of active isopenicillin N synthase. Solubilized isopenicillin N synthase of more than 95% purity was obtained by passing this material through a DEAE-Trisacryl ion exchange column. Expression studies conducted at different temperatures indicated that isopenicillin N synthase was produced predominantly in a soluble, active form when expression was conducted at 20°C, and accounted for about 20% of the total soluble protein. This high-level production facilitated the purification of soluble isopenicillin N synthase to near homogeneity in four steps. Characterization of the purified soluble and solubilized isopenicillin N synthase revealed that they are very similar.
