    Notizen: Abstract The genome of Bradyrhizobium japonicum and B. elkanii contains multiple copies of the repeated DNA sequence RSα. A collection of 18 B. japonicum, 4 B. elkanii and 72 other bacterial strains was screened by polymerase chain reaction (PCR) using a pair of primers specific for RSα. Only strains of B. japonicum and B. elkanii gave the predicted amplification product. Restriction analysis of PCR products obtained from different strains of B. japonicum showed that the RSα sequence was generally conserved. The usefulness of RSα as a specific probe for Bradyrhizobium strains capable of nodulating soybean was also demonstrated.

    Notizen: Abstract A manganese-containing superoxide dismutase (EC 1.15.1.1) was purified to homogeneity from a higher plant for the first time. The enzyme was isolated fromPisum sativum leaf extracts by thermal fractionation, ammonium sulfate salting out, ion-exchange and gel-filtration column chromatography, and preparative polyacrylamide gel electrophoresis. Pure manganese superoxide dismutase had a specific activity of about 3,000 U mg-1 and was purified 215-fold, with a yield of 1.2 mg enzyme per kg whole leaf. The manganese superoxide dismutase had a molecular weight of 94,000 and contained one g-atom of Mn per mol of enzyme. No iron and copper were detected. Activity reconstitution experiments with the pure enzyme ruled out the possibility of a manganese loss during the purification procedure. The stability of manganese superoxide dismutase at-20°C, 4°C, 25°C, 50°C, and 60°C was studied, and the enzyme was found more labile at high temperatures than bacterial manganese superoxide dismutases and iron superoxide dismutases from an algal and bacterial origin.

    Notizen: Summary Metalloenzymes as biochemical markers of nutritional status have been studied for 30 years since Brown and Hendricks proposed the use of enzyme activities as indicators for the diagnosis of micronutrient deficiencies. Enzyme activities can be used in other lines of research, such as the characterization of particular reactions of genotypes to nutritive imbalances. Catalase, peroxidase and superoxide dismutase activities were assayed in soybean cultivars growing in nutrient solutions with different Fe/Mn ratios. Considerable differences were observed between cultivars. Relationships between growth and enzyme activity were also studied. Although more information is required, our results provide an introduction to the knowledge of the mechanisms developed by genotypes in response to Fe−Mn imbalances.
