    Notes: Summary Net H+ influx measurements on the giant internodal cells of the alga,Nitella clavata, have been made by transferring the cells to solution of low pH, 4.5 to 4.7, and measuring the pH changes or the amount of acid addition required to maintain the low pH. The latter results are in very good agreement with those of Kitasato (1968) and thus support his hypothesis that theNitella membrane is very permeable to H+. Other results indicate or suggest that the internal pH is changed only slightly under these conditions and that the membrane may have a rather large capacity to adsorb H+ on the external surface.

    Notes: Abstract We present a rough-cut analysis tool that quickly determines a few potential cost-effective designs at the initial design stage of flexible assembly systems (FASs) prior to a detailed analysis such as simulation. It uses quantitative methods for selecting and configuring the components of an FAS suitable for medium to high volumes of several similar products. The system is organized as a series of assembly stations linked with an automated material-handling system moving parts in a unidirectional flow. Each station consists of a single machine or of identical parallel machines. The methods exploit the ability of flexible hardware to switch almost instantaneously from product to product. Our approach is particularly suitable where the product mix is expected to be stable, since we combine the hardware-configuration phase with the task-allocation phase. For the required volume of products, we use integer programming to select the number of stations and the number of machines at each station and to allocate tasks to stations. We use queueing network analysis, which takes into account the mean and variance of processing times among different products to determine the necessary capacity of the material-handling system. We iterate between the two analyses to find the combined solution with the lowest costs. Work-in-process costs are also included in the analysis. Computational results are presented.

    Notes: An amorphous monomeric glass is defined as a mixture of compatible organic monomeric molecules with an infinitely low crystallization rate under the most favorable conditions. These mixtures can be formed in a one-part reaction of a multifunctional nucleus with a mixture of substituents. The “noncrystallizability” of the mixture is controlled by the structural dissymmetry of the nucleus and/or the substituents and the number of components making up the mixture. We have developed an equation to calculate the number of components in a given mixture, knowing the structure and functionality of the nucleus and the number of substituents. An HPLC characterization method was developed and used to investigate the effects of various reaction conditions on the component concentration distributions and physical properties of the resulting materials. A dissymmetry number defined as the total weight percent of odd-substituted components is believed to be a potential measure of the “noncrystallizability” of the mixture. Physical properties such as glass-transition temperature can be dramatically increased through controlled oligomerization at the price of a moderate increase in melt viscosity. The organic monomeric glasses, like amorphous polymers, have good film-forming properties. However, unlike polymers, they display extremely low melt-viscosities and large positive entropy-of-mixing values and can be ground easily into extremely small particles. These properties make them ideal for certain applications where compatibility, melt-flow, and small particle size are important.

    Notes: A 1-l. fermentor was designed and tested for use as a tissue-culture vessel. It features a temperature control device, impeller agitation without the necessity of a shaft seal, and a means of measuring, recording, and controlling both pH and oxidation-reduction potential (ORP). Tests have shown the ORP to change fairly rapidly with impeller speed variations under conditions of a continuous carbon dioxide-air overlay. Working with strain L mouse fibroblasts (Earle), cell counts of more than 1.25 ×106 ml., without centrifugation and medium renewal, were achieved, and cell counts were maintained above 1 ×106 for more than 30 hr. With the vessel studied, pH control was ±0.05, the ORP control was ±10 mV. Controlled environments for tissue-cell metabolic studies are entirely feasible with this system.

    Notes: Studies on the reaction kinetics and chromatographic properties of detergent-dispersed adenylate cyclase are described. Detergent-dispersed enzyme was prepared from whole rat cerebellum and from partially purified plasma membranes from rat liver.Data were simulated to fit kinetic models for which an inhibitor is added in constant proportion to the variable substrate. Models were chosen to distinguish whether the adenylate cyclase reaction may be controlled by an inhibitory action of free ATP-4 (or HATP-3) or by a stimulatory action of free divalent cations. The various kinetic models were then tested with the dispersed brain adenylate cyclase with both Mg++ and Mn++ and in two different buffer systems. The experimental data indicate that this enzyme has a distinct cation binding site, but exhibits no significant inhibition by HATP-3 or ATP-4.The detergent-dispersed adenylate cyclase both from liver plasma membranes and from brain have been chromatographed on anion exchange material and have been chromatographed on anion exchange material and have been subjected to gel filtration. The presence of detergent was required for elution of cyclase activity from DEAE-Sephadex but was not required when DEAE-agarose was used. Dispersed brain cyclase was also chromatographed on agarose-NH(CH2)3 NH(CH2)3-NH2 which exhibits both ionic and hydrophobic properties. Fifty percent of the applied activity was recovered with a fivefold increase in specific activity. The data suggest that the relative effectiveness of a given chromatographic procedure for detergent-dispersed adenylate cyclase may reflect the in fluence of both hydrophobic and ionic factors.

    Notes: This paper presents a reliable technique for the preparation of radioautographic specimens for study by high-voltage electron microscopy. Tissues are fixed by ventricular perfusion of aldehydes, sectioned, collected on slides, and coated with nuclear track emulsion using a coating machine. Sections are developed after 17 weeks' exposure in either undiluted Microdol-X or diluted D-19 developer, fixed in 25% sodium thiosulfate, and stained through the emulsion with uranyl acetate in absolute methanol and aqueous lead citrate. The technique produces sections of even thickness and staining density with negligible numbers of background grains and non-specific labeling. In this report, the efficacy of this technique for the study of the periosteal surface of demineralized alveolar bone specimens is demonstrated.
