    Notes: Good-quality high-Tc YBa2Cu3O7-x thin films deposited by the laser ablation method were investigated by Raman spectrosocpy. The vibrational spectra of thin films of YBa2Cu3O7-x at various laser intensities and with different polarizations were measured. The Raman modes change on increasing the laser power and in general such changes are reversible. However, as the laser intensity is increased above a certain value, dramatic changes occur in the Raman peaks, as a result of damage to the sample.

    Notes: A grand-scale mass spectrometer with high mass resolution and high transmission was employed for the analysis of haemoglobin variant. Two variants were isolated from a haemolysate by chromatography. Secondary ion mass spectrometry of complex peptide mixtures derived from these variants precisely determined the molecular weight of abnormal peptides. The molecular weight, 2857.4 and 2858.4, indicated the amino acid substitutions of asparagine and aspartic acid, respectively, for lysine at position 82 of β globin chain. The mutations had been reported in haemoglobin Providence.

    Notes: Basic expressions are derived for both the retention time and the effective separation factor in serially coupled GC columns. The retention time is determined by two main parameters. The first is the fractional time spent by an unretarded solute in each column which, in turn, is determined by the relative column lengths and flow velocities through each column. The second parameter is the relative mass distribution coefficient of a particular solute in each column; a variable that can be adjusted by changing the relative temperatures of the columns. The expression for the effective separation factor relates the measured separation factor for the series combination to the separation factors on the individual columns, the fractional time spent by an unretarded peak in each column, as well as the relative values of the mass distribution coefficients of a particular solute on the different columns.

    Notes: The real plate number concept is interpreted in terms of the fundamental properties of a chromatograph. It is shown that the linear relationship between the total peak width at half height, (b0.5)T, t, and k is only an approximation of the more general linear relationship between (b0.5)2T, t and (1 + k)2. In the case where the relationship between (b0.5)T, t and k approaches linearity, it is concluded, firstly, that b0 = a, i. e., b0 is a column property and independent of extra-column phenomena and, secondly, that nreal=ntheor. When the relationship becomes non-linear, b0 is determined partly by extra-column phenomena, but not in terms of a useful relationship. It is concluded that it is unnecessary to introduce the parameters a, b0 and nreal, since they all have counterparts in the established theory of chromatography. These counterparts prove to be equally useful in the practical analysis of chromatographic systems.

    Notes: The linearity of the relationship between σT,t2 and (1 + k)2 is studied with the view of using it for the evaluation of GC equipment. Based on appropriate analytical expressions it is concluded that in general this relationship is not linear and that practical procedures for column evaluation should be developed that take this non-linearity into account.

    Notes: Capillary columns in gas chromatography, first proposed by Golay [1,2], have become well established [3] which have extremely high column efficiencies and separabilities. Everyone could imagine it to be a great tool if such a kind of high efficiency column could be established in liquid chromatography (LC). Microcapillary liquid chromatography (MCLC) may be one way of achieving high-efficiency in terms of theoretical plates. Few papers were published in this field before 1976 [4-6], but more recently, after 1977 [7-15], this field has become one of topical interest in liquid chromatography. Temporarily there are two microcapillary liquid chromatography, that is: „open-tubular“ [7,8,10-15] and „packed“ [9] MCLC. In this presentation we focus on open-tubular microcapillary liquid chromatography and a review contemporary developments of ours and others.

    Notes: The electron impact mass spectra of twenty acetates and methylates of spirostanol and related furostanol glycosides of high molecular weight (above 650) have been investigated, and the most probable fragmentation patterns of the glycoside derivatives are presented. High resolution mass spectrometry provides useful information concerning the structures of both aglycone and sugar moieties as well as the molecular size of the oligoglycosides.

    Notes: Collisionally-activated dissociation (CAD) studies of linear peptides were performed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Four different gases, viz. helium, nitrogen, argon and xenon, were introduced into a gas cell, installed close to the ion source of TOF mass spectrometer, to obtain high-energy CAD information. The CAD spectral patterns for the linear peptides obtained using the four gases were almost the same as each other. The fragments characteristically obtained in conventional high-energy CAD using tandem mass spectrometry were observed also in these spectra. A difference between CAD spectra and post-source decay product-ion spectra has been demonstrated.
