Notes: Abstract Hypocotyls of dark-grown seedlings of Ara bidosis thaliana exhibit a strong negative gravitropism, which is reduced by red and also by long-wavelength, far-red light treatments. Light treatments using phytochrome A (phyA)- and phytochrome B (phyB)-deficient mutants showed that this response is controlled by phyB in a red/far-red reversible way, and by phyA in a non-reversible, very-low-fluence response. Crosses of the previously analyzed phyB-1 allele (in the ecotype Landsberg erecta background) to the ecotype Nossen wild-type (WT) background resulted in a WT-like negative gravitropism in darkness, indicating that the previously described gravitropic randomization observed with phyB-1 in the dark is likely due to a second mutation independent of that in the PHYB gene.

Notes: Abstract We show that a 61 bp fragment derived from the promoter region of the tobacco class I β-1,3-glucanase GLB gene enhances transcription in Nicotiana plumbaginifolia protoplasts independent of orientation relative to the start of transcription. This fragment leads to a cooperative stimulation of transcription when combined with the cauliflower mosaic virus 35S as-1 enhancer element. The GLB enhancer contains two copies of the sequence AGCCGCC, which is conserved in several genes showing expression patterns similar to the GLB gene, as well as a sequence identical at 6 of 7bp. Point mutations in these three sequences eliminate the enhancer activity of the 61 bp fragment. Nuclear extracts prepared from leaves of tobacco plants contain one or more putative transcription factors that interact specifically with the GLB enhancer. This factor was much less abundant in nuclear extracts prepared from upper leaves of untreated tobacco plants than in nuclear extracts prepared from upper leaves of ethylene-treated plants or from lower leaves. Since β-1,3-glucanase genes are expressed at very low levels in upper leaves of tobacco plants, at higher levels in lower leaves, and are induced in all leaves after treatment of plants with the stress hormone ethylene, we conclude that the enhancer element interacts with one or more transcription factors whose binding activity is correlated with gene expression in vivo.

Notes: Summary A light sensitive mutant was used as a recipient in the transfer of chloroplasts from a wildtype donor. Gamma irradiated (lethal dose) mesophyll protoplasts of Nicotiana gossei were fused with mesophyll protoplasts of a N. plumbaginifolia line carrying light sensitive plastids from a N. tabacum mutant. After fusion, colonies containing wild-type plastids from the cytoplasm donor were selected by their green colour. Most of the regenerated plants had N. plumbaginifolia morphology, but were a normal green in colour. The presence of donor-type plastids was confirmed by the restriction pattern of chloroplast DNA in each plant analysed. These cybrids were fully male sterile with an altered flower morphology typical of certain types of alloplasmic male sterility in Nicotiana. The use of the cytoplasmic light sensitive recipient proved to be suitable for effective interspecific transfer of wild-type chloroplasts. The recombinant-type mitochondrial DNA restriction patterns and the male sterility of the cybrids indicated the co-transfer of chloroplast and mitochondrial traits.

Notes: Summary Protoplasts of Nicotiana tabacum SRI (streptomycin resistant) and of Nicotiana knightiana (streptomycin sensitive) were fused using polyethylene glycol treatment. From three heterokaryons 500 clones were obtained. From the 43 which were further investigated, 6 resistant, 3 sensitive, and 34 chimeric (consisting of resistant and sensitive sectors) calli were found. From eight clones, a total of 39 plants were regenerated and identified as somatic hybrids. Chloroplast type (N. tabacum = NT or N. knightiana = NK) in the plants was determined on the basis of the species specific EcoRI restriction pattern of the chloroplast DNA. Regenerates contained NT (13 plants) or NK (15 plants) plastids but only the plants with the NT chloroplasts were resistant to streptomycin. This finding and our earlier data on uniparental inheritance points to the chloroplasts as the carriers of the streptomycin resistance factor.

Notes: Summary Mitochondrial DNA (mtDNA) restriction patterns were studied in mutant, cybrid and somatic hybrid plants regenerated from Nicotiana protoplasts. It has been shown that neither components of the culture media used for protoplast culture and plant regeneration, nor the antibiotics streptomycin and lincomycin used for the mutant selection induced alterations in the mtDNA. No rearrangements were detected in the mtDNA of plants derived from homoplasmic fusions where the mtDNA of the parents was identical as judged by mtDNA restriction patterns. There were rearrangements, however, in the mtDNA of each of the cybrid plants derived from heteroplasmic fusions. Restriction patterns generated by BamHI and SalI restriction endonucleases were different from those of both parents, and were composed of parental and non-parental fragments.

Notes: Abstract Transcription of ten nuclear genes was analysed in the albostrians mutant of barley (Hordeum vulgare L.). The lack of plastid ribosomes in white seedlings of this mutant results in a complex alteration of nuclear gene expression at the transcriptional level. We found a strong reduction in the accumulation of mRNAs transcribed from nuclear genes encoding chloroplast enzymes involved in the Calvin cycle, the chlorophyll a/b binding protein, and the cytosolic enzyme nitrate reductase. In contrast, the levels of transcripts of the genes encoding the cytosolic glycolytic enzymes glyceraldehyde phosphate dehydrogenase and phosphoglycerate kinase were slightly enhanced. Accumulation of chalcone synthase mRNA even reaches much higher levels in white than in green leaves. Ribosome-deficient plastids were combined by crossing with a nuclear genotype heterozygous for the albostrians allele. Analysis of transcript levels in F 1 plants having the same nuclear genotype and differing only with respect to their content of normally developed chloroplasts versus undifferentiated mutant plastids, provided strong genetic evidence for the plastid being the origin of a signal (chain) involved in regulation of nuclear gene expression. Results of run-on transcription in isolated nuclei demonstrated that the plastid signal acts at the level of transcription; it does not interfere with gene regulation in general. Mechanisms triggering nuclear gene expression in response to light operate in white mutant leaves: the very low levels of mRNAs derived from nuclear genes encoding chloroplast proteins and the strongly enhanced level of chalcone synthase mRNA were both light inducible. Also the negative regulation of leaf thionin gene expression by light is observed in white albostrians seedlings. Furthermore, in spite of its low absolute level, the circadian rhythm in the abundance of the cab mRNA is still detectable in plastid ribosome-deficient seedlings. Thus, functional plastid protein biosynthesis and photosynthesis are not preconditions for the circadian oscillations in the level of mRNA transcribed from this gene (family).

Description / Table of Contents: Abstract
Notes: Abstract Activity and selectivity data obtained in n-hexane dehydrocyclization over Pt/Al2O3 and Pd−Pt/Al2O3 catalysts exposed to different thermal treatment processes are compared. The ageing phenomena were most pronounced on catalysts treated in hydrogen at higher temperatures. The Pd−Pt/Al2O3 catalyst possessed lower cracking activity with a parallel increase of the selectivity for benzene and iso-hexanes.

Notes: Abstract Pt/Al2O3 catalysts with good reforming activity were prepared from [(CH3)3PtX]4 (X=Cl, Br, I) complexes and tested in pulse and continuous-flow reactors in n-hexane dehydrocyclization. It has been demonstrated that [(CH3)3PtX]4 complexes are excellent precursors of supported platinum catalysts.

Description / Table of Contents: Abstract
Notes: Abstract A γ-Al2O3 supported Re−Pt bimetallic catalyst prepared by electrochemical adsorption has been studied in n-hexane dehydrocyclization, using the slug-pulse method. The activity of the Re−Pt/Al2O3 catalyst treated only in hydrogen was low, but markedly increased by a calcination step before hydrogen treatment. The modified catalyst showed higher activity and increased selectivities for benzene and iso-hexanes than the base Pt/Al2O3 catalyst.
