    Description: p38 and c-Jun N-terninal kinase (JNK) are activated in response to acute stress and inflammatory signals. Through modification of a plethora of substrates, these kinases profoundly re-shape cellular physiology for the optimal response to a harmful environment and/or an inflammatory state. Here, we utilized phospho-proteomics to identify several hundred substrates for both kinases. Our results indicate that the scale of signaling from p38 and JNK are of a similar magnitude. Among the many new targets, we highlight the regulation of the transcriptional regulators grb10-interacting GYF protein 1 and 2 (GIGYF1/2) by p38-dependent MAP kinase-activated protein kinase 2 (MK2) phosphorylation and 14–3–3 binding. We also show that the Golgi apparatus contains numerous substrates, and is a major target for regulation by p38 and JNK. When activated, these kinases mediate structural rearrangement of the Golgi apparatus, which positively affects protein flux through the secretory system. Our work expands on our knowledge about p38 and JNK signaling with important biological ramifications.

    Description: Thick sulfate evaporite accumulations are absent from Proterozoic strata between ca. 2000 and ca. 1000 Ma, and detailed sedimentologic, stratigraphic, and geochemical data for the oldest Neoproterozoic thick marine sulfate evaporite successions are largely lacking. The middle Neoproterozoic Ten Stone Formation (Little Dal Group, Northwest Territories, Canada) consists of ~500 m of pelagic lagoonal gypsite and anhydritite (rocks consisting of the minerals gypsum and anhydrite) deposited shortly before the ca. 811 Ma Bitter Springs carbon isotope anomaly in an intracratonic basin that developed prior to breakup of Rodinia. The thickness of regional stratigraphic subdivisions of this formation, defined by subtle silt- and carbonate-bearing intervals, indicates a minor terrigenous source in the southeast and a silled connection to the open ocean in the northwest. Deposition of the Ten Stone Formation began with abrupt, tectonically triggered subsidence and restriction, and ended equally abruptly, as shown by stratigraphic contacts across which lithofacies corresponding to strikingly different paleoenvironments change sharply, with no evidence for hiatus or erosion. Stratigraphic cyclicity in the evaporite succession is minimal owing to isolation of bottom-hugging, dense lagoonal brine from overlying waters. Deposition of the Ten Stone Formation in a basin that experienced intermittent, basin-scale tectonic adjustments, as recorded by details of its stratigraphy, supports the interpretation that the Mackenzie Mountains Supergroup accumulated in an extensional, tectonically active intracratonic basin whose structure resembled a lower-plate extensional system. The absence of halite from the Ten Stone Formation contrasts with its abundance in the stratigraphically lower, gypsum-free Dodo Creek Formation, suggesting that deposition of the lower to middle Little Dal Group spanned a major oxygenation event, during which the sequence of evaporite mineral precipitation from seawater changed from halite-first to sulfate-first in response to rapid accumulation of atmospheric oxygen and concomitant increase in the global marine sulfate reservoir. The limited range of sulfur isotope values in a new data set spanning hundreds of meters of gypsite indicates a strongly and persistently oxidizing mid-Neoproterozoic atmosphere, an abundance of sulfate in seawater, and marine oxygenation extending below storm wave base. The mineralogy, sedimentology, stratigraphy, and geochemistry of the Ten Stone Formation are virtually indistinguishable from those of thick, Phanerozoic "deep-water" (below wave-base) evaporite successions, and indicate that the tectonic, climatic, and geochemical conditions required for deposition of thick successions of marine sulfate evaporites were well established prior to ca. 811 Ma. Thick sulfate evaporite successions in equivalent stratigraphic positions just below the Bitter Springs carbon isotope excursion elsewhere in Laurentia, as well as on the Congo craton, and in South Australia attest to the global impact of the rapidly increased seawater sulfate reservoir prior to Rodinia’s breakup. High relative burial rates of organic matter prevailed before the breakup of Rodinia and led to oxygenation of the atmosphere-ocean system, growth of the seawater sulfate reservoir, and, in association with a warm and arid climate, deposition for the first time in Earth’s history of thick sulfate evaporites in the middle Neoproterozoic, ~100 m.y. before the first Cryogenian glacial episode. The Neoproterozoic Oxygenation Event may have taken place in several steps, the first of which preceded the Bitter Springs anomaly.

    Description: Life on Earth is thought to have coevolved with the chemistry of the oceans and atmosphere, and the shift from an anoxic to an oxic world across the Archean-Proterozoic boundary represents a fundamental step in this process. In order to understand the relative influence of biological and geological factors on this transition, we must constrain key variables in seawater chemistry before the Great Oxidation Event (ca. 2500 Ma). We present a multielement (C-S-Fe-Mo) biogeochemical study of ca. 2662 Ma shales from the Hamersley Province in Western Australia. Our data reveal a sustained episode of Fe-limited pyrite formation under an anoxic and sulfidic (euxinic) water column. This is the oldest known occurrence of euxinia in Earth's history and challenges the paradigm of persistently Fe-rich Archean oceans. Bulk trace metal chemistry and preservation of strong mass-independent S isotope fractionations in sedimentary pyrites indicate that ocean euxinia was possible prior to oxidative weathering, suggesting that sulfidic waters may have been common throughout the Archean Eon. C-S-Fe systematics suggest that oxygenic photosynthesis was the primary source of organic carbon in the basin, and the absence of Mo enrichments highlights a potential link between inefficient nitrogen fixation and the delayed arrival of the Great Oxidation Event.

    Description: Constraining oxygen levels in the early Precambrian surface ocean has been a longstanding goal, but efforts have been challenged by the availability of suitable proxies. Here we present a novel approach, iodine geochemistry, which broadens our perspective by providing constraints on shallow, carbonate-dominated marine settings. Iodate ( ) persists exclusively in oxic waters and is the sole iodine species incorporated into carbonate minerals, allowing iodine-to-calcium ratios (I/Ca) in shallow carbonates to be used as a paleoredox indicator. Our data from a series of Mesoarchean through Paleoproterozoic carbonates deposited under shallow-marine conditions reveal a progressive surface ocean oxygenation in the early Paleoproterozoic. These data seem to indicate that a largely anoxic surface ocean extended throughout the Archean until the Great Oxidation Event (GOE) at ca. 2.4 Ga, implying that previous inferences of pre-GOE oxygen production may reflect oxygen oases, transient oxidation events, or oxygen levels below those required for accumulation. The data suggest formation and persistence of and, consequently, surface ocean oxygen concentrations of at least 1 µM during the GOE. Following the initial rise of oxygen, carbonate-associated iodine in globally extensive carbonate units deposited during the Lomagundi positive carbon isotope excursion at ca. 2.22–2.1 Ga suggests a widespread aerobic iodine cycle beyond that operating prior to the event, synchronous with high relative rates of organic carbon burial and apparent expansion of oxidative conditions.

    Description: South Africa has great potential for considering wind energy as an alternative resource. The climatology allows for significant wind energy production. An accurate joint description of the wind speed (linear) and wind direction (circular) characteristics is important for wind farm development. In this paper, a bivariate class of flexible joint probability density functions of wind speed and wind direction for the use in wind energy analysis is presented. This joint model accounts for bimodality, skewness, and a dependency structure between the wind speed and wind direction. For the joint probabilistic description of the wind speed and wind direction, special cases of this bivariate class are evaluated, namely the semi-parametric Möbius model on the disc, the Möbius distribution on the disc, and the Beta type III Möbius distribution on the disc. These three special cases are applied to wind speed and wind direction data recorded every ten minutes at two locations in South Africa. Evaluation of the models is based on three different information criteria and normalized deviation. Overall, the semi-parametric model is superior to the parametric models based on the performance measures. The wind energy potential at the two locations is evaluated using the semi-parametric model.

    Description: The impact of oxygen exposure during winemaking on metal ion concentrations in wine were investigated throughout the winemaking process in a Chardonnay wine. The concentrations of Al, Ca, Co, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, Sn, and Zn were determined using inductively coupled plasma-mass spectrometry. Oxygen exposure significantly impacted 13 metal ions at different phases of winemaking. However, only the concentrations of Cr, Cu, and Fe were impacted by early oxygen exposure during pressing, with lower Cr and Cu concentrations in wines that were aerobically pressed and lower concentrations of Fe in wines that were inertly pressed. The sequestering of Al, Cu, Ni, and Zn by wine lees was significantly affected by oxygen treatment, with lees collected from wines that were treated oxidatively sequestering significantly greater amounts of Cu and Zn and removing these metals from the wine supernatant. The metal ion that was most affected by oxygen exposure during pressing and handling was Cu, with significantly lower Cu measured in wines that were produced under oxidative conditions. It is known that elevated Cu concentrations have negative implications for wine aroma and flavour. This study demonstrated that oxygen management during winemaking significantly impacts metal ion concentrations in lees and wine, which may decrease the risk of developing taints and faults.

    Description: Bacterial vaginosis (BV) and periodontal disease (PD) are characterised as bacterial dysbioses. Both are associated with an increased risk of poor pregnancy outcomes, yet it is unknown whether PD and BV are related. We characterised the oral microbiota of young South African females with a high prevalence of BV and investigated the association between oral communities and vaginal microbiota. DNA was extracted from vaginal lateral wall, saliva and supragingival plaque samples from 94 adolescent females (aged 15–19 years). 16S rRNA gene sequencing of the V4 hypervariable region was performed for analysis of the oral and vaginal microbiota and BV status was determined by Nugent scoring. The core oral microbiota was predominately comprised of Firmicutes followed by Proteobacteria and Bacteroidetes. The salivary microbiota of participants with BV was more diverse than those with lactobacillus-dominated communities (p = 0.030). PD-associated bacterial species, including Prevotella intermedia and Porphyromonas endodontalis were enriched in the supragingival microbiota of women with non-optimal vaginal communities compared to those with Lactobacillus-dominant communities, while Pseudomonas aeruginosa and Prevotella intermedia were enriched in the saliva of women with non-optimal vaginal microbiota. These data suggest a relationship between oral and vaginal dysbiosis, warranting further investigation into whether they are casually related.

    Description: The assumption of symmetry is often incorrect in real-life statistical modeling due to asymmetric behavior in the data. This implies a departure from the well-known assumption of normality defined for innovations in time series processes. In this paper, the autoregressive (AR) process of order p (i.e., the AR(p) process) is of particular interest using the skew generalized normal (SGN) distribution for the innovations, referred to hereafter as the ARSGN(p) process, to accommodate asymmetric behavior. This behavior presents itself by investigating some properties of the SGN distribution, which is a fundamental element for AR modeling of real data that exhibits non-normal behavior. Simulation studies illustrate the asymmetry and statistical properties of the conditional maximum likelihood (ML) parameters for the ARSGN(p) model. It is concluded that the ARSGN(p) model accounts well for time series processes exhibiting asymmetry, kurtosis, and heavy tails. Real time series datasets are analyzed, and the results of the ARSGN(p) model are compared to previously proposed models. The findings here state the effectiveness and viability of relaxing the normal assumption and the value added for considering the candidacy of the SGN for AR time series processes.

    Description: Maneuvers in level ice are common operations for icebreakers and polar supply vessels. Maneuvering exposes the midship and stern area to ice interaction, influencing the magnitude and frequency of ice-induced loading in these areas. However, full-scale measurements do not typically cover the midship and stern areas, as measurements have commonly focused on the bow area. Controlled maneuvering tests were conducted during the ice trials of S.A. Agulhas II in the Baltic Sea. During these tests, ice-induced loading at different hull areas was measured simultaneously with ship control, navigation, and ice condition data. This work studied the effect of maneuvers on the characteristics and statistics of ice-induced loading at different hull areas and compared the impact to ahead operations. The study showed that the maneuvers had minor impact to the magnitude, frequency, and duration of loading at the bow and bow shoulder. On the other hand, maneuvers had a clear effect on the load magnitude and frequency at the stern shoulder. Additionally, a statistical analysis showed that the load magnitude increased as a function of load duration in all hull areas. Furthermore, the analyzed measurement data are presented and made available with the paper.

    Description: Chronic cigarette smokers (CCS) are known to have elevated levels of carboxyhemoglobin (COHb). However, it is not known whether increased levels of COHb are associated with endothelial dysfunction (ED), and therefore the development of peripheral arterial disease (PAD). The aim of the study was to investigate the association of blood COHb and plasma nitric oxide (NO) levels, and whether it is an independent risk factor in the development of PAD among CCS at Dr George Mukhari Academic Hospital (DGMAH). A sample of 120 CCS with PAD and a convenience sample of 100 CCS without PAD were recruited into the study. Blood COHb levels were measured using the ABL 90 FLEX CO-oximeter automated spectroscopy. Plasma nitric oxide (NO) levels were measure using ELISA. Logistic regression analysis was used to investigate the association of blood COHb and plasma NO with PAD. Blood COHb levels of CCS with PAD were significantly higher than those of CCS without PAD, and the NO levels of CCS with PAD were significantly lower than those of CCS without PAD. Although both the blood COHb and plasma NO in CCS were significantly associated with PAD in bivariate logistic analysis, only plasma NO was independently associated with PAD in multivariate logistic analysis. This finding is consistent with the hypothesis that COHb is a cause of arterial damage in PAD, leading to reduced NO, and therefore reduced arterial dilation.
