    Notes: Abstract The energy-dependent transport of phenylalanine into isolated vacuoles of barley (Hordeum vulgare L.) mesophyll protoplasts has been studied by silicone-layer floatation filtering. The uptake of this aromatic amino acid into the vacuolar compartment is markedly increased by MgATP, showing saturation kinetics; the K m values were 0.5 mM for MgATP and 1.2 mM for phenylalanine. V max for phenylalanine transport was estimated to 140 nmol phenylalanine·(mg·Chl)-1·h-1. The transport shows a distinct pH optimum at 7.3 and is markedly inhibited by 40 mM nitrate. Azide (1 mM) and vanadate (400 μM) had no or little effect on rates of transport while p-fluorophenylalanine seemed to be an effective inhibitor, indicating a possible competition at an amino-acid carrier. Ionophores such as valinomycin, nigericin or gramicidin were strong inhibitors of phenylalanine transport, indicating that this process is coupled to both the transmembrane pH gradient (ΔpH) and the transmembrane potential (ΔΨ).

    Notes: Abstract During solar flares and coronal mass ejections, nuclei and electrons accelerated to high energies are injected into interplanetary space. These accelerated particles can be detected at the SOHO satellite by the ERNE instrument. From the data produced by the instrument, it is possible to identify the particles and to calculate their energy and direction of propagation. Depending on variable coronal/interplanetary conditions, different kinds of effects on the energetic particle transport can be predicted. The problems of interest include, for example, the effects of particle properties (mass, charge, energy, and propagation direction) on the particle transport, the particle energy changes in the transport process, and the effects the energetic particles have on the solar-wind plasma. The evolution of the distribution function of the energetic particles can be measured with ERNE to a better accuracy than ever before. This gives us the opportunity to contribute significantly to the modeling of interplanetary transport and acceleration. Once the acceleration/transport bias has been removed, the acceleration-site abundance of elements and their isotopes can be studied in detail and compared with spectroscopic observations.

    Notes: Abstract Earlier studies on the synthesis of C3-derived amino acids, plastidic isoprenoids and fatty acids from CO2 by isolated chloroplasts in the light indicate the presence of a complete, but low-capacity, chloroplast (chlp) 3-phosphoglycerate → acetyl-CoA pathway which is predominantely active in immature (developing) chloroplasts (A. Heintze et al., 1990, Plant Physiol. 93, 1121–1127). In this paper, we demonstrate the activity of the enzymes involved i.e. chlp phosphoglycerate mutase, chlp enolase, chlp pyruvate kinase and chlp pyruvate-dehydrogenase complex (PDC), in the stroma of purified barley (Hordeum sativum L.) chloroplasts of different developmental stages. The chlp phosphoglycerate mutase was partially purified for the first time. The activities of the enzymes of this chlp pathway (except PDC) were about a magnitude lower than those of the cytosolic enzymes. The chlp PDC of barley was more active than that of spinach. The apparent K m values of the enzymes of this pathway were about 100 μM or lower except for the chlp phosphoglycerate mutase which had a K m of 1.6–1.8 mM for 3-phospho-d-glycerate. Interestingly, no appreciable change in the activity of these enzymes was observed during maturation of the chloroplasts. In contrast, the activity of the reversible NADP+-glyceraldehyde 3-phosphate dehydrogenase increased about five times (from 140 to 590 nkat per g leaf dry weight). The following hypothesis is put forward to explain the regulation of carbon metabolism during chloroplast development: 3-phospho-d-glycerate is withdrawn from a common pool by the actions of 3-phosphoglycerate kinase and NADP+-glyceraldehyde-3-phosphate dehydrogenase, the activity of which increases considerably during maturation of chloroplasts. This leads to an insufficient supply of 3-phospho-glycerate for the chlp phosphoglycerate mutase, which has a low affinity for its substrate.

    Notes: Abstract To verify the enhancement in sensitivity of a Schottky barrier diode using a 90° corner reflector at submillimeter wavelength a 65∶1 scaled model of the open structure mixer was constructed and investigated at 2.8 cm wavelength. The results show clearly the advantage of a 90° corner reflector, the reality of the deviations for 45° and 60° corner reflectors found for submillimeter wavelengths, and the equality of the antenna patterns for a 90° corner reflector and a “cat-eye” (corner cube) reflector.

    Notes: Abstract The performance of a submillimeter heterodyne receiver using an HCOOH laser local oscillator and an open structure mixer with a Schottky barrier diode has been optimized for 693 GHz. Working at room temperature a single sideband (SSB) system noise temperature of 7,300 K, a mixer noise temperature of 6,100 K and a conversion loss of 12 dB has been achieved. The same receiver system has been investigated at 324 GHz using an HCOOD laser local oscillator yielding a noise temperature of 3,100 K (SSB), a mixer noise temperature of 2,400 K (SSB) and a conversion loss of 10 dB (SSB). An acousto-optical spectrometer has also been constructed, with 1024 channels and a channel-bandwidth of 250 kHz. The system NEP per channel was 2.5×10−17 W/Hz1/2 at 324 GHz and 5.0×10−17 W/Hz1/2 at 693 GHz.

    Notes: Abstract A compact heterodyne receiver system used in the Kuiper Airborne Observatory (KAO) is described. Calibration techniques and methods for eliminating standing wave effects are presented. The J=7–6 rotational transition of carbon monoxide was detected in OMC-1 and in W3 with a beam of 5.5′ HPBW. The peak antenna temperature of OMC-1 is 55 K, with the line center at +8±1 kms−1 and FWHM 6.0±0.3 kms−1. The total line flux in our beam is 7.8×10−13 Wm−2. This relatively high value seems to indicate that OMC-1 might be extended over at least several arcminutes in CO (J=7–6). In W3, the peak antenna temperature is 6±2 K, with line center at −42±2 kms−1 and FWHM ∼9 kms−1. The total line flux is 1.5×10−13 Wm−2. W3 thus emits about 50 L⊙ in CO (J=7–6) alone.
