    Biology and fertility of soils 23 (1996), S. 15-19
    ISSN: 1432-0789
Schlagwort(e): Key wordsBradyrhizobium japonicum ; Bradyrhizobium elkanii ; Repeated sequence RSα ; Polymerase chain reaction detection ; DNA hybridization ; Soil inoculant ; Nodulation tests ; N2 fixation
Quelle: Springer Online Journal Archives 1860-2000
Thema: Biologie , Geologie und Paläontologie , Land- und Forstwirtschaft, Gartenbau, Fischereiwirtschaft, Hauswirtschaft
Notizen: Abstract The genome of Bradyrhizobium japonicum and B. elkanii contains multiple copies of the repeated DNA sequence RSα. A collection of 18 B. japonicum, 4 B. elkanii and 72 other bacterial strains was screened by polymerase chain reaction (PCR) using a pair of primers specific for RSα. Only strains of B. japonicum and B. elkanii gave the predicted amplification product. Restriction analysis of PCR products obtained from different strains of B. japonicum showed that the RSα sequence was generally conserved. The usefulness of RSα as a specific probe for Bradyrhizobium strains capable of nodulating soybean was also demonstrated.

Schlagwort(e): Column liquid chromatography ; Urine clean-up ; Arsenic speciation ; Microwave assisted oxidation ; Atomic absorption spectrometry
Quelle: Springer Online Journal Archives 1860-2000
    Thema: Chemie und Pharmazie
Notizen: Summary An effective clean-up method based on the precipitation of most high molecular mass inorganic salts and organic compounds in ethanol at −15°C has been developed for the determination of arsenite, arsenate, monomethylarsonate (MMA), dimethylarsinate (DMA), arsenobetaine (AsB) and arsenocholine (AsC) in urine. Analyte recovery was close to 100%. The six species were separated on an anionic HPLC column and determined by microwave-assisted oxidation-hydride generation-atomic absorption spectrometry. The detection limits and relative standard deviation of the whole procedure were within the 8–15 μg L−1 and 2.5–5.3% ranges, respectively, which allows application to the analysis of urine of people exposed to arsenic.
