    Notes: Summary The gene for the antibacterial peptide colicin B was cloned and transformed into a host background where it was constitutively overexpressed. The purified gene product was biologically active and formed voltage-dependent, ion-conducting channels in planar phospholipid bilayers composed of asolectin. Colicin B channels exhibited two distinct unitary conductance levels, and a slight preference for Na+ over Cl−. Kinetic analysis of the voltage-driven opening and closing of colicin channels revealed the existence of at least two conducting states and two nonconducting states of the protein. Both the ion selectivity and the kinetics of colicin B channels were highly dependent on pH. Excess colicin protein was readily removed from the system by perfusing the bilayer, but open channels could be washed out only after they were allowed to close. A monospecific polyclonal antiserum generated against electrophoretically purified colicin B eliminated both the biological and in vitro activity of the protein. Membrane-associated channels, whether open or closed, remained functionally unaffected by the presence of the antiserum. Taken together, our results suggest that the voltage-independent binding of colicin B to the membrane is the rate-limiting step for the formation of ion channels, and that this process is accompanied by a major conformational rearrangement of the protein.

    Notes: Abstract People living near or in tropical forest ecosystems have traditionally extracted forest products, i.e. timber, food and medicinal plants for their livelihood. Such practice does not create too much disturbance when the population is still sparse, and the product is used only for their own needs. When population pressure becomes greater, and when the motive of extraction is profit, then the disturbance become serious and created environmental problems. Major exploitation of the Indonesian rain forest for timber began in the 1960s and is continuing today. The lowland rain forests of Sumatra and Kalimantan have been particularly logged. Exploitation has often been destructive because Forest Department rules have been widely ignored. Moreover, once roads have given access to formerly inaccessible areas, farmers have often moved in after the timber companies and then cleared the relict, regenerating forest for either permanent or shifting cultivation. The traditional shifting cultivations have been practised for years, producing millions of ha of impoverished secondary types of forest, degraded lands and alang-alang (Imperata cylindrica) grasslands. Forests have also been lost through conversion of land to plantation agriculture and transmigration programmes, mining, construction roads and railways and also natural disturbances, such as drought and fire. This paper will discuss the human impact upon tropical forest dynamics in general, with examples from Indonesian and other Southeast Asia countries' tropical forests.

    Notes: A method is described for the determination of polycyclic aromatic hydrocarbons (PAH) in the diluted exhaust gas of diesel vehicles. Sampling is done by drawing off proportional streams from the dilution tunnel. The particulates deposited on filters are sublimed, and the sublimate is purified and prefractionated on silica gel. Further separation and quantitative determination of the PAH is performed by two dimensional TLC in conjunction with In situ fluorescence spectrometry. Results of theoretical considerations and experimental data on the distribution of the emitted PAH between particulate matter and the corresponding gas phase in diluted exhaust are presented. A modified Langmuir adsorption model is used to explain the effects of dilution ratio and sample temperature in the dilution tunnel. Comparison of the emission values for PAH obtained from diluted and undiluted exhaust shows good agreement. Furthermore, the PAH contents of the exhaust gas of several diesel-engined cars are also compared for various driving cycles.
