    Notes: Abstract Cysteine is found to accelerate electroless nickel (EN) deposition and hydrogen evolution simultaneously. Changes in the activation energy of the EN reaction and the electrochemical behaviour of the EN bath due to the presence of cysteine were studied. The composition of the deposit formed was determined by XPS. The results show that the activation energy of the EN reaction is decreased when cysteine is added to the bath, and also that cysteine only influences the anodic process and has no direct effect on the cathodic process. The high resolution XPS spectra show that cysteine is adsorbed in the surface layer, and S2− in the inner layer of the deposit. It is deduced that cysteine may participate in the formation of a reactive intermediate and facilitate the oxidation of H2PO 2 − , which results in the acceleration of EN plating.

    Notes: Abstract Native β-cyclodextrin, deprotonated at its secondary hydroxyl groups has been studied as pseudostationary phase for electrokinetic chromatographic separation of highly hydrophobic compounds. Using this pseudostationary phase the separation of five polycyclic aromatic hydrocarbons (PAHs) is shown. The influence of the main parameters (field strength, pH, concentration of cyclodextrin, urea and organic solvent) on the separation of PAHs and migration behaviour of β-CD is discussed.

    Notes: Abstract Twenty-eight unrelated persons heterozygous for familial hypercholesterolemia (FH) were screened to assess the frequency and nature of major structural rearrangements at the low-density lipoprotein (LDL) receptor gene in Korean FH patients. Genomic DNA was analyzed by Southern blot hybridization with probes encompassing exons 1–18 of the LDL receptor gene. Two different deletion mutations (FH29 and FH110) were detected in three FH patients (10.7%). Each of the mutations was characterized by the use of exon-specific probes and detailed restriction mapping mediated by long-PCR (polymerase chain reaction). Mutation FH29 was a 3.83-kb deletion extending from intron 6 to intron 8 and FH110 was a 5.71-kb deletion extending from intron 8 to intron 12. In FH29, the translational reading frame was preserved and the deducible result was a cysteine-rich A and B repeat truncated protein that might be unable to bind LDL but would continue to bind β-VLDL. FH110 is presumed to be a null allele, since the deletion shifts the reading frame and results in a truncated protein that terminates in exon 13. Sequence analysis revealed that both deletions have occurred between two Alu-repetitive sequences that are in the same orientation. This suggested that in these patients the deletions were caused by an unequal crossing over event following mispairing of two Alu sequences on different chromatids during meiosis. Moreover, in both deletions, the recombinations were related to an Alu sequence in intron 8 and the deletion breakpoints are found within a specific sequence, 27 bp in length. This supports the hypothesis that this region might have some intrinsic instability, and act as one of the important factors in large recombinational rearrangements.

    Notes: Abstract  A protocol is presented for genetically engineering Cymbidium orchid using particle bombardment. This protocol enabled the routine transformation of orchid plants that were previously difficult to transform. Liquid culture was used to generate a large number of protocorm-like bodies (PLBs) to be bombarded and to promote continued development of the bombarded meristematic tissue. Plasmid DNA (pKH200) carrying the GUS-INT and NPTII genes flanked by tobacco matrix attachment regions was introduced into the meristematic cells of PLBs by particle acceleration. The transformed PLBs were proliferated and selected for kanamycin resistance conferred by the introduced NPTII gene. Shoot regeneration was then induced from the kanamycin-resistant PLBs, and transgenic plantlets were produced. Both the kanamycin-resistant PLBs and regenerated shoots expressed the GUS-INT gene. The presence of the introduced gene in the transformed orchid plants was confirmed by PCR analysis, sequencing and Southern blot analysis of the PCR product. The recovered transgenic plants were established in soil and acclimatized in the greenhouse.

    Notes: Abstract  We describe a protocol for Agrobacterium tumefaciens-mediated transformation of hybrid cottonwoods (Populus sections Tacamahaca Spach. and Aigeiros Duby). The protocol has allowed routine transformation of several economically important cottonwood hybrids (Populus trichocarpa Torr. & Gray×P. deltoides Bartr. ex. Marsh. and P. deltoides×P. nigra L.) that were previously difficult to transform. The procedure was applied to 11 different hybrid cottonwood genotypes and one P. deltoides genotype using kanamycin as the selection agent. Additional experiments showed a very strong interaction between auxin preculture and the effectiveness of various cytokinins for induction of shoot organogenesis. The data also demonstrated the superiority of Agrobacterium strain EHA105 over C58 and LBA4404 for T-DNA transfer based on transient assays with a reporter gene.

Wear resistance of composites is improved due to the presence of reinforcements, but no noticeable improvements are observed in the wear resistance with more than 20% addition of reinforcements. To analyse wear mechanisms, wear surfaces are examined by scanning electron microscopy (SEM). The major wear mechanisms of discontinuous metal matrix composites (MMC)s are strongly dependent on sliding speeds. Dominant mechanism is the adhesive-abrasive wear at low and intermediate sliding speeds, and melt wear at high sliding speeds. Weight loss is linearly increased with the sliding distance. The effect of reinforcements' orientations on wear behaviours is also discussed.

    Notes: Abstract New dyes with characteristic fluorescence lifetimes have been developed for bioanalytical applications. Based upon the concept of “multiplex dyes,” we have designed rhodamine dyes with nearly identical absorption and emission spectral characteristics but different fluorescence lifetimes. Extending this principle to applications with laser diodes, new rhodamines with functional groups for covalent coupling of analytes have been developed. The new labels exhibit absortion and fluorescence beyond 600 nm and have a high quantum efficiency, even in aqueous buffer systems.
