Notes: Abstract We describe preliminary experimental results concerning the operation of a 3 m arm-length Michelson interferometer with two Fabry-Perot cavities whose mirrors and beam splitter are suspended independently by wires. The reflected light beams from the two Fabry-Perot cavities are recombined to obtain interference at a photo-detector; this scheme is necessary for future power-recycled laser interferometers used to detect gravitational waves. The fundamental properties of the interferometer are presented, including the power spectral density of the displacement noise.

Notes: Summary The phonon energy spectra of a polycrystalline α-Fe foil were observed at 150 K and 300 K by using the nuclear resonant scattering of synchrotron radiation. In each spectrum, inelastic scattering was observed at both sides of the elastic peak. It was found that the ratio of the elastic-scattering component and the asymmetry of the intensity of the side bands observed at 150 K are larger than those observed at 300 K, respectively. The observed temperature-dependent spectra are in good agreement with the spectra calculated from the phonon energy distribution function. One of the advantageous features of this method is that the excitation of only a specific element is possible. Our results show that this method is applicable to the study of lattice dynamics and opens a new field of the nuclear resonant scattering spectroscopy.

Notes: Abstract The effect of tetrachloroethylene on Phase I and II drug-metabolizing enzymes in rat liver was examined. Rats were treated orally with tetrachloroethylene daily for five days, at doses of 125, 250, 500, 1,000 and 2,000 mg/kg. The higher doses (〉500 mg/kg) of tetrachloroethylene induced the hepatic microsomal 7-pentoxyresorufin O-depentylase and 7-benzyloxyresorufin O-debenzylase activities associated with the CYP2B subfamily. 7-ethoxyresorufin O-deethylase activity was also induced about 2-fold compared with that of control rats at 500, 1,000, and 2,000 mg/kg dose levels of tetrachloroethylene. However, 7-ethoxycoumarin O-deethylase and 7-methoxyresorufin O-demethylase activities were increased significantly at only the 1,000 mg/kg dose level of tetrachloroethylene (1.4- and 1.5-fold). Although other cytochrome P450-mediated monooxygenase activities such as nitrosodimethylamine N-demethylase, aminopyrine N-demethylase and erythromycin N-demethylase were also induced by tetrachloroethylene, the relative induction to control activity was lower than those of 7-pentoxyresorufin O-depentylase and 7-benzyloxyresorufin O-debenzylase. Western immunoblotting showed that the levels of CYP2B1 and CYP2B2 proteins in liver microsomes were increased at doses of 1,000 and 2,000 mg/kg of tetrachloroethylene. In addition to cytochrome P450-mediated monooxygenases, there was significant induction of the Phase II drug-metabolizing enzymes, DT-diaphorase, glutathione S-transferase activities towards 1-chloro-2,4-dinitrobenzene and 1,2-dichloro-4-nitrobenzene, and UDP-glucuronyltransferase activities towards 4-nitrophenol and 7-hydroxycoumarin. The results indicate that tetrachloroethylene induces both Phase I (CYP2B-mediated monooxygenase) and Phase II drug-metabolizing enzymes (DT-diaphorase, glutathione S-transferase and UDP-glucuronyltransferase) in the rat liver.

Notes: Abstract The X-ray absorption in a 5 μm thick Ni foil for energies from 8.2 keV to 8.8 keV was measured by the photoacoustic method using a gas-microphone-cell. Results are presented for both photoacoustic amplitude and phase as a function of incident photon energy. The absorption fine structure above the NiK-edge is clearly resolved. The data are compared with a simple theory for the signal generation, and discrepancies are explained by discussing the various energy dissipation channels following the X-ray absorption.

Notes: Summary A family in which two first cousins were found to have the Prader-Willi syndrome was investigated cytogenetically. Although G-banding analysis of metaphase chromosomes failed to demonstrate abnormality, close analyses on the fine prometaphase bands by G-banding and the DA-DAPI bands by double stainings revealed a distinct chromosome abnormality in this family. A reciprocal translocation, rep(14:15)(q11.2;q13), was detected in three family members: the mother, the maternal grandmother, and a maternal uncle of the proband. And, the proband and one of the first cousins had an unbalanced translocation that was derived from their carrier parents. The karyotypes of the affected cousins were determined as 46,XY or XX,-15,+der(14),rcp(14;15)(q11.2;q13). Therefore, they were considered to have an identical cytogenetic abnormality: a partial trisomy of the 14pter→q11.2 segment and a partial monosomy of the 15pter→q13 segment. Detailed clinical features of the proband and his affected cousin are described, main features associated with the Prader-Willi syndrome having been observed in both cousins. These observations support a definite relationship between the Prader-Willi syndrome and chromosome 15.

Notes: Abstract We have developed a high energy, high resolution monochromatic x-ray computed tomography system using synchrotron radiation. Our system is installed in a vertical wiggler beamline at the Photon Factory of the National Laboratory for High Energy Physics that can produce high brilliance x-rays in the high energy region. At the present stage, we have obtained monochromatic x-rays up to 75 keV, succeeded in taking computed tomography images of materials up to 50 keV with large absorption coefficients that could not be imaged by conventional methods, and observed three-dimensional fine structures in such materials. When the K-edge subtraction method was applied, the system could quantitatively and nondestructively measure the trace distribution of a specific element in the object of interest. In this paper we introduce the experimental system, mainly the crystal monochromator, evaluate the results, and discuss the performance of the system.

Notes: Abstract Objectives: The aim of this study was to develop limited sampling models for estimating the area under the concentration-versus-time curve (AUC) of carboplatin. Methods: Based on pharmacokinetic analyses of 14 patients who received 300 mg · m2 of carboplatin over a 90-min infusion following irinotecan, we developed limited sampling models with stepwise multiple linear regression analysis. We validated these models to be unbiased and precise using pharmacokinetic data of a second group of 14 patients. We also compared the observed and the predicted AUC in the patients using Calvert's formula with the patients' renal function. Results: We developed the following models: AUC (mg · ml−1 · min) = 0.784 × C4 + 1.30 (r 2 = 0.930) and AUC = 0.100 × C0.25 + 0.597 × C4 + 0.140 (r 2 = 0.992), where C0.25 and C4 denote unbound plasma concentrations (μg · ml−1) of carboplatin at 0.25 h and 4 h after the end of infusion, respectively. These models were validated to be unbiased and precise: a mean prediction error (MPE) with standard deviation (SD) = 2.41 (9.45)% and a root mean squared error (RMSE) = 9.42% for the one-sample model, and MPE with (SD) = 1.22 (5.56)% and RMSE = 5.49% for the two-sample model. We also calculated predicted AUC in the patients using Calvert's formula: MPE with (SD) =−5.87 (21.5)% and RMSE = 21.5%. Conclusions: These estimations were, as expected, more accurate than the prediction using Calvert's formula based on patients' renal function. The result of this study confirmed the idea that the pharmacokinetic parameters derived from limited sampling models would be more suitable for pharmacokinetic analysis of carboplatin than those obtained using Calvert's formula.
