Notes: Abstract. Euphorbia dulcis endosperm is the site of controlled long lasting endocellular lysis involving segregation and autophagy of portions of the cytoplasm within the endoplasmic reticulum membrane. The lysis products may constitute a food source for the benefit of the early developing embryo.

Notes: Summary Ultrastructural changes in the integumentary cell walls of Nerium oleander L. were observed, starting with the beginning of nucellus degeneration. The cell walls in direct contact with the nucellus, followed in a regular progression by those of the next 2–3 cell layers, were seen to increase rapidly in thickness and, in contact with the plasmalemma, to develop a peculiar layer characterized by the presence of numerous membrane-like structures. Morphological and cytochemical findings indicate a membraneous nature of these wall structures; the structures exhibit a marked affinity to potassium permanganate, ruthenium red and phosphotungstic acid, and possess a three-layered configuration. Moreover, the structures were found to be disorganized by phospholipase C. Some of the wall structures appear to be pitted, sac-shaped formations; others to be single sheets. Both types exhibit a direct continuity with the plasmalemma after digestion of the wall material by cellulase. The origin and development of these structures are discussed.

Notes: Summary The development of characteristic cytoplasmic inclusions from endoplasmic-reticulum (ER) cisternae in nucellar cells of some species of Euphorbia has been studied by electron microscopy. The formation of these organelles is preceded by the appearance of rough ER cisternae filled with an electron-dense material and forming complicated networks. Vesicular structures are formed which grow rapidly to give electron-dense, spherical dilations. On the outer surface of their limiting membrane numerous ribosomes and often polysomes are present. This membrane can be seen to remain continuous with the membranes of one or more cisternae of the rough ER up to when the dilations have a maximum diameter of 2.5–3 μ. At this time, continuity between the ER cisternae and the spherical dilations ceases. After this the new cell organelles remain unchanged in size, shape, and electron-density until the cell is disintegrated by the growing embryo-sac. The fate of the contents of these organelles is discussed.

Notes: Abstract Estrogen replacement therapy is effective in the prevention of postmenopausal osteoporosis, and a direct action of 17-β-estradiol (17βE2) on osteoblastic and osteoclastic cells has been demonstrated. The inhibition of bone resorption by ipriflavone (IP), an isoflavone derivative devoid of estrogenic properties but active in potentiating the effects of estroge on bone tissue, has been shown in in vitro and in vivo studies and confirmed by clinical data. To investigate the molecular mechanisms that underlie IP effect, we studied the possible interactions of IP and its four main in vivo metabolites (I, II, III, and V) with the estrogen receptor (ER) in the human preosteoclastic cell line FLG 29.1, whose growth and function are modulated by the compound. In parallel experiments, the human breast cancer cell line MCF7 was also analyzed. IP binding sites were demonstrated in the nuclear fraction of FLG 29.1 cells. 17βE2 and other steroid compounds failed to displace IP binding to intact FLG 29.1 cells. Similarly, IP and metabolites I, III, and V were not able to displace 17βE2 binding to intact MCF7 cells, whereas metabolite II showed an IC50 of 61 nM. 17βE2 binding to FLG 29.1 cells was increased after preincubation with metabolites I, III, and V. IP and its metabolites did not induce FR-dependent gene expression in FLG 29.1 and MCF7 cells transfected with a reporter gene and an estrogen response element (ERE). These results suggest that IP effects on osteoclast precursors are not mediated by a direct interaction with the ER, even if a crosstalk between the mechanisms of action of IP and 17βE2 cannot be excluded.

Notes: Summary Rates and yields of nitrite formation in solutions, upon irradiation of aqueous sodium nitrate with ultraviolet light, are found to be consistently and considerably enhanced by their chloride content. The process is investigated under conditions met both, in the laboratory and under sunlight illumination, and the effects of altitude and temperature on the reaction studied in the field, are evaluated.

Notes: Summary Mechanically stirred actinometric solutions of uranyl oxalate, sensitive to radiation below 4300 Å, are found to respond consistently to conditions of solar radiation which are met in Italy during summer, at elevations between 0–3500 metres. The instrumentation developed permits to monitor the course of radiation-dependent chemical reactions in solution, under sunlight.

Notes: Summary The process of nitrite formation, by the action of the near-ultraviolet radiation on sodium nitrate solutions, is investigated both in the laboratory and at different elevations under sunlight. The release of NO x , from solutions irradiated, is found to compete with the generation of nitrite which is proportional to the radiation absorbed by the solution.
