    Description: This review provides a summary of work in the area of ensemble forecasts for weather, climate, oceans and hurricanes. This includes a combination of multiple forecast model results that does not dwell on the ensemble mean, but uses a unique collective bias reduction procedure. A theoretical framework for this procedure is provided, utilizing a suite of models that is constructed from the well-known Lorenz low-order non-linear system. A tutorial that includes a walk-through table and illustrates the inner workings of the multimodel superensemble's principle is provided. Systematic errors in a single deterministic model arise from a host of features that range from the model's initial state (data assimilation), resolution, representation of physics, dynamics, and ocean processes, local aspects of orography, water bodies and details of the land surface. Models, in their diversity of representation of such features, end up leaving unique signatures of systematic errors. The multimodel superensemble utilizes as many as 10 million weights to take into account the bias errors arising from these diverse features of multimodels. The design of a single deterministic forecast models that utilizes multiple features from the use of the large volume of weights is provided here. This has led to a better understanding of the error growths and the collective bias reductions for several of the physical parameterizations within diverse models, such as cumulus convection, planetary boundary layer physics and radiative transfer. A number of examples of forecasts of weather, seasonal climate, hurricanes and the sub-surface ocean where the performance of forecast skills of member models, the ensemble mean, and the superensemble are provided.

    Description: Experimental studies (circular dichroism and ultra-violet (UV) absorption spectra) and large scale atomistic molecular dynamics simulations (accompanied by order parameter analyses) are combined to establish a number of remarkable (and unforeseen) structural transformations of protein myoglobin in aqueous ethanol mixture at various ethanol concentrations. The following results are particularly striking. (1) Two well-defined structural regimes, one at x EtOH ∼ 0.05 and the other at x EtOH ∼ 0.25, characterized by formation of distinct partially folded conformations and separated by a unique partially unfolded intermediate state at x EtOH ∼ 0.15, are identified. (2) Existence of non-monotonic composition dependence of (i) radius of gyration, (ii) long range contact order, (iii) residue specific solvent accessible surface area of tryptophan, and (iv) circular dichroism spectra and UV-absorption peaks are observed. Interestingly at x EtOH ∼ 0.15, time averaged value of the contact order parameter of the protein reaches a minimum, implying that this conformational state can be identified as a molten globule state. Multiple structural transformations well known in water-ethanol binary mixture appear to have considerably stronger effects on conformation and dynamics of the protein. We compare the present results with studies in water-dimethyl sulfoxide mixture where also distinct structural transformations are observed along with variation of co-solvent composition.

    Description: Approximately 10–20 per cent of active galactic nuclei (AGN) are known to eject powerful jets from the innermost regions. There is very little observational evidence if the jets are powered by spinning black holes and if the accretion discs extend to the innermost regions in radio-loud AGN. Here, we study the soft X-ray excess, the hard X-ray spectrum and the optical/UV emission from the radio-loud narrow-line Seyfert 1 galaxy PKS 0558–504 using Suzaku and Swift observations. The broad-band X-ray continuum of PKS 0558–504 consists of a soft X-ray excess emission below 2 keV that is well described by a blackbody ( kT ~ 0.13 keV) and high-energy emission that is well described by a thermal Comptonization (compps) model with kT e ~ 250 keV, optical depth  ~ 0.05 (spherical corona) or kT e ~ 90 keV,  ~ 0.5 (slab corona). The Comptonizing corona in PKS 0558–504 is likely hotter than in radio-quiet Seyferts such as IC 4329A and Swift J2127.4+5654. The observed soft X-ray excess can be modelled as blurred reflection from an ionized accretion disc or optically thick thermal Comptonization in a low-temperature plasma. Both the soft X-ray excess emission when interpreted as the blurred reflection and the optical/UV to soft X-ray emission interpreted as intrinsic disc Comptonized emission implies spinning ( a  〉 0.6) black hole. These results suggest that disc truncation at large radii and retrograde black hole spin both are unlikely to be the necessary conditions for launching the jets.

    Description: The effect of velocity slip at the walls on the linear stability characteristics of two-fluid three-layer channel flow (the equivalent core-annular configuration in case of pipe) is investigated in the presence of double diffusive (DD) phenomenon. The fluids are miscible and consist of two solute species having different rates of diffusion. The fluids are assumed to be of the same density, but varying viscosity, which depends on the concentration of the solute species. It is found that the flow stabilizes when the less viscous fluid is present in the region adjacent to the slippery channel walls in the single-component (SC) system but becomes unstable at low Reynolds numbers in the presence of DD effect. As the mixed region of the fluids moves towards the channel walls, a new unstable mode (DD mode), distinct from the Tollman Schlichting (TS) mode, arises at Reynolds numbers smaller than the critical Reynolds number for the TS mode. We also found that this mode becomes more prominent when the mixed layer overlaps with the critical layer. It is shown that the slip parameter has nonmonotonic effect on the stability characteristics in this system. Through energy budget analysis, the dual role of slip is explained. The effect of slip is influenced by the location of mixed layer, the log-mobility ratio of the faster diffusing scalar, diffusivity, and the ratio of diffusion coefficients of the two species. Increasing the value of the slip parameter delays the first occurrence of the DD-mode. It is possible to achieve stabilization or destabilization by controlling the various physical parameters in the flow system. In the present study, we suggest an effective and realistic way to control three-layer miscible channel flow with viscosity stratification.

    Description: Silicon drift detector based X-ray spectrometer diagnostic was developed to study the non-thermal electron for Aditya tokamak plasma. The diagnostic was mounted on a radial mid plane port at the Aditya. The objective of diagnostic includes the estimation of the non-thermal electron temperature for the ohmically heated plasma. Bi-Maxwellian plasma model was adopted for the temperature estimation. Along with that the study of high Z impurity line radiation from the ECR pre-ionization experiments was also aimed. The performance and first experimental results from the new X-ray spectrometer system are presented.

    Description: An ATP-dependent RNA ligase from Methanobacterium thermoautotrophicum (MthRnl) catalyzes intramolecular ligation of single-stranded RNA to form a closed circular RNA via covalent ligase-AMP and RNA-adenylylate intermediate. Here, we report the X-ray crystal structures of an MthRnl•ATP complex as well as the covalent MthRnl–AMP intermediate. We also performed structure-guided mutational analysis to survey the functions of 36 residues in three component steps of the ligation pathway including ligase-adenylylation (step 1), RNA adenylylation (step 2) and phosphodiester bond synthesis (step 3). Kinetic analysis underscored the importance of motif 1a loop structure in promoting phosphodiester bond synthesis. Alanine substitutions of Thr117 or Arg118 favor the reverse step 2 reaction to deadenylate the 5'-AMP from the RNA-adenylate, thereby inhibiting step 3 reaction. Tyr159, Phe281 and Glu285, which are conserved among archaeal ATP-dependent RNA ligases and are situated on the surface of the enzyme, are required for RNA binding. We propose an RNA binding interface of the MthRnl based on the mutational studies and two sulfate ions that co-crystallized at the active site cleft in the MthRnl–AMP complex.

    Description: A photo multiplier tube (PMT) array based spectroscopic diagnostic with fast time response of 10 μ s and spatial resolution ∼3 cm has been developed and installed on Aditya tokamak to study the spatial and temporal behavior of H α emissions from typical discharges. Collimated light has been collected from the plasma along 16 lines of sight passing through entire plasma poloidal cross section of Aditya and detected by two 8 channels PMT arrays after selecting H α emission using interference filter. The studies are carried out during plasma formation phase of Aditya by changing vertical field and its delay with respect to loop voltage. It is observed that plasma initiated in the high field side in typical discharges of Aditya. The plasma formation position is matched with null field location estimated through simulation.
