Notes: Gliding motility was investigated in the marine diatom, Amphora coffeaeformis. Ultrastructural, biochemical, and pharmacological protocols were employed to probe the possible involvement of cytoskeletal proteins and a secretory process in gliding motility. Motility rate was measured using a video recording apparatus, and the effects of various cytoskeleton-disrupting drugs on motility were tested. Cytochalasins D and E, podophyllotoxin, and vinblastine (all at 25 μUg/ml) reversibly inhibited motility, as did monensin (10 μUM) and pronase (25 μUg/ml). Biochemical protein analysis of whole-cell extracts by one- and two-dimensional polyacrylamide gel electrophoresis revealed polypeptides comigrating with rabbit skeletal muscle actin and bovine brain tubulin; however, specific assays used to separate actin from whole-cell preparations gave ambiguous results. Ultrastructural studies revealed the presence of extracellular material between the raphe canal and the substratum in motile cultures. An assay was devised for the detection of radioactively labeled material (MW 〉 1800 Daltons) released by motile cultures into the culture medium. When cultures were treated with either an anticytoskeletal drug or monensin, motility was inhibited while the amount of measured radioactivity increased over solvent-treated control groups. The results from this study indicate possible roles for both actin- and tubulin-based structures in gliding motility of Amphora. Though secretion may be necessary for gliding to occur, its exact relationship to motility was not deduced. The data obtained in this study are compatible with a theory for the mechanism of gliding which involves the surface translocation of externally exposed membrane proteins against an immobile matrix of substratum-attached secreted material to generate the force required for movement.

Notes: Under negative ion chemical ionization conditions, fluorene yields an anion at [M + 14] mass units. This species has been determined to correspond to [M + O - 2H]-, with the loss of two hydrogen atoms at the C-9 position and addition of oxygen to form a structure similar to 9-fluorenone.

Notes: Viscosities were obtained on oil solutions of two ethylene-propylene-diene terpolymers (EPDMs) and their blends. For the amorphous terpolymer with 59 mol% ethylene, intrinsic viscosities were constant between-10 and 40°C. The viscosities decreased rapidly at low temperature for blends of this material with as little as 20 wt % of a slightly crystalline EPDM with 79 mol% ethylene. Dynamic viscosity measurements on 1.0% solutions of blends likewise gave considerably smaller values at low temperature than measurements on an amorphous EPDM of similar molecular weight. The data are in agreement with the view that longer ethylene sequences that crystallize in the bulk polymer can organize in oil into ordered domains that interfere less with flow than the disordered amorphous polymer regions. © 1993 John Wiley & Sons, Inc.

Notes: Liquid SO2 sealed into tubes made of a fluorocarbon copolymer permeates the walls of the tube at a temperature-dependent but accurately reproducible rate. Sulfur dioxide dispensers made in this way are called permeation tubes and are useful for calibrating instruments that measure SO2 concentrations in air. The National Bureau of Standards calibrates SO2 permeation tubes and makes them available as Standard Reference Materials. The permeation rate in a batch of nominally identical tubes varies enough that each Standard Reference Material tube must be individually calibrated. Changes in the length or radial dimensions of the tubes are much too small to explain most of this variation. An excellent (negative) correlation is found between the measured permeation rate and the density of the polymer (or weight per unit length). Since both the measured density and the permeation rate for this semi-crystalline polymer depend upon morphological factors, but in different ways, x-ray diffraction measurements of the thickness and orientation of the lamellar crystals were made and a mathematical model was set up to identify the morphological factors which can cause variations in the permeation rate.

Notes: We have been investigating the synthesis of endohedral metallofullerenes since we successfully opened an orifice on the framework of C60 in the form of a cobalt complex having the metal sitting on top of the opening. A second approach aims at the synthesis of spherically-shaped acetylenic macrocycles, which are expected to rearrange to endohedral metal complexes of fullerenes in a controlled process analogous to the gas-phase coalescence of mono- and polycyclic polyynes during fullerene formation by the graphite evaporation method. Since the potential benefits from obtaining endohedral metal complexes of C60 are enormous, both approaches are being actively pursued in our group.
