Some methods for the determination of the distribution of the substituents were compared on amyloses and lichenins acetylated to different degrees of substitution in a homogenous medium: (1) tritylation, (2) nitrogen dioxide-oxidation of free primary hydroxyl groups, and (3) oxidation of the secondary hydroxyl groups with sodium periodate. At degree of substitution below 2 all three methods gave only sligtly differing results. The 3rd method showed the best, and the 2nd method the lowest reproducibility. The determination of the distribution of substituents gave the following results. (1) If the acetates were prepared by partial esterification, the primary hydroxyl groups were substituted preferentially at the beginning of the reaction. Low concentrations and reaction temperatures increase this effect, but do not allow to prepare primary monoacetates. (2) Acetates obtained by partial alkaline or acidic saponification from the triacetate showed a statistical distribution of the substituents. Both effects demonstrate that the bond energy is responsible for the distribution of the substituents, while steric factors are active only at high degrees of substitution.
Notes:
