    Notizen: Abstract Poly(A)-containing messenger RNA was purified from polyribosomes isolated from the primary leaves of 7-day-old dark-grown seedlings of Phaseolus vulgaris var. Masterpiece. Analysis of the messenger RNA on 2.4% polyacrylamide gels showed that it consists of a heterogeneous population of molecules with an average molecular weight of 500,000. The nucleotide composition of the RNA was 16.0% cytidylic acid, 39.4% adenylic acid, 21.3% guanylic acid and 23.2% uridylic acid. Based on the degree of resistance of the RNA to digestion with ribonucleases A and T1 the average length of the poly(A) sequence was calculated to be 120 nucleotides. No significant differences in mobility in polyacrylamide gels, nucleotide composition or polyadenylic acid content were found between the poly(A)-containing mRNA from polyribosomes of primary leaves of dark-grown plants and those given a 16 h white light treatment. Purified poly(A)-containing mRNA was shown to direct the incorporation of [35S]methionine into proteins in an in vitro protein-synthesising system from wheat germ. The protein products were fractionated according to molecular size by electrophoresis in 15% polyacrylamide/urea/SDS gels and the protein bands were detected by fluorography. Messenger RNAs directing the synthesis of three polypeptides with molecular weights of 34,000, 32,000 and 25,000 were detected in polyribosomes of plants following white light treatment. These messenger RNAs were absent, or present in much lower amounts, in polyribosomal messenger RNA from leaves of dark-grown plants, although they were present in total cell poly(A)-containing RNA. This indicates that certain messenger RNAs may be stored in the dark and that light stimulates these RNAs to engage in polyribosome formation. Continuous far-red (730 nm) irradiation for 4 h also caused the appearance of these messenger RNAs in the polyribosomes although 5 min red light followed by 4 h darkness had little effect. This suggests that phytochrome acting in the “high energy” mode, may be the photoreceptor responsible for initiating the response.

    Notizen: Abstract The internal potential of flexor cells in slices of the laminar pulvini of Phaseolus coccineus has been recorded by standard microelectrode techniques in 100 eq m-3 external salt solutions of various ionic compositions. The measured values are between-15 and-60 mV depending on the external medium. Treating the results with the Goldman equation yields the following relative permeabilities: K+, 1.00; Na+, 0.24; Cl-, 0.19; NO 3 - , 1.6. The membrane potential was only slightly sensitive to external pH and Ca2+. Metabolic inhibitors (azide, cyanide and salicylhydroxamic acid, carbonyl cyanid m-chlorphenyl hydrazone) caused only slight depolarizations (ca. 4 mV), which differed from the ion-induced changes by their slow time courses. The results are consistent with the hypothesis that the relatively impermeable Cl- is actively transported and osmotically efficient, whereas the well-permeable K+ passively follows Cl- to maintain electroneutrality and is osmotically of only minor significance.

Bean cells grown in suspension culture on maintenance medium have been shown to differentiate (xylem and phloem production) when they are transferred to an induction medium containing an increased ratio of naphthylacetic acid to kinetin.-2. Conditions for the extraction and isolation of undegraded mRNA are described. The mRNA has been translated by 2 in vitro protein synthesizing systems; the wheat germ and the mRNA-dependent-reticulocyte-lysate preparations.-3. A greater proportion of the mRNA from cells on maintenance medium coded for high molecular weight products compared with that from cells on induction medium.-4. mRNA from cells on induction medium coded for a relatively large amount of a few polypeptides whereas that from cells on maintenance medium coded for a large number of polypeptides all present in fairly equal amounts.

    Notizen: Abstract Chlorophyllase (chlorophyll-chlorophyllidohydrolase, EC 3.1.1.14) was isolated and purified from Phaseolus vulgaris L. chloroplasts and etioplasts dissolved in 1% Triton X-100 and 10% glycerol. A 100 and 40-fold purification, respectively, was achieved. Enzyme preparations from both sources had similar affinities for chlorophyll a when assayed in a Triton X-100 medium. When electrophoresed in sodium dodecyl sulphate polyacrylamide gels the major band in both preparations migrated as a peptide of 30,000 daltons. Chlorophyll containing liposomes were also used as a substrate for chlorophyllase. The rate of hydrolysis did not follow Michaelis-Menten kinetics. When chlorophyllide a or methyl chlorophyllide a was incorporated in the liposomes, then in the presence of phytol dissolved in methanol, methylchlorophyllide a and chlorophyll a were shown to be synthesized. Apparently the purified enzyme in the presence of lipids, is endowed with both synthetic and hydrolytic activity.

    Notizen: Abstract Gibberellic acid (GA3), applied as a dispersion in aqueous lanolin to the stumps of decapitated stems of P. vulgaris plants, was found to promote the transfer of 14C-and 32P-labelled assimilates to the site of hormone application. Measurements of the component transfer processes, operating between source and sink (site of hormone application), showed that GA3 was not acting to promote assimilate transfer by increasing the photosynthetic rate of, or the assimilate export rate from the source, nor by altering the mobilizing ability of the competing root sink. Here, it also was found that the time between GA3 application and detection of an enhanced transport flux was independent of the length of the transport pathway. Overall, the evidence obtained indicated that GA3 was not acting on any transfer process remote from its point of hormone application but was acting locally at this latter point.

    Notizen: Abstract Leaf water potentials of Phaseolus vulgaris L. plants exposed to a -3.0 bar root medium were reduced to between -7 and -9 bars within 25 min and remained constant for the next several hours. This treatment led to considerable variation between leaves in both abscisic-acid (ABA) content and Rs, although the two were well correlated after a 5-h treatment. There was an apparent 7-fold increase in leaf ABA levels necessary to initiate stomatal closure when plants were exposed to a -3.0 bar treatment, but when plants were exposed to a -5.0 bar stress Rs values increased prior to any detectable rise in ABA levels. To explain these seemingly contradictory results, we suggest that the rate of ABA synthesis in the leaf, rather than the total ABA content, determines the status of the stomatal aperture.

    Notizen: Summary Fertilization and early embryo and endosperm development were examined in Phaseolus vulgaris x P. acutifolius, P. vulgaris x P. lunatus crosses and their reciprocals. The number and length of pollen tubes were not different between selfings and interspecific crosses. Fertilization was completed in all matings and the time of fertilization was maternally dependent which may reflect the degree of maturation of embryo sacs at pollination. A large difference between reciprocal crosses was found in the time of endosperm and embryo division in relation to the time of fertilization. When P. vulgaris was the female parent and P. acutifolius the male parent, endosperm division occurred at the same time as in P. vulgaris upon selfing, while in P. vulgaris x P. lunatus crosses the time of endosperm division was intermediate as compared with the two parents. The time lapse between fertilization and endosperm and embryo division in P. acutifolius x P. vulgaris crosses was longer than in either parent upon selfing. In P. lunatus x P. vulgaris crosses, endosperm division occurred in only 7–12% of the ovules at 72 hours after pollination. Embryo development in these ovules was limited to the four cell stage although the endosperm was at the free nuclei stage. The severe delay in embryo and endosperm divisions may be the major cause of early pod abscission in P. lunatus x P. vulgaris crosses.

    Notizen: Summary The influence of genotypic combinations on the growth of hybrid embryos between Phaseolus vulgaris and P. lunatus, and between P. vulgaris and P. acutifolius was examined. All embryos obtained from P. vulgaris × P. lunatus crosses developed only to a stage which appears to be comparable to the pre-heart-shape stage of selfed embryos. Reciprocal crosses were attempted, but pods abscised at a very early stage. Embryos derived from P. vulgaris × P. acutifolius and reciprocal crosses attained the cotyledon stage although no mature seeds were formed. A distinct characteristic of these embryos was the uneven development of the two cotyledons. The rate of growth and final size of these hybrid embryos seemed to be influenced by the genotypes of both parents. Immature embryos were cultured on defined medium and the effects of glutamine and gibberellin (GA3) were examined. Glutamine was effective in increasing the survival rate; gibberellin had no apparent effect. Plants derived from cultured embryos of P. vulgaris × P. lunatus, P. vulgaris × P. acutifolius and P. acutifolius × P. vulgaris were obtained.
