    Description: The spread of farming out of the Balkans and into the rest of Europe followed two distinct routes: An initial expansion represented by the Impressa and Cardial traditions, which followed the Northern Mediterranean coastline; and another expansion represented by the LBK (Linearbandkeramik) tradition, which followed the Danube River into Central Europe. Although genomic data now exist from samples representing the second migration, such data have yet to be successfully generated from the initial Mediterranean migration. To address this, we generated the complete genome of a 7,400-year-old Cardial individual (CB13) from Cova Bonica in Vallirana (Barcelona), as well as partial nuclear data from five others excavated from different sites in Spain and Portugal. CB13 clusters with all previously sequenced early European farmers and modern-day Sardinians. Furthermore, our analyses suggest that both Cardial and LBK peoples derived from a common ancient population located in or around the Balkan Peninsula. The Iberian Cardial genome also carries a discernible hunter–gatherer genetic signature that likely was not acquired by admixture with local Iberian foragers. Our results indicate that retrieving ancient genomes from similarly warm Mediterranean environments such as the Near East is technically feasible.

    Description: Polycyclic aromatic hydrocarbons (PAHs) are compounds that can be quantified by fluorescence due to their high quantum yield. Haloalkalitolerant bacteria tolerate wide concentration ranges of NaCl and pH. They are potentially useful in the PAHs bioremediation of saline environments. However, it is known that salinity of the sample affects fluorescence signal regardless of the method. The objective of this work was to carry out a comparative study based on the sensitivity, linearity, and detection limits of the excitation, emission, and synchronous fluorescence methods, during the quantification of the residual anthracene concentration from the following haloalkalitolerant actinomycetes cultures Kocuria rosea, Kocuria palustris, Microbacterium testaceum, and 4 strains of Nocardia farcinica, in order to establish the proper fluorescence method to study the PAHs biodegrading capacity of haloalkalitolerant actinobacteria. The study demonstrated statistical differences among the strains and among the fluorescence methods regarding the anthracene residual concentration. The results showed that excitation and emission fluorescence methods performed very similarly but sensitivity in excitation fluorescence is slightly higher. Synchronous fluorescence using  nm is not the most convenient method. Therefore we propose the excitation fluorescence as the fluorescence method to be used in the study of the PAHs biodegrading capacity of haloalkalitolerant actinomycetes.

    Description: Erythroid cells undergo enucleation and the removal of organelles during terminal differentiation. Although autophagy has been suggested to mediate the elimination of organelles for erythroid maturation, the molecular mechanisms underlying this process remain undefined. Here we report a role for a Bcl-2 family member, Nix (also called Bnip3L), in the regulation of erythroid maturation through mitochondrial autophagy. Nix(-/-) mice developed anaemia with reduced mature erythrocytes and compensatory expansion of erythroid precursors. Erythrocytes in the peripheral blood of Nix(-/-) mice exhibited mitochondrial retention and reduced lifespan in vivo. Although the clearance of ribosomes proceeded normally in the absence of Nix, the entry of mitochondria into autophagosomes for clearance was defective. Deficiency in Nix inhibited the loss of mitochondrial membrane potential (DeltaPsi(m)), and treatment with uncoupling chemicals or a BH3 mimetic induced the loss of DeltaPsi(m) and restored the sequestration of mitochondria into autophagosomes in Nix(-/-) erythroid cells. These results suggest that Nix-dependent loss of DeltaPsi(m) is important for targeting the mitochondria into autophagosomes for clearance during erythroid maturation, and interference with this function impairs erythroid maturation and results in anaemia.     Description: Apoptosis in the immune system is critical for maintaining self-tolerance and preventing autoimmunity. Nevertheless, inhibiting apoptosis in lymphocytes is not alone sufficient to break self-tolerance, suggesting the involvement of other cell types. We investigated whether apoptosis in dendritic cells (DCs) helps regulate self-tolerance by generating transgenic mice expressing the baculoviral caspase inhibitor, p35, in DCs (DC-p35). DC-p35 mice displayed defective DC apoptosis, resulting in their accumulation and, in turn, chronic lymphocyte activation and systemic autoimmune manifestations.     Description: Notch signaling affects many developmental and cellular processes and has been implicated in congenital disorders, stroke, and numerous cancers. The Notch receptor binds its ligands Delta and Serrate and is able to discriminate between them in different contexts. However, the specific domains in Notch responsible for this selectivity are poorly defined. Through genetic screens in Drosophila, we isolated a mutation, Notch(jigsaw), that affects Serrate- but not Delta-dependent signaling. Notch(jigsaw) carries a missense mutation in epidermal growth factor repeat-8 (EGFr-8) and is defective in Serrate binding. A homologous point mutation in mammalian Notch2 also exhibits defects in signaling of a mammalian Serrate homolog, Jagged1.     Notes: The dependence of S impurity incorporation into GaAs vs growth rate, and substrate temperature has been investigated during vapor phase epitaxial growth. The specific technique used, close space vapor transport, has been selected to allow the growth rate Vg and the substrate temperature θ to vary independently and in very large ranges. The concentration ND of incorporated impurities has been determined for a variety of values of Vg and θ, using an electrical method. Comparison of the experimental data with a simple model gives the variation of the sticking coefficient versus temperature. The model assumes that the impurities incorporated are those which stick on the surface during the time a monolayer is grown, when desorption can be neglected during this time. This sticking coefficient is thermally activated with an activation energy of 2 eV. The agreement between the experimental data and the variation of ND vs Vg and θ deduced from this model, confirms its validity and demonstrates that high purity GaAs can be grown by this technique under high growth rate conditions. © 1995 American Institute of Physics.

    Notes: Photoacoustic signals generated in absorbing media can be used for real-time alignment of laser cavity components, for measurements of positions and diameters of pulsed laser beams, and for control of the degree of overlap of two or more, focused and unfocused pulsed laser beams. The developed techniques require an absorbing medium and a piezoelectric detector. Measurements and control can be performed by real-time inspection of the acoustic signals in an oscilloscope. These methods are convenient for experiments in nonlinear optics in which the power requirements lead to focusing and superposition of the beams, and for alignment of laser cavities in the liquid phase as well as gas and solid state infrared lasers. © 1995 American Institute of Physics.

    Notes: Time-resolved spectroscopy has been used in a Xe pulsed discharge. The analysis reveals a delay in the onset of emission which is characteristic of the ion, suggesting that this can be related to the time for exciting each ionization stage, for given discharge conditions. This lag is longer for higher ions. The delay determination was done characterizing it for several lines of each ion; only some are listed below. The results enable a characterization of all the nonclassified Xe lines which are known to lase in the visible and the near UV, as belonging to three different groups of ions. One has temporal lags which coincides with those of Xe iv well-classified lines; the other two, each having longer time delays, are assigned as transitions in higher ions.
