    Notes: Conditions are reported for the reverse-phase HPLC analysis of gossypol and of some of its derivatives, including anhydrogossypol, gossypolone, gossypol hexaacetate, and Schiff's base derivatives. It is shown that field desorption (FD) and chemical ionization (CI) mass spectrometry have advantages over electron impact (EI) mass spectrometry for the characterization of these compounds and that combined high-performance liquid chromatography/mass spectrometry (LC/MS), especially when used in the CI mode, is particularly effective.

    Notes: An improved analog four-channel selected ion recording system is described, in which major modifications permit a decrease in the dwell time to 33 ms per channel, thus minimizing the mass cycling error. Synchronization with a sinusoidal sweep voltage superimposed on the normal accelerating voltage (8 kV) enables two channels to be monitored simultaneously in real time and each mass to be recorded continuously. These improvements allow measurement of ion current ratios with a precision of 0.2% over a wide dynamic range, permitting accurate determination of isotopic enrichment in biomedical assays even when this enrichment is derived from a single label. Use of the system is illustrated by the analysis of palmitate turnover in dogs (using [1 - 13C] palmitic acid) with an average standard deviation corresponding to the detection of 0.04% excess of [1 - 13C] palmitate.

    Notes: A method has been developed for the quantitation of the putative phenolamine neurotransmitter octopamine, and its precursor tyramine, in brain tissue. The procedure employs methane chemical ionization of the pentafluoropropionate derivatives of octopamine and tyramine together with the use of deuterated internal standards and selected ion monitoring. Deuterated analogues of octopamine and tyramine are added to brain homogenates in aqueous perchloric acid and ion exchange is used to isolate the brain amines. The method is capable of measuring 20 pg of octopamine and tyramine. The measured concentration (ng g-1 wet tissue) of octopamine and tyramine in rat brain was as follows: whole brain (less cerebellum) (0.6 and 2.2); hypothalamus (3.2 and tyramine value not statistically significant); striatum (0.5 and 11.8) and cortex (0.6 and 1.0). Administration of pargyline resulted in an increase (around ten-fold) in octopamine and tyramine concentration in all the above brain regions. In contrast α-methyltyrosine produced only a small increase (50%) in the concentration of tyramine in the striatum.

    Notes: The value of selected ion monitoring in analyzing biological radio isotope incorporation experiments by radiogas chromatography mass spectrometry is illustrated with reference to the biosynthesis of the mycotoxin mycophenolic acid and the mode of action of the anticholesterolemic drug 20,25-diazacholesterol. It is shown that the increased sensitivity and specificity of the selected ion monitoring mode detector permits straightforward detection and identification of the relatively small cellular pools associated with metabolic intermediates. The computer program RADSIM is described. Problems that still exist in using radiogas gas chromatography mass spectrometry technology to analyse isotope incorporation experiments are discussed.

    Notes: Gas chromatography using a short packed column (45 cm, 0.2 cm i.d., 2% OV-101 on Gas-Chrom Q) with mass spectrometric detection in the selected ion monitoring mode has been found satisfactory for the analysis of lower as well as higher polybrominated biphenyls. Acceptable sensitivity (〈1 ng) may be achieved for this method by focusing selectively at either the low (m/z 20-600) or the high (m/z 600-1000) range of the quadrupole filter (low range for mono- through hexabromobiphenyl, high range for hexa- through decabromobiphenyl). A tuning technique has been developed for low range and high range polybrominated biphenyls using the ion abundances of perfluorotributylamine as a standard. Standard ions for the quantitation of mono- through decabromobiphenyls were selected and validated. The technique was applied to the analysis of a variety of environmental samples.

    Notes: Two common methods of applying dead-time corrections in EDS systems, both of which are carried out internally in the electronics, are examined. The Barnhart method shows serious discrepancies when making dead-time corrections and in certain cases no correction is made at all. The problem is associated with the reliance on the fast amplifier to count all x-rays collected by the detector. In practice, however, the fast amplifier does not always succeed in carrying out this function since x-rays with energies 〈 1500 eV may fall below its threshold level. The problems become worse when a formvar window is substituted for beryllium since a greater proportion of low energy x-rays can now reach the detector. The magnitude of the discrepancy is shown also to be a function of specimen and electron accelerating voltage. Similar experiments with an EDS system using the Covell method show that no such discrepancies arise.

    Notes: A study of the low-energy region (below 1. 5 keV) of energy dispersive x-ray spectra has been carried out. Firstly, because some counting systems do not provide a reliable dead time correction, a new method of dealing with the problem is given. Next, the effects of different positioning of the energy discriminator within the analyser circuitry are discussed with reference to the consequences for light element analysis. Electronic noise counts can present a problem for analysis of x-rays of less than 300 eV and a straightforward method for noise removal is described. Finally, an unexpected artifact in the spectrum below 500 eV is identified; its size is related to the total number of counts in the spectrum and a practical method of dealing with it is demonstrated. Once the above factors have been taken into account a low energy spectrum is produced, consisting solely of characteristic x-ray lines and a true continuum background.
