    Notes: The variable-temperature proton nmr spectra of the oligoribonucleotides in the series CpApX and the series ApGpX, X = A, G, C, U, together with the parent dimers CpA and ApG have been measured. A complete analysis of all the nonexchangeable base proton resonances and ribose H-1′ proton resonances was made. The presence of trends in the shielding abilities of the various bases at both the nearest-neighbor and next-nearest-neighbor positions were identified. The observed shieldings could be used to predict the chemical shifts of protons in related systems. Based on the empirical results from ribodinucleoside monophosphates, the temperature-dependent behavior of the J1′2′ coupling constants of the triribonucleotides suggested that the compounds in the CpApX series stacked from the 5′-end to the 3′-end, while those in the ApGpX series stacked from the 3′-end to the 5′-end.

    Notes: The exo- and endo-irontricarbonyl complexes of 5,6-dimethylidene-2-exo-norbornyl alcohols 10x, 10n, p-bromobenzenesulfonates 11x, 11n, acetate 12x and of the 2,3-dimethylidene-7-anti-norbornyl alcohols 17x, 17n, p-bromobenzenesulfonates 19x, 19n and acetates 20x, 20n have been prepared. The SN1 buffered acetolyses of 11x, 19x and 19n gave 12x, 20x and 20n, respectively (retention of configuration). The first-order rate constants of the acetolyses have been evaluated and compared with those of the acetolyses of the uncomplexed 5,6-dimethylidene-2-exo-norbornyl (14) and 2,3-dimethylidene-7-anti-norbornyl p-bromobenzenesulfonates (18). A rate retardation effect of ca. 1.5 · 105 was measured for 11x → 12x (65°) compared with the acetolysis of 14. The retardation effect is larger (〉 5 · 107) with 11n. Contrastingly, the acetolysis 19x → 20x was slightly accelerated with respect to that of the uncomplexed p-bromobenzenesulfonate 18. An unsignificant rate-retardation effect was measured for the acetolysis 19n → 20n. The results are interpreted in terms of competitive inductive destabilization and charge-induced dipole stabilizing interaction by the exocyclic diene-iron tricarbonyl fragment. PMO. arguments give a rationale for the difference in polarizability between the diene-Fe(CO)3 group in 19 and that in the endo-7-norbornadienyl-iron tricarbonyl system.

    Notes: The mobility of polymer chain segments is shown to play a major role in the diffusion of dyes in nylon 66. The rate of dye diffusion has been related to the time-dependent mechanical properties, such as creep and stress relaxation, which are controlled by the mobility of the chain segments. The theoretical relations derived by Fujita et al. relating the diffusion of small molecules in amorphous polymers to other properties are shown to be applicable for the dyeing of semicrystalline nylon. Data showing the effects of surface area, temperature, and the amount of dye absorbed at saturation on the dyeing rate are presented and discussed.

    Notes: Three high molecular weight polycaprolactones (M̄w = 35,000, 18,600, and 7,130) were utilized as the sole carbon source by five of six fungi tested by the American Standards for Testing and Materials (ASTM) agar plate method. The fungi were Aspergillus flavus, A. niger, A. fumigatus, Chaetomium globosum, Pencillium funiculosum, and a Fusarium sp. Quantitative analysis of degradation was performed using gel permeation chromatography (GPC). GPC analysis demonstrated differences between the activities of organisms which appeared similar by the ASTM method, and showed that, while all molecular weight species within each polymer were hydrolyzed, in several cases low molecular weight end products were not assimilated. Depending on the organism, the dominant factor determining degradability was either polymer molecular weight or degree of crystallinity.

    Notes: A correlation for estimating the diffusion coefficients of protein molecules is presented. The correlation is based upon literature values of the protein diffusion coefficients and molal volumes for 143 proteins. The correlation can be used for the estimation of diffusion coefficients using only molecular weight. Accuracy is such that a linear regression on 301 proteins showed 75% of the diffusion coefficients estimated fell within 20% of the experimental values. The relationship between this correlation, the Stokes-Einstein equation, and the Wilke-Chang correlation is discussed.

    Notes: Continuous hydrolysis of triglyceride in organic solvent systems using Rhizopus arrhizus mycelia as a source of insolubilized lipase has been studied in packed-bed and stirred-tank reactors. Typically a packed bed reactor containing 1 g of mycelia fed at 1 mL/min with a solution of 2.5% (w/v) olive oil in di-isopropyl ether gave a fatty acid yield of 45% at 30°C. The optimum water concentration was found to be 0.17% (w/v) except under conditions of high oil feed concentration and high yield where no optimum was established. No temperature optimum was observed over the range 20-55°C. Calculated activation energies of 13-20 kJ/mol, depending on temperature, were lower, while Km(app) values of 0.1-0.3M were higher than those for hydrolysis in conventional aqueous emulsion systems. No evidence of any significant diffusional limitation, which could account for these values, was obtained. The mycelia showed a loss of activity of 0.6-1.0%h at 30°C. The packed bed proved markedly superior to the stirred tank for this system.

    Notes: The strength and rheological characteristics of isoelectric soya protein precipitate, prepared in continuous tubular and batch-stirred tank reactors, were determined in relation to the separation efficiency and sludge discharge characteristics of intermittent disk and scroll discharge centrifuges. The batch tank precipitate showed greater resistance to aggregate shear breakup than the tubular reactor precipitate. This difference was of significance to the separation efficiency of the scroll centrifuge only. The tubular reactor precipitate sludge showed greater resistance to shear deformation and resulted in drier sludge for a given scroll differential speed. some compressive dewatering occurred for both types of precipitate sludge but shear-induced dewatering is proposed as the major mechanism. This study illustrates the need to integrate the design of protein precipitate formation and centrifugal recovery operations.

    Notes: An improvement in the resistance of isoelectric soya protein precipitate to capillary shear disruption was achieved by aging in a stirred tank. The aggregate strength was found to depend on the extent and duration of agitation prior to exposure to shear. An optimum value of an aging parameter Gt ∼ 105 was determined, where G is the rms velocity gradient in the aging vessel and tis the time of aging. The disruption of precipitate aggregates by exposure to high rates of shear for short times was dependent on the rate of shear and time of exposure. The dominant mechanism of aggregate breakup was fragmentation, with erosion occurring to a lesser extent. The size of the fragments produced by shear disruption was weakly dependent on the rate of shear, with higher rates of shear producing smaller fragments.

    Notes: A theoretical description of a low-frequency, dual-source acoustic treatment of protein precipitates is presented. Two mechanisms for the potential improvement of the centrifugal separation of precipitates are proposed. They are acoustic aggregation, which causes an increase in particle size, and aggregate deformation, which results in an increase in aggregate density. Experiments with isoelectric soya protein precipitate indicated that the extent of ageing prior to acoustic conditioning is important. Support for both aggregation and dewatering was found and a maximum increases in the particle settling velocity of 20% was observed.
