Notes: Abstract The Ca2+ antagonist binding sites associated with the voltage dependent calcium channel in rabbit myocardium were found to distribute with the sarcolemmal Na− + K+ ATPase and adenylate cyclase activities during subcellular fractionation on sucrose-density gradients. The equilibrium dissociation constants (KD) for the binding of [3H]nitrendipine and [3H]verapamil were 0.31 ± 0.04 nM and 4.1 ± 0.5 nM respectively, and displayed an average density of 0.55 ± 0.05 pmol/mg and 0.4 ± 0.03 pmol/mg protein respectively for the most enriched membrane fraction. The Ca2+2 antagonist binding sites were solubilized from the membranes with the detergent 3-[(3-cholamidopropyl)dimethylammonio]propanesulfonate, and specific binding sites for [3H]PN200-110, [3H]verapamil and [3H]diltiazem were isolated on a wheat-germ lectin column. The binding sites for [3H]PN200-110 were enriched about 2500 fold as compared with the original homogenate and displayed a density of 28.5 ± 8 pmole/mg protein in the isolated fraction. Sodium dodecyl sulfate gel electrophoresis of the isolated drug binding proteins indicated enrichment of proteins of Mr 170000, 140000, 130000, 100 000 and 53000. The isolated receptor contained an intrinsic kinase activity that phosphorylated glycoproteins of Mr 170 000 and 53000. Exogenously added cAMP-kinase stimulated phosphorylation of the 170000, 100000, 53 000 and 28000 Mr glycoproteins in the receptor fraction. The results of this study indicate that the binding sites for [3H]nitrendipine, [3H]PN200-110, [3H]verapamil and [3H]diltiazem residue on glycoprotein(s) which are of sarcolemmal origin, and co-purify together on wheat germ lectin columns. The polypeptide composition of the Ca2+ antagonist binding sites from cardiac muscle appears to be very similar to that of the dihydropyridine receptor in skeletal muscle.

Notes: Abstract Tritiated analogues of the Ca2+ channel blockers such as [3H] PN200-110, [3H] verapamil and [3H] diltiazem have been used to identify and isolate Ca2+ antagonist receptors. The Ca2+ antagonist binding sites were solubilized from skeletal muscle transverse tubules with the detergent CHAPS and purified by wheat germ lectin column chromatography and sucrose density gradient centrifugation. The isolated proteins retained their ability to bind the various classes of Ca2+ channel blockers. Polypeptides of 170, 150, 108, 56, and 32 kDa were found to be present in the purified receptor fraction when analysed by sodium dodecyl sulfate polyacrylamide gel electrophoresis under non-reducing conditions. The apparent molecular weight of the 170 kDa polypeptide changed to 145 kDa in the presence of reducing agents, as where the apparent molecular weight of the 150, 108, 56 and 32 kDa peptides remained unchanged. An endogenous protein-kinase present in the original membranes, co-purified with the receptor and stimulated the phosphorylation of the 150 and 56 kDa polypeptides in the isolated fraction.

Notes: Summary In normal (untreated) rats the mean length ratio of postsynaptic to presynaptic membrane was 2.7±0.8 for neuromuscular junctions of slow-twitch soleus muscle fibres and 4.2±1.0 for neuromuscular junctions of fast-twitch extensor digitorum longus muscle fibres; this difference was significant (P〈0.001). After experimental double innervation by fast and slow muscle nerves for four months, the ratio was (1) 2.9±0.8 for the original slow-twitch fibre end-plate and 2.8±0.8 for the newly established one, both not significantly different from that of the normal slow-twitch fibres; and (2) 2.2±0.5 for the original fast-twitch fibre end-plate and 2.2±0.7 for the newly established one, both significantly smaller than that of the normal fast-twitch fibres (P〈0.001). This means that the double innervated slow-twitch muscle fibres retained their original neuromuscular junction type, whereas the doubly-innervated fast-twitch muscle fibres underwent a dramatic transformation of their neuromuscular junction from the fast-muscle to the slow-muscle type. In both doubly innervated fibres, the ultrastructural characteristics of neuromuscular junctions, whether altered or not, were identical at both end-plate regions.

Notes: Abstract In this paper, moiré contour fringes is applied to study the femur's articular surface of the knee of Pongidae. The preliminary division of the femur's articular surface of knee into three types is proposed. The moiré contour fringes ofthe medial condyle is taken as a mark according to the references. Owing to the fact that the moiré contour fringes obtained from experiments after the 2nd order of fringe basically follow a certain rule, an investigation is made on the distribution of the angle α which is defined as the angle of the major axis of the 2nd order's near-oval shaped moiré contour fringe on the medial condyle with the horizontal axis. Preliminary distribution graphs are given in the paper.

Notes: Abstract In this paper, the translation-torsion coupling earthquake responses. of single-story ecoentric factory buildings with consideration of whole-space-work of the roof in 3-dimension non-inertia system are analysed and studied; and its calculation model has been established. And what is more, corresponding motion equations of earthquake responses have been derived, analysed and discussed. And it points out that under the action of the earthquake including rotational component, the principle of superposition of independent action for each earthquake component and mode-superposition method of dynamic responses analysis are already not applicable. In this paper, the translation-torsion coupling earthquake responses of some single-story eccentric factory building are calculated and analysed by comparison methods without consideration of the earthquake torsional component. The conclusion is that when whole-space-work of the roof is considered, the results of calculation are more conformable to the actual carthquake damage. Moreover, this paper gives its simplified method for calculation and conditions in which the deformation of the roof can be neglected.

Notes: Abstract In this paper. first we introduce the shooting method (including the method of adjoints and the method of quasilinearization) for two-point boundary value problems of ordinary differential equations with parameters, and give out an example of solving hydrodynamic lubrication equation by using the shooting method. The calculated results are satisfactory.

Notes: Abstract Phosphorus segregation to the free surface of an amorphous Fe44Ni36P14B6 alloy during annealing in the temperature range 180 to 300° C has been studied by Auger electron spectroscopy. The phosphorus concentration increased with annealing temperature with an abrupt change appearing at about 250° C. At 276° C the phosphorus Peak Height Ratio on the surface was three times as high as that of a sputtered surface. A step-heating scheme has been employed to observe the surface segregation of phosphorus in the temperature range 25 to 290° C. On heating, the segregation behaviour can be correlated with a degradation in toughness properties. It is concluded that the segregation of phosphorus onto the free surface of this amorphous alloy is an indication of some local atomic migration processes, for example clustering, occurring inside the bulk.

Notes: This paper presents a simple extension of the patch test to mixed formulations to provide the necessary and sufficient conditions for convergence. The general algebraic conditions of Babuska and Brezzi are given a simple form and a conceptual application of the patch tests serves to point out the instability of several well known formulations for incompressible problems. The test is also applied to mixed displacement - strain formulations of elasticity, and limiting formulations are noted.

Notes: Abstract Pure iron foils have been implanted with Ti and with Ti plus C and characterized by conversion electron Mössbauer spectroscopy (CEMS) and Auger electron spectroscopy (AES). Both paramagnetic and magnetic phases are observed and attributed to amorphous Fe−Ti−C alloys of differing Ti and C contents. Estimates of the thicknesses of the amorphous layers based on the CEMS data are in good agreement with the AES concentration depth profiles.
