    Notes: : Based on the high level of extractability of myosin subunits (light chains), even after prolonged heat treatment of muscle, a new method to evaluate the dark muscle content in the fish meat and products of mackerel is proposed. Tissue-specific rabbit antisera with myosin light chains (A1 from ordinary muscle and D1 from dark muscle) from mackerel Scomber japonicus were obtained. Mackerel meat paste (surimi) was dissolved in 8 M urea containing 1% SDS, and diffused on agar plates containing antiserum against A1 or D1 by single radial immunodiffusion (SRID). The results obtained showed that the area of halos formed in the plates was quite proportional to the content of dark muscle.

    Notes: The resurgence in mycobacterial infection worldwide has led to renewed attention to the pathogenesis of Mycobacterium species. Although interferon-γ (IFN-γ) is a principal mediator of macrophage activation, macrophages infected with Mycobacterium are poor in response at the cytokine. However, the molecular mechanisms underlying mycobacterial infection remain unclear. The purpose of this study was to elucidate the mechanism of the poor response to IFN-γ in mycobacterial infection. Our data clearly demonstrate that this is due to induction of suppressor of cytokine signal (SOCS) negative regulators of IFN-γ signal transduction that closely correlates with the inhibition of JAK/STAT signaling and gene expression stimulated by IFN-γ. Mycobacterium bovis bacillus Calmette-Guérin infection induces the production of SOCS-1 and SOCS-3 in murine J774 macrophages. The level of SOCS-1 mRNA increased 1 h and reached a maximum 3 h after the addition of the bacteria. SOCS-3 mRNA expression appeared as early as 1 h after the infection. We also observed that trehalose 6,6′-dimycolate/cord factor, a major component of the Mycobacterium tuberculosis cell wall, induces expression of SOCS and inhibits IFN-γ-stimulated phosphorylation of STAT1 extensively in the cells. The results in this study suggest that a molecular mechanism of mycobacterial infection affects the unresponsiveness to IFN-γ in the subsequent growth and spread of macrophages.

    Notes: Co/Pt multilayer disks with a double-layer and a quadri-layer structure were prepared on a 2 P (photopolimerized) glass substrate with 1-μm-pitched grooves. The read/write characteristics of the disks were studied using Ar gas laser at a wavelength of 488 nm. It was confirmed that very small domains 0.3 μm in length were stably recorded. The carrier-to-noise ratio was 29 and 32 dB (30 kHz BW) at a mark length of 0.3 μm for double-layer and quadri-layer disks, respectively, being larger than that measured for a TbFeCo disk because of a higher figure of merit. In higher-density recording below 0.5-μm mark length, a quadri-layer disk shows a larger recording power margin than a double-layer disk due to the smaller expansion of domain with power, originating from the sharper thermal distribution.

    Notes: Bronze-processed multifilamentary Nb3Sn superconducting composite wires were loaded and then unloaded at room temperature, resulting in a large change in the critical current (Ic) and the upper critical magnetic field (Hc2) at 4.2 K: the Ic and Hc2 increased, reaching maximum and then decreasing with an increasing applied stress level. The changes in Ic and Hc2 below the loading stress to cause breakage of the Nb3Sn were described well from the viewpoint of the change in residual strain in Nb3Sn. When the applied stress was high enough to cause breakage of the Nb3Sn, the Ic was reduced seriously. From the reduction in Ic in such a case, the strength distribution of Nb3Sn was estimated by using the Weibull distribution function. The result indicated that the shape parameter for the Weibull distribution was 12 and the average strength was 1.02 GPa in the present samples. From the calculation of the residual strain at 4.2 K in combination with the estimated strength distribution of Nb3Sn, the change in Ic at 4.2 K as a function of applied stress at room temperature could be described well.

    Notes: Thin films have been deposited by XeCl excimer laser ablation of a bulk yttria-stabilized zirconia (YSZ) target. The deposited thin films were characterized in terms of spatial distribution, composition, microstructure, and solid particulates on the film surface through deposition process control. The deposition of dense YSZ films, 0.5–2 μm thick, with an excellent adhesive property at 800 °C was performed on the CeO2-Sm2O3 substrate maintained at 500 °C under an O2 ambient of 5×10−2 Torr. The crystallinity of YSZ thin films was found to be promoted by post-deposition annealing. The YSZ film consisted of a cubic phase with predominant growth along the (200) plane.

    Notes: Co/Pt and Co/Pd multilayers were prepared by two source dc-magnetron sputtering. It was found that magneto-optical and magnetic properties of the multilayers were affected by total film thickness. Kerr rotation angle of the films was remarkably enhanced at the film thickness below several hundred angstroms. The theoretical calculation clarified that the increase of Kerr rotation was due to optical interference and multiple reflection. The shape of Kerr loop also depends upon the film thickness and a perfect squareness is attained only at the ultrathin region. The improvement in the squareness of Kerr loop can be explained by the change in the domain structure from a stripe domain to a single domain. These behaviors at the ultrathin region in the multilayers are suitable for new magneto-optical recording media.

    Notes: The presence of a high-frequency ((approximately-greater-than)6 MHz) oscillation in the discharge current and voltage of a plasma-focus device is found experimentally and is explained by using equivalent circuit analyses. The occurrence of such an oscillation is attributed to the excitation of the resonant oscillation in the discharge circuit. It is also shown that the high-frequency response of the driver circuit, which consists of a capacitor bank and cables, plays an important role in determining the high-frequency behavior of the discharge current and voltage.

    Notes: The ciliated protozoan Paramecium contains hemoglobin in heterogeneous monomeric forms. In particular, Paramecium caudatum is characterized by the presence of a major component called Hb10 and a basic component named bHb. We found that in P. caudatum both of these hemoglobin components show some variation according to stock. The types and distributions of these hemoglobin components were examined on 16 stocks in five different syngens and one stock in an unidentified syngen using high performance liquid chromatography. The results indicate that in a variety of stocks the major component, Hb10, was divided into three types, A, B or A + B, and that the basic hemoglobin component was composed of a combination of two or three variants out of four possible, i.e. bHb 1, bHb 2, bHb 3 and bHb 4. Neither the Hb10 types nor the bHb variants, however, could be used to distinguish syngen in P. caudatum, since all of the Hb10 types and bHb variants were widely distributed over syngens and identical profiles appeared to some stocks in different syngens.

    Notes: SYNOPSIS. The helical coiling state of a glycerinated stalk in the family Vorticellidae had been regarded, without any evidence, as a reversible and completely equilibrated mechano-chemical system regulated by the external free Ca2+ concentration. Our present detailed observations of many individual stalks of glycerinated Vorticella convallaria revealed that the contraction-extension cycle of the helical stalks caused by increasing and decreasing free Ca2+ concentrations in the medium represents a kind of hysteresis. This phenomenon was observed in extensively glycerinated Vorticella in a medium of very low or physiological ionic strength, as well as in briefly glycerinated ciliates. With regard to the configurational change in various parts of a glycerinated stalk during the contraction-extension cycle, it was found that the initial bending of the stalk caused by increasing free Ca2+ concentration begins to take place near the zooid (head) while the initial reextension of the stalk with decreasing free Ca2+ concentration begins to take place near the zooid (head) while the initial reextension of the stalk with decreasing free Ca2+ concentration takes place simultaneously throughout the entire stalk. The Ca2+ threshold for complete reextension of the part of the stalk near the rootlet was found to be much higher than that near the zooid.

    Notes: The microstructural factors limiting the critical current density Jc have been investigated for the silver-sheathed Bi2223 tapes prepared by the powder-in-tube technique. In a hierarchy among them, which factor is the most effective in determining Jc depends on how extensively each of them occurs. The Jc was found to increase when the volume fraction of 2223 phase increases, suggesting the importance of the connectivity among 2223 grains. The Jc also increased with decreasing amount of residual carbon. The temperature and magnetic field dependences of Jc indicated characteristics of Josephson currents through superconducting–normal–superconducting barriers. After a discussion based on the alternative current limiting mechanisms proposed up to now, it is suggested that the homophase grain boundary limits the Jc. © 1996 American Institute of Physics.
