    Notes: The mechanical properties, tensile strength, and elongation were investigated for poly(vinyl chloride) (PVC) samples mixed with dioctylphthalate (DOP) at concentrations from 0 to 100 parts per hundred parts PVC at 23°C. It was found that the tensile strength decreased with the increase of concentration, and the elongation was increased until a concentration of 30 DOP content, and then decreased. This leads to the suggestion that intermolecular plasticization is dominant until 30 DOP content, while interstructural plasticization is prevailing for higher concentrations. The permittivity ε′ and the dielectric loss factor ε″ of the same samples have been measured in the frequency range 102-105 Hz at temperatures from 3 to 96°C. Results show that as the DOP content increases in PVC, the dielectric absorption becomes broader, and the glass transition temperature Tg is lowered. The magnitude of the loss peak decreases with an increase of DOP content to a minimum at concentrations from 40 to 60 DOP content. At higher concentrations the loss peak is increased and Tg is unaltered. Another absorption was observed at 100 Hz and at high temperatures, which was attributed to Maxwell-Wagner effect or direct current conductivity or both of them. It was found that the sample containing 40 parts DOP in 100 parts PVC possesses the best mechanical and electrical properties.

    Notes: The aim of the present study was to investigate the expression of the mammary derived growth inhibitor (MDGI) and the subcellular localization of MDGI related antigens in bovine mammary glands. Cell-free translation of poly (A+) = RNA, immunoprecipitation with rabbit anti-MDGI-antibodies, and estimation of the relative contents of MDGI by a radioimmunoassay in mammary tissue of different functional statess revealed that the 13 kDa MDGI was dramatically increased in terminally differentiated mammary tissue compared with the proliferating tissue from pregnant animals. To address the question of tissue localizationl, polyclonal anti-MDGI antibodies and antibodies directed aganist a sythetic peptide corresponding ot residues 69 to 78 of MDGI were used. Western blotting of tissue fractions revealed the cytosolic and microsomal localization of MDGI. Additionally, both types of antibodies a 70-kDa antigeninthe unclear fractionof differentiated mammary glands. Salt extraction and DNase I digestion of isolated unclei, as well as chromatin purification, indicated an association of the 70-kDa antigen with the chromatin. By means of the immunogold technique, MDGI-related antigens were localized within euchromatic unclear regions of epithelial cells in the intact differentiated mammary gland. The immunostaining was markedly diminished in the proliferating tissue. This finding raises the possibility that MDGI and the 70-kDa antigen influence cell proliferation by acting on geneexpression within the unclei of mammary glands.

    Notes: Dipeptides of the aromatic fluorescent amino acid, pyrenylalanine, are studied using both stationary and transient fluorescence techniques. Since the conformational transitions of the peptide chain are slow compared to the decay of the pyrene excited state, both ground state conformations, adopted by the peptide, i.e., C5 and C7, can be monitored separately. Kinetic models are proposed to describe the molecular dynamics of the peptide chain as probed by the intramolecular excimer formation between both pyrene chromophores. These kinetic schemes explain the influence of solvent, chain chirality, main chain-side chain interactions, and nature of the protecting groups on the emission spectrum and the fluorescence decay profile of these model peptides. These schemes also provide a tool to calculate rate constants of conformational transitions and excimer formation. By comparing the kinetic and thermodynamic parameters of the various compounds, the influence of a structural modification on the molecular dynamics of the peptide chain is determined.

    Notes: The aqueous pseudoheterogeneous polymerization of methyl methacrylate in the absence and presence of some metal chlorides or sulfates was carried out in nitrogen atmosphere at 40°C using sodium bisulfite as initiator. The metal chlorides or sulfates were found to catalyze the rate of polymerization of methyl methacrylate to different extents. The highest effect was obtained in the presence of copper salts, a moderate effect was obtained when iron salts were used, and the least effect was caused by the others. All 13C-NMR spectra for the polymers obtained in the absence and presence of some metal salts were those to be expected for radically initiated poly(methyl methacrylate)s of conventional, slightly syndiotactic-rich microstructures prepared at normal temperature. The spectra were found to be very similar to that for a radical poly(methyl methacrylate) first analyzed by Inoue et al. The ratios of isotactic (mm): heterotactic (mr,rm): syndiotactic (rr) triads were found to be close to 6 : 32 : 62. The polydispersity for some poly(methyl methacrylate)s determined by gel permeation chromatography technique was found here to vary rather widely and ranges between 1.8 and 8.92.

    Notes: A series of electrically conducting pyrrole “black” polymers has been prepared by chemical oxidation of pyrrole with a variety of ferric salts. The synthesis and properties of these materials are described and compared with electrochemically-prepared analogs. Pyrrole “blacks” with strong acid anion “dopants” exhibited the highest conductivities, up to 62 Ω cm-1 for the triflate-doped polymer. The adjustability and ease of synthesis of the ferric-derived polypyrroles render these particularly promising for large-scale processing of conducting polymers.

    Notes: To study the molecular aspects of the regulation of transcription of a multigene family, we have isolated and sequenced cDNA and genomic clones coding for the α-tubulin of the sea urchin Paracentrotus lividus. Two cDNA clones, Pα 10 and Pα 4, contain respectively the coding information for 391 C-terminal and for 338 N-terminal amino acids of the 452 residues that constitute the complete protein. They show silent nucleotide substitutions only, suggesting that Pα 10 and Pα 4 represent the cloned copies of two allelic gene transcripts, which encode for two α-tubulin isoforms with identical amino acid sequence in the region of the overlap. The comparison of the predicted amino acid sequence of the composite Pα 4-10 and of the mouse M α-6 (Villasante et al., Mol. Cell Biol 1986; 6:2409-2419) reveals a conservation of 97% between the two polypeptides. By RNA blotting hybridization six major α-tubulin transcripts were identified. Two, of 3.5 kb and 2.0 kb, are expressed in the unfertilized eggs and during early cleavage. The other two maternal mRNAs, of 2.4 kb and 1.8 kb, are expressed in both early and late cleavage embryos, but in the intestine the 1.8 kb RNA, which specifically reacted with the 3′ specific probe of the Pα 10 cDNA, is the only transcript detected. Finally, the 1.5 kb and 1.9 kb mRNAs represent the transcription of stage- and tissue-specific genes, respectively. In fact, the former becomes detectable at blastula stage and accumulates during late development, whereas the latter is found in the testis only. The sequence data of the 3′ terminus of the α-3 genomic clone suggests that it encodes for a divergent α-tubulin, and it most probably corresponds to the testis-specific gene.
