    Description: Amyloid deposition in senile plaques and the cerebral vasculature is a marker of Alzheimer's disease. Whether amyloid itself contributes to the neurodegenerative process or is simply a by-product of that process is unknown. Pheochromocytoma (PC12) and fibroblast (NIH 3T3) cell lines were transfected with portions of the gene for the human amyloid precursor protein. Stable PC12 cell transfectants expressing a specific amyloid-containing fragment of the precursor protein gradually degenerated when induced to differentiate into neuronal cells with nerve growth factor. Conditioned medium from these cells was toxic to neurons in primary hippocampal cultures, and the toxic agent could be removed by immunoabsorption with an antibody directed against the amyloid polypeptide.     Description: Smell identification ability was measured in 1955 persons ranging in age from 5 to 99 years. On the average, women outperformed men at all ages, and nonsmokers outperformed smokers. Peak performance occurred in the third through fifth decades and declined markedly after the seventh. More than half of those 65 to 80 years old evidenced major olfactory impairment. After 80 years, more than three-quarters evidenced major impairment.     Description: The neu oncogene, identified in ethylnitrosourea-induced rat neuroglioblastomas, had strong homology with the erbB gene that encodes the epidermal growth factor receptor. This homology was limited to the region of erbB encoding the tyrosine kinase domain.     Notes: A method for the quantitative determination of chlorpromazine and five of its major metabolites in a single sample of biological fluid in the ng/ml range has been developed utilizing gas chromatography/mass spectrometry with selected ion recording. The assay is highly specific and quantification is accomplished by an inverse stable isotope dilution technique, using deuterium-labeled variants of the compounds as internal standards. In this way the concentrations of chlorpromazine and five of its major metabolites (the sulfoxide, the N-oxide, the monodemethylated, the didemethylated, and the 7-hydroxylated compounds) can be determined in biological fluids. Levels in humans have been measured both in plasma and in red blood cells and are compared to those found in related in vitro studies.

    Notes: Doubly charged ion mass spectra have been obtained for 15 n-alkane hydrocarbons. Spectra were measured using a Nier-Johnson geometry Hitachi RMU-7L mass spectrometer operated at 1.6kV accelerating voltage. Fragment ions, which resulted from C—C bond rupture and extensive H loss, dominated the spectra. Molecular ions have not been observed. The most intense ions in the doubly charged ion mass spectra of n-alkanes were [C2H4]2+, [C3H2]2+, [C4H3]2+, [C5H2]2+, [C6H6]2+, [C6H8]2+, [C7H6]2+, [C7H8]2+, [C8H6]2+ and [C8H8]2+. Appearance energies for forming the prominent doubly charged fragment ions have been measured and range from 27.5 eV to energies greater than 60eV. A geometry optimized SCF approach has been used to compute the energies and structures of prominent ions in the doubly charged mass spectra.

    Notes: The liquid SIMS mass spectra of silver acetate dissolved in a glycerol matrix is discussed, with emphasis on the formation of a ‘silver mirror’ on the surface of the glycerol droplet owing to reduction of the silver acetate. Silver clusters containing up to three silver atoms have been observed from this mirrored surface; Ag3+ cluster ions are not observed in the spectrum when conditions are such that the mirror is not formed. For example, use of a slightly oxidizing matrix (o-nitrophenyl octyl ether or m-nitrobenzylalcohol) prevents formation of the ‘mirror’; only Ag+ is sputtered from this surface.

    Notes: The chemical shifts of N-benzylpyridinium salts have been measured in D2O, and the substituent induced chemical shifts of the methylene protons are in substantial agreement with those expected from the overall electronic effects of the substituents. An additivity effect of the substituents on the methylene resonance is also observed.
