Zusammenfassung Es werden feinstrukturelle Veränderungen anLeishmania donovani im Lebergewebe vom Goldhamster vor und nach Tiefgefrierung in flüssigem Stickstoff beschrieben. Als Gefrierschutzmittel wurde DMSO in einer Endkonzentration von 5% benutzt. Um festzustellen, ob morphologische Veränderungen an dem Parasiten aufgrund des Tiefgefrierprozesses oder vorbereitenden Arbeiten resultierten, wurden vor der Tiefgefrierkonservierung Kontrollen untersucht. Die meisten der unbehandelten und 20 min nach Entnahme der Leber fixierten Parasiten wiesen eine normale Struktur des Cytoplasmas und Kernes auf. Nach 90 min Aufbewahrung in PSG-3-Puffer mit und ohne DMSO-Zusatz (5%) bei 4°C fanden sich jedoch Veränderungen, wie Erweiterung des Mitochondrium und der Geißeltasche, Aufblähung der parasitophoren Vakuole, Verlust des an Ribosomen reichen Cytoplasmas, teilweise Schädigung der mitochondrialen Matrix, Kondensation der lamellären Struktur des Kinetoplasten und Aufhellung des Euchromatins. Nach Tiefgefrierung wurden nur wenige Parasiten mit normaler Struktur des Cytoplasmas und Kernes gesehen. In der Regel hatten wiedergewonnene Leishmanien ähnliche, aber auffälligere Veränderungen des Cytoplasmas als Parasiten vor der Tiefgefrierung. Zellen mit fortgeschrittener Desintegration wiesen jedoch charakteristische durch Tiefgefrierung verursachte Alterationen, wie Spaltbildung, im Cytoplasma, Zerstörung der Parasiten-Membran, Verlust der pelliculären Mikrotubuli und eine stufenweise fortschreitende Schädigung der Kerne. Mäßig geschädigte, Kerne hatten Chromatin, das deutlich denaturiert war und eine augenfällige Trennung von Euchromatin und Heterochromatin zeigte, wohingegen schwergeschädigte Kerne Auflösung und manchmal Ruptur der Kernhülle aufwiesen. Reste des Karyoplasmas sahen dann aus wie grobschollige oder kondensierte Plaques. Die endgültige Zerstörung einer Zelle war durch eine fast völlige Elution des Cytoplasmas und Kondensation des Karyoplasmas gekennzeichnet.

Abstract The ultrastructural changes inLeishmania donovani in parasitized liver tissue of the golden hamster prior to and after storage in liquid nitrogen are described. Dimethysulphoxide (DMSO) at a final concentration of 5% was used as the cryoprotective agent. To ascertain whether morphological changes in parasites resulted from deep-freezing or preparative procedures, controls prior to cryopreservation were investigated. The majority of untreated parasite cells fixed 20 min after removing the liver had normal cytoplasmic and nuclear structure. However, suspending the respective samples for 90 min at 4°C in PSG-3-buffer with and without the addition of DMSO (5%) caused changes in the parasites, such as enlargement of the mitochondrion and flagellar pocket, inflation of the parasitophorous vacuole, loss of cytoplasm rich in ribosomes, partial damage to the mitochondrial matrix, condensation of the lamellar structure of the kinetoplast as well as translucency of euchromatin of the nucleus. After cryopreservation only a few parasite cells with normal structure of the cytoplasm and the nucleus were noted. As a rule, recovered leishmaniae had similar but more conspicuous alterations of the cytoplasm than parasites prior to deep-freezing. Thus all transition stages of damaged parasites were observed. However, cells with advanced disintegration of cytoplasm exhibited characteristic changes caused by deepfreezing or thawing, such as fissuration of the cytoplasm, destruction of the unit plasma membrane, loss of the pellicular microtubules, or gradual damage to nuclei. Moderately damaged nuclei had chromatin which was markedly denatured, showing an obvious separation of euchromatin and heterochromatin, whereas severely damaged nuclei revealed disintegration and sometimes rupture of the nuclear envelope. Remnants of the karyoplasm then appeared like coarse or condensed plaques. The final destruction of the cell was marked by almost total elution of cytoplasm and condensation of karyoplasm.
