    Notes: The facilitated transport of CO2 through ion exchange membranes containing organic amine counterions was studied. Steady state CO2 fluxes were measured from pure gas streams and mixtures with CH4. Facilitation factors for CO2 up to 26.7 were calculated from transport data. Transport measurements were binary mixtures of CO2 and CH4 yielded ratios of CO2 flux to CH4 flux ranging from 29.0 to 264. Conversion of flux ratios to permeabilities yielded separation factors up to 551 for CO2 over CH4, which are compared to literature data for polymeric membranes. The transport data support a mobile, reactive-carrier facilitated transport mechanism with a carbamate zwitterion complex. Effective diffusivities for CO2 and the carrier-gas complex were determined from transport data. A reaction equilibrium model predicted facilitation factors that were in very good agreement with experimental values. Potential applications are also discussed.

    Notes: Human polymorphonuclear leukocyte elastase (PMN elastase) is inhibited by L-659, 286 (7α-methoxy-8-oxo-3-[[(1,2,5,6-tetrahydro-2-methyl-5,6-dioxo-1,2,4-triaz-in-3-yl)thio]methyl]-5-thia-1-aza-6R-bicyclo [4.2.O]oct-2-ene-2-pyrrolidine carboxamide-5,-dioxide) with a Ki of 0.4 μM. This inhibition is time-dependent, rapid, and only slowly reversible, with a t1/2 of 〉 3 days at 25°C. L-659, 286 is also highly selective for PMN elastase, as it does not inhibit thrombin, trypsin, papain, plasmin, chymotrypsin, or cathepsin G. L-659, 286 administered intratracheally inhibits lung damage caused by administration via the same route of human PMN elastase into hamsters. In marmosets, L-659, 286 is cleared from blood very rapidly after an intravenous injection but is recovered in bronchoalveolar lavage fluid for several hours after intratracheal administration.

    Notes: Laminar mixing in a single screw extruder between two miscible liquids is considered. Studies are presented which examine the mixing performance in single screw extruders with mixing sections, variable geometry screws, and multi-viscosity mixtures. Model extruders, set up on engine lathes, were used to mix colored silicone rubber which eventually cured in the screw channel. The cured silicone was then removed and sliced to measure the mixing. Results show mixing as a function of both down channel location and total average strain. For the mixing section study, results support the theory of interfacial area reorientation as the key mechanism in mixing section performance. Increasing viscosity ratio for the multi-viscosity study showed a decrease in mixing rate.

    Notes: Intermediate subviral particles (ISVP) derived from reovirus represent a simple model system for the switch-on of transcriptase function. In such particles the endogenous transcriptase is present in a switched-off form, one step removed from the switched-on state. Switch-on of transcriptase function is an active process in this system and can be triggered by K+ ions. A variety of agents which affect gene expression in cells were tested for an effect on switch-on in ISVP. Marked effects on switch-on in ISVP were observed with a diverse group of test agents, including DMSO and other solvents, BUdR, TdR, caffeine, theophylline, and temperature. The correlation in response between ISVP and cells suggests that the ISVP system may be useful as a model for studying the biochemical mechanisms underlying the perturbative effects of such agents on gene expression in cells.

    Notes: The effects of the male antifertility agent, α-chlorohydrin, six of its derivatives, and glycidol were studied on the metabolism of washed ram spermatozoa in vitro with fructose as substrate. The α-chlorohydrin derivatives were the amino, the phosphorylated, and four glycol-bridge (ketal) compounds. All compounds except glycidol, in a concentration between 0.1 and 100 mM, reduced the aerobic glycolsis and/or oxidation of fructose. However, there was not a high correlation between the ability of these compounds to inhibit the metabolism of ram spermatozoa in vitro and their antifertility activity when administered to male rats. Other factors are clearly involved in their antifertility activity, eg, the concentration of the compounds in the epididymis and their conversion of either more or less spermicidal compounds in the body.

    Notes: The morphological and ultrastructural changes that occur during preparation of porcine, bovine, and murine spermatozoa for flow cytometric quantification of the relative DNA content of the X- and Y-chromosome-bearing sperm populations were examined. Ejaculated spermatozoa from the boar and bull were washed using a series of dimethyl sulfoxide (DMSO) solutions prior to fixation, whereas the epididymal mouse spermatozoa were washed only in phosphate-buffered saline (PBS). Spermatozoa from all three species were then fixed in ethanol and processed for fluorochrome staining by a treatment regimen consisting of sulfhydryl reduction and proteolysis. The processed sperm nuclei were stained for DNA with the fluorochrome, 4′-6-diamidino-2-phenylindole (DAPI) before quantification by flow cytometry. Scanning and transmission electron micrographs of sperm heads taken at various steps of the preparation and staining procedures show 1) that the rigorous washing procedure disrupted the plasma and outer acrosomal membranes, 2) that ethanol fixation resulted in removal of the outer membranes and disintegration of the nuclear envelope, and 3) that thiol and proteolysis treatment removed the remaining cellular organelles including the tail and rapidly induced partial decondensation of the tightly packed chromatin. Sequential micrographs showed that the nuclear matrix of all three species increased in thickness about twofold during the preparation and staining. Consequently, the harsh procedures currently used for quantitative staining of DNA for high-resolution flow cytometric analyses destroy most cellular organelles and thereby prevent simultaneous characterization of DNA content and other sperm cell constituents.

    Description: ISEE-3 Geotail observations are used to investigate the relationship between the interplanetary magnetic field, substorm activity, and the distant magnetotail. Magnetic field and plasma observations are used to present evidence for the existence of a quasi-permanent, curved reconnection neutral line in the distant tail. The distance to the neutral line varies from absolute value of X = 120 to 140 R/sub e near the center of the tail to beyond absolute value of X = 200 R/sub e at the flanks. Downstream of the neutral line the plasma sheet magnetic field is shown to be negative and directly proportional to negative B/sub z in the solar wind as observed by IMP-8. V/sub x in the distant plasma sheet is also found to be proportional to IMF B/sub z with southward IMF producing the highest anti-solar flow velocities. A global dayside reconnection efficiency of 20 + or - 5 percent is derived from the ISEE-3/IMP-8 magnetic field comparisons. Substorm activity, as measured by the AL index, produces enhanced negative B/sub z and tailward V/sub x in the distant plasma sheet in agreement with the basic predictions of the reconnection-based models of substorms. The rate of magnetic flux transfer out of the tail as a function of AL is found to be consistent with previous near-earth studies. Similarly, the mass and energy fluxes carried by plasma sheet flow down the tail are consistent with theoretical mass and energy budgets for an open magnetosphere. In summary, the ISEE-3 Geotail observations appear to provide good support for reconnection models of solar wind-magnetosphere coupling and substorm energy rates.

    Description: A decrease in the solar radius is determined using the technique of Dunham and Dunham (1973), in which timed observations are made just inside the path edges. When the method is applied to the solar eclipses of 1715, 1976, and 1979, the solar radius for 1715 is 0.34 + or - 0.2 arc second larger than the recent values, with no significant change between 1976 and 1979. The duration of totality is examined as a function of distance from the edges of the path. Corrections to the radius of the sun derived from observations of the 1976 and 1979 eclipses by the International Occultation Timing Association are also presented.

    Description: Absorption features from the 8 micron SiO fundamental (upsilon = 1-0) and hot bands (upsilon = 2-1) have been observed in sunspots at sub-Doppler resolution using a ground-based tunable diode laser heterodyne spectrometer. The observed line widths suggest an upper limit of 0.5 km/s for the microturbulent velocity in sunspot umbrae. Since the silicon monoxide abundance is very sensitive to sunspot temperature, the measured equivalent widths permit an unambiguous determination of the temperature-pressure relation in the upper layers of the umbral atmosphere. In the region of SiO line formation (log P sub g = 3.0-4.5), the results support the sunspot model suggested by Stellmacher and Wiehr (1970).
