    Notes: Abstract Rosmarinic acid synthase from cell cultures of Coleus blumei Benth. was purified to apparent homogeneity by fractionated ammonium sulfate precipitation (60–80% saturation), hydrophobic interaction chromatography, affinity chromatography and gel filtration. This purification procedure resulted in a 225-fold-enriched specific enzyme activity with a yield of 9%. The protein preparation was apparently pure according to sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and two-dimensional gel electrophoresis. The apparent molecular mass determined by gel filtration and SDS-PAGE was 77 kDa, indicating that rosmarinic acid synthase is a monomeric enzyme.

    Notes: Abstract A biosynthetic pathway for rosmarinic acid is proposed. This pathway is deduced from studies of the enzymes detectable in preparations from suspension cells of Coleus blumei. Phenylalanine is transformed to 4-coumaroyl-CoA by the enzymes of the general phenylpropanoid pathway: phenylalanine ammonia-lyase (EC 4.3.1.5), cinnamic acid 4-hydroxylase (EC 1.14.13.11) and hydroxycinnamic acid:CoA ligase (EC 6.2.1.12). Tyrosine is metabolized to 4-hydroxyphenyllactate by tyrosine aminotransferase (EC 2.6.1.5) and hydroxyphenylpyruvate reductase. The ester can be formed from 4-coumaroyl-CoA and 4-hydroxyphenyllactate by the catalytic activity of rosmarinic acid synthase with concomitant release of CoA. Microsomal hydroxylase activities introduce the hydroxyl groups at positions 3 and 3′ of the aromatic rings of the ester 4-coumaroyl-4′-hydroxyphenyllactate giving rise to rosmarinic acid.

    Notes: Xenon-129 NMR is used to probe macroscopic distributions of aromatic molecules adsorbed in a packed bed of 1-μm NaY zeolite particles. Relative rates of guest transport through the intracrystalline (micro) and intercrystalline (macro) pores play a unique role in the axial distribution of sorbate molecules, such as hexamethylbenzene, in a zeolite powder. Xenon-129 NMR spectra show that a sharp HMB adsorption front advances through a bed of dehydrated NaY crystallites at 523 K. However, at 573 K or in the presence of coadsorbed water, HMB species disperse through the bed without forming a sharp boundary between adsorption zones.When guest transport is controlled by pseudosteady-state diffusion in the macropores, axial penetration of the bed by vapor-phase guest species occurs in a sharp adsorption front. A shrinking-core transport model then quantitatively estimates the intracrystalline diffusivities of HMB in dehydrated and partially hydrated NaY zeolite of 10-11 and 10-13 m2/s, respectively, at 523 K. Xenon-129 NMR proves to be a powerful tool for probing adsorbed guest distribution in zeolites, allowing relative time scales to be established for transport of molecular guests in NaY powders.

    Notes: The mixing and segregation behavior of liquid fluidized beds containing two different-sized particle species are described mathematically. This model is based on a mass balance, rather than a volume balance, which has been used by most previous researchers. The model also includes the effects of density gradients that are present in the bed. Particle dispersion coefficients in a binary-solid fluidized bed are evaluated using this model. Further, a correlation that allows the calculation of these particle dispersion coefficients is presented, which describes not only dispersion coefficients calculated using the above model, but also those presented by other researchers. It is applicable over a broad range of variables of practical interest and is based on relevant dimensionless parameters to account for the influence of important particle and fluid properties on the particle dispersion characteristics.

    Notes: Adsorption of a strongly bound absorbate in a bidisperse-pore system is modeled using a Turner pore structure. This idealized system leads to an analytical solution for the micropore uptake of adsorption that is valid for a part of the adsorption-time profile. The analytical solution yields a well-defined criterion that establishes the boundary between an adsorption process dominated by macropore resistance and one dominated by micropore resistance. The range of the analytical solution is extended to include the entire adsorption-time profile using an approximate analytical solution and two asymptotic solutions.

    Notes: This paper discusses the procedure developed for simulating water sprays at a reduced scale (i.e., physical modeling) and, more impotantly, compares the model spray characterstics to corresponding full scale spray characteristics. An ideal physically modeled spray would simulate actual spray momentum, water volume flow rate, air entrainment rate, discharge water pressure, spray angle, spray pattern and drop size distribution. Full scale measurements or estimates of air discharge velocity, momentum, entrainment rate and drop size distributions were obtained for two types of sprays at two discharge pressures, various spray angles and at a 500 gpm flow rate. Physical modeling was conducted to simulate the full scale nozzles at a 1:50 scale reduction using commercially available spray nozzles. Measurements of spray momentum, volume flow rate and air entrainment rate were obtined for a subset of spray angles used in the full scale testing. Particle size distribution was not measured since it was considered of secondary importance in the simulation. The results of the study showed that model and full scale momentum, volume flow rate and air entrainment rates compared well with corresponding full scale observations (i.e., within 10 percent for momentum, 20 percent for volume flow rate and 25 percent for air entrainment rate). The results indicate that the method used for simulating water sprays is valid and that the model sprays adequately simulate the mechanical action of full scale sprays on vapor clouds. The results provide evidence that physical modeling can be used to evaluate the effectiveness of water sprays as a mitigation technique.
