    Notizen: Abstract The basis for the hypersensitive response of glycogen phosphorylase to epinephrine stimulation was investigated in adult rat cardiomyocytes isolated from normal and alloxan-diabetic animals. To assess potential G-protein involvement in the response, normal and diabetic derived myocytes were incubated with either cholera or pertussis toxin prior to hormonal stimulation. Pretreatment of cardiomyocytes with cholera toxin resulted in a potentiated response to epinephrine stimulation whereas pertussis toxin did not affect the activation of this signaling pathway. To determine if the enhanced response of phosphorylase activation resulted from an alteration in adenylate cyclase activation, the cells were challenged with forskolin. After 3 hr in primary culture, diabetic cardiomyocytes exhibited a hypersensitive response to forskolin stimulation relative to normal cells. However, after 24 hr in culture, both normal and diabetic myocytes responded identically to forskolin challenge. The present data suggest that a cholera toxin sensitive G-protein mediates the hypersensitive response of glycogen phosphorylase to catecholamine stimulation in diabetic cardiomyocytes and this response which is present in alloxan-diabetic cells and is induced in vitro in normal cardiomyocytes is primarily due to a defect at a post-receptor site.

    Notizen: Abstract Freshly isolated ventricular myocytes have been used extensively as an adult cardiac model system. Due to their inability to undergo cytokinesisin vitro and their dedifferentiated properties in long-term culture, they can not be used for extended studies. Recent reports tell of the establishment of fetal and neonatal cardiac cell lines and the development of adult cardiomyocytes from transgenic animals. A recent report by Kirshenbaum [1], is the first to demonstrate insertion of genes in to adult ventricular myocytes using viral infection. This paper discusses the infection of primary adult differentiated cardiomyocytes with the SV40 large T antigen and subsequent proliferation under temperature sensitive control. Upon further characterization, the cells could be used as a model to study muscle differentiation and repair as well as adult cardiac cell physiology.

    Notizen: Summary Protein phosphatase 2A1 was purified from rat skeletal muscle and used to produce antisera to the three subunits of the holoenzyme. Affinity purified antibodies specific for the subunits of the phosphatase enzyme were found to recognize the type 2A1 and 2A2 phosphatase from rat skeletal muscle, heart, liver, brain and erythrocytes and were used to investigate the effects of diabetes on the levels of this enzyme in liver and heart. Phosphorylase phosphatase assays coupled with immunoblot analysis of fractionated rat liver and heart cytosol from normal and diabetic animals show no apparent differences in the quantity or activity of these enzymes following the induction of alloxan diabetes. When considering these results and the normal physiological concentrations of known effectors of these enzymes, it is likely that protein phosphatase 2A1 and 2A2 are not responsible for the dephosphorylation of phosphorylase a under physiological conditions.

    Notizen: Abstract Carbonic anhydrase IdMichigan, an electrophoretic variant of human red cell carbonic anhydrase I, was purified from erythrocytes obtained from an individual heterozygous for the trait. Primary structural analysis indicates that a lysine residue has exchanged for a threonine residue in the variant enzyme. After isolation, there was approximately 1.8 times as much normal as variant enzyme. Thermostability studies demonstrated that carbonic anhydrase Id was more thermolabile than the normal enzyme. The normal and variant enzymes showed no differences in specific carboxylesterase activity or CO2 hydratase activity. Utilizing the carboxylesterase activity toward β-naphthyl acetate, the normal and variant enzymes had similar Michaelis constants, pH profiles, and rates of inhibition by acetazolamide. Immunochemical studies did not demonstrate an antigenic difference for the variant enzyme.

    Notizen: Abstract The phosphoglucomutase (PGM) electrophoretic phenotype of the mouse (Mus musculus) consists of several distinct components which can be grouped into two major zones designated PGM-1 and PGM-2. Evidence presented here indicates that each zone is controlled by a single genetic locus denoted Pgm-1 and Pgm-2, respectively. Two variant forms segregated at the Pgm-1 locus. They were codominantly expressed and inherited as alleles at an autosomal locus. The alleles were termed Pgm-1 a (fast) and Pgm-1 b (slow). These alleles were separately fixed in a number of inbred strains of mice. Preliminary evidence based on wild mouse phenotypes indicates that variant forms also exist for PGM-2 which are inherited as alleles at an autosomal locus. Genetic linkage relationships have not been determined for these loci. PGM-1 variants and PGM-2 were expressed in mouse fibroblasts in vitro.

    Notizen: Abstract Cluster Analysis has been shown to be partially suitable for the classification of glasses. The expansion of the clusters on the Seger's diagram allows an investigation of the influence of the different glass components. Variation of the SiO2 content results in a smaller change of properties than variation of the Na2O/RO relationship. Principal Component Analysis is convenient for showing correlations between the composition of the examined glasses, the glass structure parameters, and the different electrode properties.

    Notizen: Zusammenfassung Kurz zusammengefasst sind die wichtigsten, hier niedergelegten Tatsachen folgende: Es wurde eine Methode für die totale Entfernung der Leber bei Hunden ausgebildet, welche frei von den meisten Fehlerquellen ist, die früheren Exstirpationsmethoden des Organs bei Säugetieren anhaften. Es wurde gefunden, dass eine geeignete Einführung von Glukose bei derart operierten Hunden das Leben viele Stunden nach Verlust des gesamten Lebergewebes erhält. Von den vielen an hepatektomierten Tieren gemachten Beobachtungen möchten wir hervorheben, 1. dass die Leber zur Erhaltung des Blutzuckerspiegels absolut wesentlich ist; 2. dass die Leber der wichtigste Faktor, wenn nicht der einzige, bei dem Prozess der Desamidierung und der Harnstoffbildung ist, und 3. dass Bilirubin auch ohne Mitwirkung der Leber gebildet werden kann.

    Notizen: Abstract Erythrocyte and tissue lactate dehydrogenase (LDH) electrophoretic patterns of 26 rodent species from ten families were examined. The LDH B gene was observed to range in erythrocyte expression from species without detectable B subunits to those which predominantly expressed B subunits. However, the shift in erythrocyte B gene expression was not observed in the tissue LDH electrophoretic patterns between rodent species. Species which did not express erythrocyte B subunits, or only small quantities of B subunits, were restricted to the suborder Myomorpha. In erythrocytes of other rodent species, and most mammals, LDH B subunits are expressed equally or in excess of A subunits. The results suggest either structural differences in the LDH B gene between Myomorph and non-Myomorph rodents or a regulator gene which controls the expression of the B gene in Myomorph erythrocytes. Existing evidence favors the latter hypothesis.
