    Notes: Summary Although only one gene is known to be functional, numerous glyceraldehyde-3-phosphate dehydrogenase (GAPDH) related sequences are scattered throughoutMus musculus andRattus rattus genomes. In this report we show that: (1) GAPDH pseudogenes are repeated to comparable extents, at least 400 copies, in 12 other Muridae species; (2) the complete, or nearly so, sequence of GAPDH messenger RNA is amplified, and a high proportion, if not all of these copies, are intronless; (3) GAPDH pseudogenes are preferentially located in heavily methylated and DNAse I-insensitive regions of chromatin; and (4) the presence of atypical GAPDH-related mRNAs in different cellular contexts raises the possibility that more than one GAPDH gene is transcribed.

    Notes: Abstract Throughout manufacturing history, management has searched for tools to help it set product prices accurately. Traditionally, manufacturing managers have analysed efficiency by cost makeup with the basic elements: materials, labour, overheads. Overhead costs are allocated to units of production based on a theorical correlation with labour costs. With automation, direct labour has come to be only 1 to 20% of the total manufactured costs of products. The CIM infrastructure must incorporate a wide range of management facilities. It is now time to focus on an integrative approach which embraces design, planning, process and product costing. With the direct value approach we describe a concept which gives pertinent information to adapt strategy speedily for a better profitability. In addition an economic weighting coefficient for the analysis of job contribution in the creation of added value, it is possible to control product pricing. In this way, economic information can be an element of the product and process database and thus build up an operational decision support system by means of sharing common information.

    Notes: Abstract On the basis of the GRIM4-S1 satellite-only Earth gravity model, being accomplished in a common effort by DGFI and GRGS, a combination solution, called GRIM4-C1, has been derivcd using 1° × 1° mean gravity anomalies and 1° × 1° Seasat altimeter derived mean geoid undulations. In the meantime improvements could be achieved by incorporating more tracking data (GEOSAT, SPOT2-DORIS) into the solution, resulting in the two new parallel versions, the satellite-only gravity model GRIM4-S2 and the combined solution GRIM4-C2p (preliminary). All GRIM4 Earth gravity models cover the spectral gravitational constituents complete up to degree and order 50. In this report the emphasis is on the discussion of the combined gravity models: combination and estimation techniques, capabilities for application in precise satellite orbit computation and accuracies in long wavelength geoid representation. It is shown that with the new generation of global gravity models general purpose satellite-only models are no longer inferior to combination solutions if applied to satellite orbit restitution.

    Notes: Summary Techniques are described for growth of primary human tumor colony-forming units in agar in glass capillary tubes. Tissue recovered from an invasive procedure is made into a single cell suspension. The cells are washed and placed in tissue culture media and agar in glass capillary tubes. After 10 to 14 d of incubation, colonies of tumor cells are visible in the capillaries. This technique can be used to determine the sensitivity pattern of patients' tumors to conventional and investigational anticancer agents. The technique can also be used to screen for antitumor activity of new chemical entities.

    Notes: Summary We have studied the influence of several parameters (glutaraldehyde concentration, pH and contact time) on the activation of an amine silica with glutaraldehyde. In order to take interactions between parameters into consideration, we have used the Response Surface Methodology which allows us to obtain an equation (not a single estimation). The optimal conditions for glutaraldehyde activation of amine Spherosil beads were as follow: the porous silica was activated for 1 h at 25°C with an 8% (V/V) solution of glutaraldehyde at pH 6.4 these conditions were the same with RNA or CMP used as substrates.

    Notes: Abstract Located in northern Québec, the Lac Shortt Mine was a small gold mine consisting of a thin subvertical orebody which was mined in three main phases. High stress and rockbursting conditions were experienced when ore was extracted in the upper zone between the surface and a depth of 500 metres during the first two phases of mining. Severe rockbursts were experienced in late 1989 near the shaft and in the footwall development following a deepening of the mine shaft to a depth of 830 m and partial development of footwall drift access for the third phase of mining (the mining of the lower zone starting at a depth of 830 m moving upward toward a depth of 500 m). A 16-channel Electrolab MP250 microseismic system, with a Queen's University Full-Waveform “piggy-back” system, was installed underground at the site due to these problems. It was expected that the thinning sill would be subjected to an ever-increasing load as the thickness of the 500 m sill pillar decreased in the face of the mining excavation from below. A monitoring program consisting of the microseismic monitoring system, a range of conventional geomechanics monitoring tools as well as the undertaking of periodic seismic tomography surveys to assess the ongoing state of stress and rock mass condition within the sill was therefore warranted. The anomalously high-magnitude stress field and the brittle rockmass created a situation in which rockmass failure was common and violent. In the creation and thinning of the sill pillar, the location of banded microseismic activity was crucial in tracing rockmass failure and the associated ground control problems. Reliable source-location determination enabled the identification of areas of stress increase. The movement of the rockmass “failure front” could be followed, and was responsible for stope dilution, footwall and orebody development deterioration, and caving. Source-mechanism analyses gave accurate double-couple solutions for approximately forty percent of these events having at least ten recognizable polarities. Results suggested movement along vertical north-south striking or vertical east-west striking features. Underground observation of damaged access points showed that vertical north-south striking joints were experiencing failure. The microseismic activity, which was consistently concentrated close to the southwest and northeast corners of current production stopes, could be explained by a stress field oriented obliquely to the strike of the orebody, as measured prior to shrinkage of the sill pillar byin situ stress measurements and observed borehole overbreaks. The orientations of theP andT axes for the microseismic activity further confirmed that the stress field oriented obliquely to strike. While an increase in compressional-wave velocity of 2.3 percent, corresponding to a measured stress increase of approximately 10 MPa could be measured by repeated tomographic surveys, it was relatively small and only a factor of two or so above the velocity measured uncertainty. The relative insensitivity of thein situ rock mass modulus to the applied stress is believed to be largely due to the rockmass discontinuities being relatively closed prior to stress increase, as substantiated by the small deformations seen by the extensometer and borehole camera. This situation existed because of the very high pre-mining stress level. The experimental demonstration that the rock could not absorb substantially increased load through the mechanism of discontinuity closure or tightening (which would be reflected in the modulus) may be evidence in itself of potentially burst-prone ground, such as encountered at Lac Shortt.

    Notes: Abstract The increasing activity of Vulcano Island (Italy) since 1985 led to the initiation of continuous geochemical monitoring of the lateral soil gas emissions. On the basis both of their relative geochemical characteristics and of local considerations, three gaseous components were selected for monitoring, namely CO2, He and 222Rn. Monitoring has been performed by means of specific analysers. Gases extracted from a water well located at the foot of the active cone were selected for monitoring, on the basis of their geochemical and isotopic characters that indicate their genetic link with central high temperature fumarolic gases emitted at the crater. Very strong variations of gas composition can be observed within one day (from 1 to about 94% for CO2). Some variations display a daily character and can be correlated with that of atmospheric pressure. The three monitored gases are highly correlated, suggesting very high kinetics of gas transfer in the system. Because of these considerable variations of chemical composition, bulk concentrations obviously are not suitable for monitoring at Vulcano. However, the evolution with time of ratios such as 222Rn/CO2 and He/CO2 (the latter being corrected for atmospheric contamination) supplies numerical parameters that the expected to characterize the intensity of the degassing process. A new input of magmatic gases, that would lead to an increase in the 222Rn/CO2 and He/CO2 ratios, should therefore be detected by such a monitoring station.

    Notes: Abstract A rabbit serum which had previously been reported to have an immunological affinity for centrosomes of human cell lines was shown also to be specific for the nucleus. Optical and ultrastructural immunolocalization in HeLa cells showed that this specificity is restricted to the fibrillar centre of nucleoli either in untreated or actinomycin D treated interphase cells. In mitotic cells discrete labelling was observed on chromosomes and shown to correspond, on spread metaphase plates, to the short arms of acrocentric chromosomes, i.e. to the nucleolar organizer regions (NORs). Using independent cell fractionation procedures in the human T-lymphoblastic KE 37 cell line and purification of immunoglobulins by affinity to antigens detected by electrophoresis and blotting, a strict correlation between immunoreactive proteins and cytological staining was established. The nucleolar specificity was shown to correspond to a protein with an Mr of 80,000 while the centrosomal specificity corresponded principally to a protein doublet of 60,000–65,000. These antigens share common epitopes as shown by the staining of both NOR and centrosome by immunoglobulins purified by affinity to either type of protein.
