    Notizen: Recent carbonate sediments from Jamaican north coast fringing reefs were collected along three parallel traverses in the vicinity of Discovery Bay. Each traverse extended from near shore across the back reef, reef crest, and fore reef to a depth of 75 m. Relative abundances of the biotic constituents vary between sites, reflecting general patterns of reef community composition. The sediment is dominated by highly comminuted coral fragments (27·1% to 63·1%), plates of the calcareous green alga Halimeda (0·4% to 38·7%), coralline algae (4·7% to 16·2%) and the encrusting foraminiferan Homotrema rubrum (0·7% to 9·5%), with lesser amounts of other taxonomic groups (non-encrusting foraminifera 1·3–5·5%; molluscs 1·4–7·0%; echinoderms 0·9–5·0%). Coral fragments, coralline algae and particles of Homotrema rubrum dominate the sediments of the shallow portions of the fore reef (5–15 m), whereas plates of Halimeda are most abundant in sediments from the back reef and deeper portions of the fore reef (〈inlineGraphic alt="geqslant R: gt-or-equal, slanted" extraInfo="nonStandardEntity" href="urn:x-wiley:00370746:SED77:ges" location="ges.gif"/〉 24 m). Q-mode cluster analysis, using sediment constituent data, resulted in the delineation of four reef biofacies over the depth range of this study (1–75 m).

    Notizen: The Escherichia coli glpT gene encodes a transport protein that mediates uptake of sn-glycerol-3-phosphate. This permease is a member of a class of bacterial organophosphate permeases which transport substrates by antiport with inorganic phosphate. The glpT gene product, probably an oligomer of a single polypeptide chain, is thought to span the cytoplasmic membrane several times, as predicted by the hydropathic profile. Protein fusions, in which varying lengths of the amino-terminal end of the permease is attached to alkaline phosphatase (phoA) and to β-galactosidase (lacZ) were constructed. On the assumption that phoA fusions only exhibit high enzymatic activity when fused to extra-cytoplasmic regions of the target protein, whereas lacZ fusions will only be active when the β-galactosidase portion is attached to cytoplasmic domains of the target protein, the activities of the fusions were used to test a two-dimensional model for the permease. The model proposes that glpT contains 12 transmembrane segments divided by a larger cytoplasmic region. Despite some limitation caused by hot-spot sites of transpositions, the TnphoA approach was consistent with the model. In contrast, we feel that the enzymatic activity of lacZ fusions is only a limited parameter for studying the topology of a complex membrane protein.

    Notizen: The specific activities of 6 enzymes, which are involved in the synthesis and catabolism of membrane lipids, were monitored in plasma membranes isolated from petunia petals during senescence. These included phosphatidylinositol (PI) kinase (EC 2.7.1.67), phosphatidylinositol monophosphate (PIP) kinase (EC 2.7.1.68). diacylglycerol (DAG) kinase (EC 2.7.1.107), phospholipase A (EC 3.1.1.4) and PIP- and PIP2-phospholipase C˙(EC 3.1.4.3). Using endogenous substrate, the [32P]PA and [32P]PIP2 formation increased to 140 and 200%, respectively, of the day 1 value by 4 days after harvest. There was no significant change in [32P]PIP formation during the same time period. On the fifth day the petals wilted and the [32P]PA and [32P]PIP formation declined significantly. In contrast, the [32P]PIP2 formation remained high in the day 5 petals. When the lipid kinase activities were assayed in the membranes in the presence of exogenous substrate the specific activity of all of the enzymes increased. and the changes in [32P]PA production over the 5-day period were similar to those observed with endogenous substrate. When exogenous PI and PIP were added, however, there was no longer an increase in [32P]PIP2 formation by plasma membranes of day 4 petals and [32P]PIP formation significantly decreased. The relative decrease in PIP and PIP2 formation by day 4 membranes when exogenous substrate was added may have resulted from differences in the lipase activities in the day 1 and day 4 membranes. The plasma membrane A-type phospholipase activity increased throughout the 5 day period, and phospholipase C activity increased two-fold between day 1 and day 4. Such changes in the metabolism of the plasma membrane lipids during flower senescence would affect the ability of the petals to use inositol phospholipid-based signal transduction pathways.
