    Notes: The histology and mucus histochemistry of the pleuronectid post-gastric alimentary canal was examined using light and electron microscopy. Distinct differences in goblet cell mucus histochemistry were observed between species, with the two closest taxonomic species, the winter flounder and the yellowtail flounder showing the most diversity and the halibut showing regional variation. Numbers of goblet cells within post-gastric regions did not differ significantly between species, but were significantly different between regions within species increasing toward the rectum. The post-gastric region was divisible into two areas based upon the ultrastructural features of lipid digestion and absorption in the intestine and pyloric caeca, and of exogenous protein in the rectum. The combination of species-specific histochemical differences in mucus and general histological and ultrastructural differences within the post-gastric regions between these species suggest a correlation between lumenal environmental conditions/histology and natural prey preference.

    Notes: The distribution of digestive enzymes was studied using histoenzymological methods in yellowtail and winter flounder larvae reared on three different diets: live food, weaned at day 15, or starved. Alkaline phosphatase, dipeptidyl peptidase IV, aminopeptidase M and esterase were present at 3 days post-hatch and became differentially distributed coinciding with morphological development. For larvae fed a live diet, activity of these enzymes was present in the intestine of both species and rectum of yellowtail flounder. Alkaline phosphatase was also present in the post-oesophageal swelling (stomach anlage) of winter flounder. Activity of all enzymes was absent in starved winter flounder larvae and a decrease in aminopeptidase M and esterase activity occurred in starved yellowtail flounder larvae. Acid phosphatase was not identified in either species. The eVect of weaning on enzymatic activity was not evaluated fully as the larvae did not survive long enough after the introduction of the artificial diet to complete experimentation.

    Notes: The histological development and mucous histochemistry of the alimentary tract in larval yellowtail flounder were studied using light microscopy. Samples were taken when the larvae were first offered food at 3 days post-hatch, then at 7, 10, 29, 36, and 46 days post-hatch, at which time they were metamorphosing. Regional partitioning of the digestive tract into the buccal cavity, pharynx, oesophagus, post-oesophageal swelling (PES), intestine, and rectum was complete by day 10. Goblet cells were present only in the buccal cavity, pharynx and intestine by day 7, but increased in number and distribution as development continued. By day 29, the posterior zone of the oesophagus had a marked increase in goblet cell density and mucosal folding. At the transition from oesophagus to PES/stomach stratified epithelium with goblet cells changed abruptly to a columnar epithelium with no goblet cells. Multicellular glands in the PES of 36-day larvae allowed it to be defined as a stomach. The distinct brush border of columnar epithelium and the presence of goblet cells characterize the intestine and rectum. All goblet cells throughout the digestive tract were strongly positive for acid mucins as was the luminal layer of the stratified epithelia lining the buccal cavity, pharynx and oesophagus. The PES/stomach epithelium stained weakly for neutral mucins. No mucin staining was associated with the gastric glandular epithelium. The brush borders of the intestine and rectum were strongly positive for combinations of neutral and acid mucins.

    Notes: The crystal structure of bovine lens γIIIb-crystallin at 2.5 Å resolution previously reported was interpreted using a consensus sequence derived from related vertebrate sequences on the assumption that γIIIb-crystallin derived from the γC-crystallin gene. It has recently been shown that γIIIb is a product of the bovine γD gene. The structure of γIIIb has now been refined with the bovine γD sequence using new 1.95 Å resolution synchrotron data. The crystallographic R factor was 20.4% for all 33 104 reflection data between 8.0 and 1.95 Å measured at 277(1) K. The electron density fully supported the assignment of the γD sequence to γIIIb. The crystal belongs to space group P212121 with two molecules of molecular mass 20 749 Da in the asymmetric unit in which 219 water molecules were located. The two-domain four-Greek-key motif highly symmetrical protein is very similar in structure to γB-crystallin (81% sequence identity). There is a single amino-acid deletion in γD in the linker region connecting the two domains. The intermolecular oganization in the crystal lattice is quite different from γB as a result of key mutations involving surface residues Leu51, Ile103 and His155. These point mutations will contribute to the intermolecular behaviour of the γ-crystallins in the eye lens, where they are major components of the densely packed, high refractive index regions of the lens.

    Notes: Abstract The presence of elastic fibres in the extracellular matrix (ECM) provides physiologically important elastic properties for many tissues. Until recently, microfibrils, one component of the ECM, were thought primarily to serve as a scaffolding on which elastin is deposited during development to form elaunin fibres [1]. The most prominent protein that forms mammalian microfibrils is fibrillin. It is known that mutations in the fibrillin gene cause a heterogenous connective tissue disease called marfan syndrome [2], so information on mechanical properties of microfibrils or their role in tissue function would be useful. Microfibrils are also found in the ECM of some invertebrate tissues, and there is growing evidence that the protein forming the structure is homologous to mammalian fibrillin [3, 4]. It has been shown that the microfibril-based arterial wall of the lobster has viscoelastic properties [5], and we have now utilized this primitive artery to measure the modulus of elasticity of microfibrils. It is similar to that of the rubber-like protein elastin.

    Notes: Abstract. Several modifications of the 3H-tetracycline bone labeling method for measuring whole skeleton bone resorption were tested. Under steady state conditions of whole skeleton resorptive activity, bone labeling for intervals longer than 2 weeks prior to experimentation did not significantly alter the urinary 3H-tetracycline loss curve. The utilization of nonlinear regression analysis showed that the urinary loss of 3H-tetracycline was best described by double exponential equations, indicating the loss of label from two distinct and independent exchangeable bone compartments. This conclusion was supported by the finding that soft tissues were effectively depleted of 3H-tetracycline by 24 hours after the final injection of label. Hence, it was concluded that approximately 40% of the 3H-tetracycline loss from skeletal bone is associated with a ``fast'' compartment which is depleted within 6 or 7 days after label loading. The size and rate of 3H-tetracycline loss from the fast compartment decreased (40%) with age such that the depletion time remained constant between 8 and 24 weeks of age in both male and female rats. The remaining 60% of 3H-tetracycline loss from a ``slow'' compartment which was depleted in about 70 days in young (8 week) rats. This compartment, which is believed to reflect cell-mediated resorption of calcified bone, decreased in size with age in both male (50%) and female (30%) rats. The rate of label loss from this compartment, however, remained relatively high so that the depletion time decreased (∼35%) between 8 and 24 weeks of age. By determining whole skeletal mass and calculating these parameters on the basis of skeletal mass, we were further able to demonstrate significantly higher resorptive activity in female than in male rats by 24 weeks of age.

    Notes: Abstract. The dry weight and calcium content of whole skeletons was determined in both male and female weanling, adolescent, and mature rats of the spontaneously hypertensive (SHR), Wistar Kyoto (WKY), and Sprague-Dawley (SD) rat strains. Dry skeletal mass, calculated as percent of body mass (%BM), increased with maturity in each strain. In adolescent and mature animals, skeletal development (%BM) was greater in female rats. SHR showed greater skeletal development (%BM) than normotensive rat strains and this difference was detectable even in weanling animals. Despite these age, strain, and gender-related differences in skeletal development, the calcium content per unit skeletal mass was identical among the various groups examined. Regression analysis of the plot of skeletal mass as a function of body mass indicated that only a portion of mature skeletal development was related to body mass. The results show that skeletal mass developed in the rat is only partially determined by structural support demands.

    Notes: Abstract Isolates ofF. oxysporum collected from symptomless carnation cuttings from Australian carnation growers properties, together with isolates from national collections, were screened for pathogenicity and grouped according to vegetative compatibility and random amplified polymorphic DNA (RAPD) patterns. The collection of 82 Australian isolates sorted into 23 different vegetative compatibility groups (VCGs). Of 69 isolates tested for pathogenicity, 24 were pathogenic to carnations, while the remaining 45 were non-pathogenic. All pathogenic isolates were within two VCGs, one of which was also compatible with an isolate obtained from an international culture collection, and which is known to represent VCG 0021 and race 2. Race status of the two pathogenic VCGs remains unknown. The RAPD assay revealed distinct DNA banding patterns which could distinguish pathogenic from non-pathogenic isolates as well as differentiate between isolates from the two pathogenic VCGs.

    Notes: Abstract Rat aortic smooth muscle exhibits a remarkable capacity for stress relaxation, the release of tension following tissue stretch. Stress relaxation was markedly enhanced in contracted aortic rings compared with unstimulated tissue. The magnitude of stress relaxation in contracted aortic rings correlated well with the passive tension imposed on the tissue by stretching, but showed little relationship to changes in tissue length or to the level of tension developed in response to agonist stimulation prior to stretch. The enhancement of stress relaxation in precontracted tissue was not affected by intimal rubbing or treatment with L-NAME. By comparison, the removal of extracellular calcium markedly attenuated stress relaxation. In addition, the use of cytochalasin B to block actin polymerization inhibited stress relaxation, whereas colchicine, a drug used to cause microtubule disassembly, had no effect on the phenomenon. The results indicate that the enhanced stress relaxation in contracted tissue is a calcium-dependent process and is not due to passive tissue elastic properties. We suggest that stress relaxation may not involve cross-bridge formation but could be explained by the remodelling of a portion of the tension-bearing actin cytoskeleton.
