    Notes: Abstract We examined chemosensitivity to 5-fluorouracil (5-FU) in four human gastric cancer cell lines, by analyzing the expression of p53 and its related genes. Treatment with 1mM 5-FU induced variable degrees of apoptosis in the cultured cells. The apoptotic indices 72 h after treatment were approximately 14% in MKN-74 (wild-type p53 gene), 12% in MKN-45 (wild-type), 3% in MKN-28 (mutated) and 0.5% in KATO-III cells (deleted), respectively. On the other hand, 50 μM 5-FU had little effect on the induction of apoptosis in MKN-74 cells, the value being approximately 2% after 72 h. Induction of P53 expression was noted 3 h after initiating the treatment, followed by the induction of P21/Waf1 after 6 h in both MKN-74 and MKN-45 cells. The same expression mode was noted in MKN-74 treated with 50 μM 5-FU. Conversely, the level of P53 expression was constant in MKN-28 cells and absent in KATO-III cells, in which P21/Waf1 had never been induced. The Bax/Bcl-2 expression ratio was gradually elevated for up to 72 h in MKN-74 and MKN-45 cells treated with 1mM 5-FU; in contrast, it was unchanged in MKN-28 and KATO-III cells, and MKN-74 treated with 50 μM 5-FU. These results might indicate that (1) 1mM 5-FU induces apoptosis in cultured gastric cancer cells carrying the wild-type p53 gene, but not those carrying the mutated type or a gene deletion, and (2) the elevated Bax/Bcl-2 expression ratio plays a more crucial role than the higher expression of P21/Waf1 in the induction of p53- gene dependent apoptosis.

    Notes: Abstract The cell surface Fas antigen transducts an apoptotic signal by its crosslinking with Fas ligand or anti-Fas antibody in a variety of human cultured cells. In this study, we examined the expression of Fas antigen and its mediation of apoptosis in six human colorectal carcinoma cell lines. A flow cytometric analysis revealed that LoVo, DLD-1, WiDr and SW837 cell lines showed higher expression levels of Fas antigen, in contrast to lower expression in COLO201 and COLO320DM. Interferon-γ enhanced the expression of Fas antigen in all of the cell lines examined. Both Fas ligand and Fas-associated phosphatase-1 (FAP-1) were expressed only in COLO320DM. Anti-Fas antibody induced apoptosis in LoVo carrying wild-type p53 gene, but not in the other five cell lines carrying mutated p53 gene. The transfection of wild-type p53 gene using an adenovirous vector upregulated P53 protein in WiDr and SW837 cells, both of which showed, however, no increase in apoptotic cells by anti-Fas antibody treatment. These results indicated that (1) Fas antigen was variably expressed, regardless of the p53 gene status and (2) the susceptibility to anti-Fas antibody-mediated apoptosis did not correlate to Fas, Fas ligand or FAP-1 expression levels. Therefore, we conclude that wild-type P53 expression might not necessarily be essential for Fas-mediated apoptosis in human colorectal carcinoma cell lines.

    Notes: New semirigid thermotropic liquid crystalline (LC) copolyurethanes 4 and 5 made up of biphenyl units and partially fluorinated aliphatic chains in the backbones were synthesized by melt polycondensation of a mixture of a dioxydihexanol of biphenyl 1 and two fluorine-containing diols 2a,b taken in definite feed mole ratios with alkylene diphenyl dicarbamates 3a-i having various lengths of aliphatic chains. The assigned structures of copolyurethanes 4 and 5 were identified by FTIR, 1H- and 13C-NMR spectra, and elemental analyses. The thermal and mesogenic properties were evaluated by differential scanning calorimetry (DSC), thermal mechanical analyses (TMA), polarizing microscopy, and temperature-changeable X-ray analyses, whose measurements indicated that the copolymers 4 and 5 form thermotropic nematic phases and have glass transition steps around room temperature. It is suggested that the incorporation of partially fluorinated aliphatic chains into the backbones has no drastic effect on the LC formation in the semirigid copolyurethanes 4 and 5. © 1998 John Wiley & Sons, Inc.     Notes: The interfacial activity and the molecular structure of functional monomers are critical factors that determine the success of synthesizing metal-imprinted polymers by surface template polymerization. From this point of view, first we prepared three distinct novel bifunctional organophosphorus monomers that are interspaced, in each case, by an alkyl spacer having a specific length. Each monomer carries two phosphonic acid groups and two aromatic groups in its molecular structures. Further, by using the synthesized bifunctional monomers, we prepared highly selective Zn(II)-imprinted polymers by the surface template polymerization initiated from a water-in-oil emulsion. To evaluate the template effect, we conducted diagnostic adsorption studies on Zn(II)-imprinted and unimprinted polymers with zinc ions. A high interfacial activity was found to be required for the functional monomers to create the predominant template effect. It became clear that Zn(II)-imprinted polymers having bifuctional monomers with 12-length alkyl chains (1,12-dodecanediol-O, O′-diphenyl phosphonic acid: DDDPA) yielded the best results. Moreover, analysis results of adsorption behavior supported a high-performance of the Zn(II)-imprinted polymers with DDDPA. © 1998 John Wiley & Sons, Inc. The result was given by the Arrhenius equation, kd = A exp(-E/(RT)) with A = 3.0 × 1013 s-1 and E = 124 kJ · mol-1.

    Notes: Novel semi-rigid fluorocarbon-containing coplycarbonates composed of biphenyl moiety and rigid rod-like hexafluoropentane chain were prepared by melt polycondensation of 6,6′-(biphenyl-4,4′-diyldioxy)dihexanol (1) and 2,2,3,3,4,4-hexafluoro-1,5-pentanediol (2) with alkylene diphenyl dicarbonates 3 of various aliphatic lengths (m = 6, 8, 10 and 12). The structures of the copolymers 4 were confirmed by FT-IR, 1H and 13C NMR spectra and elemental analyses. The thermal and liquid-crystalline (LC) properties were examined by differential scanning calorimetry (DSC), polarizing microscopy and temperature-changeable X-ray analyses. These measurements indicated that the copolymers show well-defined thermotropic nematic textures and have block-like sequences. It is suggested that the introduction of the hexafluoropentane chain into the main chain gives more stable LC phases.

    Notes: Gas chromatography was employed to quasi-continuously determine the amount of carbon dioxide that evolved from carbonate apatite specimens during sintering. Assuming that the carbonate in the specimens decomposed to carbon dioxide on a mole-for-mole basis, the determination of the carbon dioxide evolved allowed for the determination of the amount of carbonate that remained in the specimens during different stages of sintering. Previously, this measurement could be carried out only after sintering was completed. Comparison of data obtained from specimens compacted isostatically at 600 MPa for sintering with powder specimens indicated that the amount of carbonate remaining in the sintered apatite mass strongly depended on heating rates, heating temperatures, and holding-time intervals. © 1995 John Wiley & Sons, Inc.

    Notes: Block copolymers consisting of poly(γ-benzyl L-glutamate) and poly(ethylene oxide) as the hydrophobic and hydrophilic components, respectively were prepared. Cell attachment onto the surfaces of block copolymers fabricated either as well-defined ordered Langmuir-Blodgett (LB) films or solvent cast microphase-separated structures was studied. On ordered LB surfaces, adherent fibroblasts were larger than on the microphase-separated cast surfaces within 15 min. The difference in cell adhesion between LB films and cast surfaces increased with increasing PEO contents in block copolymer. Adherent cells increased with an increase in surface pressure of LB films in any polymer. Phase-contrast microphotographs of adherent cells showed rapid and extensive morphologic changes associated with the LB surface as compared to cast film surfaces. The number of cells grown on the LB surface is greater than that on the cast film. © 1996 John Wiley & Sons, Inc.
