    Notes: Although it has been assumed that the effects of nonsteroidal antiinflammatory drugs (NSAIDs) are mainly the result of their action on local synthesis of prostaglandins, there is growing evidence to suggest that they may also exert a central analgesic action. Some authors have suggested that inhibition of prostaglandin synthesis in the brain could contribute to the analgesic action. The effect of dexketoprofen trometamol (tromethamine salt of the enantiomer (+)-S-ketoprofen) on prostaglandin synthesis was investigated in rat brain fragments and in cyclooxygenase preparations from rat brain microsomes. Effects of the (-)-R-enantiomer and the racemic mixture were also evaluated. Significant levels of prostaglandin F2α (PGF2α) were synthesized in rat brain fragments after 10 min of incubation at 37°C. Dexketoprofen was found to be a potent inhibitor of this PGF2α production in rat brain (IC50 = 6.2 nM), and it completely suppressed PGF2α production at 1 μM concentration. In addition, inhibition of PGF2α synthesis by dexketoprofen was highly stereoselective since the enantiomer (-)-R-ketoprofen was significantly less potent (IC50 = 294 nM); with this enantiomer, even at high concentrations such as 1 μM, less than 60% inhibition was achieved. These results correlated with those obtained in the study of racemic ketoprofen and its enantiomers on cyclooxygenase activity of rat brain microsomes, where dexketoprofen also inhibited enzymatic activity stereoselectively. IC50 values obtained for dexketoprofen, (-)-R-ketoprofen, and rac-ketoprofen were 3.5 μM, 45.3 μM, and 5.8 μM, respectively. The above results could be related to the potent analgesic effect of dexketoprofen observed in vivo, which was also stereoselective. Taken together, these findings suggest that prostaglandin synthesis inhibition in rat brain by dexketoprofen could be associated, at least in part, with the analgesic effect of this NSAID.     Notes: Phenoxarsin-10-yl derivatives of 2-amino-cyclopent-1-ene-1-carbodithioic acid, (ACDA), and its N-alkyl derivativesO(C6H4)2 AsS2C - C5H6 -  NHR-2 (R = H, CH2CH3, CH2C6H11,), have been prepared by reacting O(C6H4)2AsCI with the corresponding ACDA 1,1-dithioic acid. The compounds were obtained by stirring stoichiometric amounts of the reagents in ethanol, over 24 h, at room temperature. The scale of the preparations were in the order of 2 mmol and the yields of the compounds ca 75%. The reactions were carried out in absolute ethanol. The compounds were characterized by IR, mass and NMR (1H, 13C) spectroscopy. The molecular structure of O(C6H4)2AsS2C - C5H6 - NH2-2 was determined using X-ray diffractometry, achieving an R-value of 6.3%; this compound is monomeric and contains an asymmetric monometallic biconnective 1,1-dithiolato ligand [As - S(1) 2.272(2) å, As … S(2) 3.125(2) å]. An intramolecular hydrogen bond is established between one hydrogen atom of the NH2 group and the sulfur [S(2)] atom involved in the secondary interaction to arsenic. The dihedral angle (150.3(3)°) of the phenoxarsine moiety is practically unaffected by substitution of chlorine on arsenic by the carbodithioato ligand.

    Notes: Abstract Acetonylacetone underwent both acid‐ and base‐catalyzed intramolecular cyclizations to 2,5‐dimethylfuran and 3‐methyl‐2‐cyclopenten‐1‐one, respectively, on pure AlPO4, whereas its modification with increasing amounts of cesium oxide developed AlPO4‐based materials with increased basic properties and, hence, higher selectivities to the base‐catalyzed cyclization process. However, the poisoning of surface acid and base sites indicated that acetonylacetone conversion into 3‐methyl‐2‐cyclopenten‐1‐one can also be attributed to the joint participation of surface acid and base sites. So, acetonylacetone cannot be used for the simultaneous characterization of acidic and basic surface properties of acid–base heterogeneous catalysts.

    Notes: The hydroxo complex (Bu4N)2[Ni2(C6F5)4(μ-OH)2]reacts with 2,3,4,5,6-pentafluoro benzenamine (C6F5-NH2), 1,3-diaryltriaz-1-enes (ArNH—N=N—Ar, Ar = Ph, 4-MeC6H4, 4-MeOC6H4), 7-aza-1H-indole (= 1H-pyrrolo[2.3-b]pyridine; Hazind), N-phenylpyridin-2-amine(pyNHPh), and N-phenylpyridine-2-carboxamide (py-CONHPh) at room temperature in acetone to give the binuclear complexes (Bu4N)2[Ni2(C6F5)4(μ-C6F5NH)2] (1) and (Bu4N)2[{Ni(C6F5)2} 2(μ-OH)(μ-azind)] (2) and the mononuclear complexes Bu4N[Ni(C6F5)2(ArN3Ar)] (3-5), Bu4N[Ni(C6F5)2(pyNPh)] (6), and Bu4N[Ni(C6F5)2(pyCONPh)] (7). The hydroxo.complex (Bu4N)2[{Ni(C6F5)2-(μ-OH)}2] promotes the nucleophilic addition of water to pyridine-2-carbonitrile, 2-aminoacetonitrile, and 2-(dimethylamino)acetonitrile, and complexes 8-10 containing pyridine-2-carboxamidato, 2-aminoacetamidato and 2-(dimethylamino)acetamidato ligands are formed. Analytical (C, H, N) and spectroscopic (IR, 1H and 19F-NMR, and FAB-MS) data were used for structural assignments. A single-crystal X-ray diffraction study of (Bu4N)2[{Ni(C6F5)2}2(μ-OH)(μ-azind)] (2) established the binuclear nature of the anion; the two Ni-atoms are bridged by an OH group and a 7-aza-7H-indol-7-yl group, but the central Ni—O—Ni—N—C—N ring is not planar, the dihedral angle between the Ni—O—Ni and Ni—N—C—N—Ni planes being 84.4°.

    Notes: The kinetics of the reaction between pyridoxal 5′-phosphate (PLP) with carbidopa, hydralazine, and isoniazid, in aqueous solution at variable pH and constant ionic strength of 0.1M was studied spectrophotometrically. The rate constants of formation and hydrolysis of the resulting Schiff base, and its stability were determined in a wide range of pH. A comparison is made of the formation rate constants with those of PLP with hydrazine. The reactivity shows the sequence isoniazid 〉 hydrazine 〉 carbidopa 〉 hydralazine in the whole range of pH studied. The Schiff bases studied are more stable than those formed by PLP and hexylamine and as stable as those described for the reactions of PLP with poly(L-lysine) or copolypeptides containing L-lysine.

    Notes: In the context of our studies on ruthenium(II) complexes containing polyazaheterocyclic ligands, we have determined the rate constants of quenching by molecular oxygen (kq) of the metal-to-ligand charge-transfer-excited state of a series of homoleptic [RuL3] complexes (where L stands for 2,2′-bipyridine (bpy), 1,10-phenanthroline (phen), 2,2′-bipyrazine (bpz), 4,7-diphenyl-1,10-phenanthroline (dip), diphenyl-1,10-phenanthroline-4,7-disulfonate (dpds), and 1, 10-phenanthroline-5-octadecanamide (poda)) in H2O and in MeOH. These compounds are singlet-oxygen (O2(1Δg)) sensitizers, and quantum yields of singlet-oxygen production (ΨΔ) in both solvents are also reported. Values of kq and ΨΔ depend on the nature of the ligand L and on the solvent, ΨΔ values showing a large range of variation (0.2 to 1.0). In MeOH, the only pathway for quenching of the excited [RuL3] complexes by molecular oxygen is energy transfer: the fraction of quenched excited states yielding singlet oxygen (ƒΔT) is unity for all compounds in the series investigated. Changing from MeOH to H2O has several remarkable effects: higher kq and lower ΨΔ values are observed; ƒΔT drops to ca. 0.5 except for [Ru(bpz)3]2+. In fact, [Ru(bpz)3]2+ is by far the weakest reductant in the series and behaves differently from the other complexes, with lowest kq and ΨΔ values and a ƒΔT equal to 1 in both solvents. Results are interpreted on the basis of the role played by charge-transfer interactions between the sensitizer excited state and molecular oxygen in the quenching mechanism. RuII Complexes based on the 4,7-diphenyl-1, 10-phenanthroline (dip) ligand are very efficient and stable singlet-oxygen sensitizers with ΨΔ values close to unity in air-saturated MeOH.

    Notes: The degradation of cefaclor (1), an oral cephalosporin antibiotic, was studied at 37° in a neutral aqueous medium by HPLC and 1H-NMR. Under these conditions, 1 underwent intramolecular aminolysis by the 7-side-chain NH2 group on the β-lactam moiety to give a piperazine-2,5-dione. The most prominent peak in the HPLC profile of a degradation solution from 1 was isolated by prep. HPLC. Mechanistically, the formation of this degradation product cis-11 from 1 involves the contraction from a six-membered cephem ring to a five-membered ring, which presumably takes place via a common episulfonium ion intermediate 9 (see Scheme). Loss of the Cl-atom from 3-chloro-3-cephem is a general reaction subsequent to β-lactam ring opening.

    Notes: The syntheses of several N-aryl-3-amino-4-nitroisothiazol-5(2H)-imines 12 from 3, 3-diamino-2-nitro-thioacrylamides 11 are reported (Scheme 3). In polar solvents, a spontaneous isomerization of some of the prepared isothiazol-5(2H)-imines 12 yielded benzothiazoles 13 (Scheme 4). In the case of 2-alkyl-substituted derivatives of type 12, the isomerization occurred only at higher temperatures. Electronic influences of different substituents on the rate of the isomerization were studied, and a polar reaction mechanism is proposed in Scheme 6. The structures of 12e and 13e were established by X-ray crystallography. Conformational analyses of 3-(methylamino)-2-nitro-N-phenyl-3-(pyrrolidin-1-yl)thioacrylamide (111) by NMR and X-ray methods were performed with the aim of explaining the distinct behavior of this amide towards Br2 or diethyl azodicarboxylate.

    Notes: A comprehensive study of the alkaline hydrolysis of the β-lactam ring of azetidin-2-one was carried out using ab initio molecular-orbital calculations at the RHF/6-31 + G* level. The influence of the solvent on this reaction was investigated by using the reaction field method (SCRF); the solvent was found to suppress the interference of some gas-phase reactions and allow the presence of a transition state to be detected as the nucleophile approaches the β-lactam ring. The transition state corresponds to a structure where the OH- group lies at a distance of 1.927 Å from the C=O group of the β-lactam ring and exhibits a potential barrier of 13.6 kcal/mol.
