    Notes: The emergence of new races of Phytophthora infestans has necessitated the search for additional sources of potato germplasm with resistance to late blight. This study examined 281 clones, derived from 72 families of a diploid random-mated hybrid population of Solanum phureja×Solanum stenotomum. The clones were evaluated in a replicated field trial for 2 years with the control cultivar ‘Atlantic’ in Pennsylvania, USA. The P. infestans US-8 A2 mating type culture was used to inoculate spreader rows of susceptible S. tuberosum cv.‘Russet Burbank’. Percent defoliation caused by the late blight fungus was estimated visually in each plot three times near the end of the growing season. Area under the disease progress curve (AUDPC) was estimated and showed that late blight was more severe in 1997 than in 1996. Genetic differences among clones and significant, but small, clone–environment interaction were detected for AUDPC. Broad-sense and narrow-sense heritability estimates, over years, were 0.79 ± 0.05 (P = 0.05) and 0.78 ± 0.29, respectively. Seventy-five percent of the diploid clones had a significantly lower mean AUDPC than Atlantic. These results support the idea that this diploid population is worthy of use in breeding for late blight resistance in tetraploid potato cultivars.

    Notes: Computer-assisted semen analysis (CASA) technology was applied to the measurement of sperm motility parameters in the common carp Cyprinus carpio. Activated sperm were videotaped at 200 frames s−1 and analysed with the CellTrak/S CASA research system. The percentage of motile cells and both sperm head curvilinear velocity and straight-line velocity were measured following exposure of carp sperm to three predilution conditions and activation in media of differing ionic strengths and osmotic pressures. The highest percentage of motile sperm was obtained following predilution of sperm in seminal plasma and activation in Na-HEPES buffer pH 8.0. This level of motility was equalled after predilution in 200 mm KCl for 2 h. Straight-line velocities and curvilinear velocities of 130 μm s−1 and 210 μm s−1, respectively, were observed. Duration of motility was higher under seminal plasma predilution conditions (over 50% motile sperm at 55 s post-activation). The application provides a sound basis for the assessment of Sperm Characteristics in fish.

    Notes: Sperm motility variables from the milt of the common carp Cyprinus carpio were assessed using a computer-assisted sperm analysis (CASA) system across several months (March-August 1992) known to encompass the natural spawning period. Two-year-old pond-raised males obtained each month were primed with an exogeneous hormone injection of carp pituitary extract and stripped of milt 18–24h later. The milt was diluted, activated and videotaped using a high-speed (200 Hz) videocamera and recorder. Videotaped samples were subsequently analysed using the CellTrak/S CASA system (Motion Analysis Corp.) for percent motility, curvilinear and straight-line velocity. In addition to assessing changes in motility parameters across several months, a comparison was made between two predilution/ activation media combinations (homologous seminal plasma/NaCl+HEPES v. 2-h incubation in 200 mM KCl+Tris/Tris). The percentage of motile cells assessed immediately after activation was significantly greater in the summer months (May, June, July) when compared to a spring sampling point (March); when assessed 1 min after activation, cells prediluted in seminal plasma maintained a higher percentage of motility than those prediluted in KC1. Curvilinear and straight-line velocities exhibited a slight seasonal trend; variations in response to the predilution treatments were observed with these measurements. Sperm count gradually increased through April and May (9–63 × 109 to 2–38 × lO10ml– 1 milt), declined in June and July (to 1–83 × lO10ml– 1 milt), and was followed by a steep increase in August (2–74 × 1010 ml– 1 milt). Mean seminal plasma osmolality remained relatively constant (250–265 mOsmol kg – 1) throughout the sampling period.

    Notes: An understanding of the mechanisms involved in the regulation of reproductive processes in yellow perch is fundamental for intensive culture of this commercially important, freshwater, perciform fish. This paper describes the annual reproductive cycle of female and male perch. It presents the current state of knowledge on the regulation of ovarian steroidogenesis, final maturation of oocytes and ovulation in this species. Moreover, data concerning biochemistry of spermatozoa and seminal plasma as well as testicular steroidogenesis are summarized. In addition to endocrinological factors, the involvement of environmental (photoperiod and temperature) cues in the regulation of yellow perch reproduction is reviewed. Finally, practical aspects of controlling perch reproduction are outlined.

    Notes: Abstract— In order to study the cyclic stress-strain behaviour under variable amplitude loading in terms of dislocation glide character and microstructure, various metals and alloys were cyclically loaded in single-step tests and in an incremental step test (IST). The cyclic stress-strain curves (CSS curves) which correspond to the different loading conditions were compared and the shapes of the hysteresis loops were analysed with respect to Masing or non-Masing behaviour.As expected, cyclic loading with constant amplitude leads to the formation of a characteristic dislocation arrangement that depends on the amplitude applied. The amplitude-related microstructure corresponds to the observation that Masing behaviour was not found in single-step tests. However, in the IST a state of saturation, or at least approximate saturation, is attained after some or many loading blocks. In the case of single-phase polycrystalline materials, wavy slip behaviour was found to be a prerequisite for the fulfilment of Masing behaviour. Then, the CSS curve, obtained by means of IST, intersects that of the single-step tests at an intermediate amplitude. Planar-slip materials show very similar CSS curves in spite of considerable differences in the dislocation arrangements for different testing modes. The behaviour of particle-hardened alloys depends very strongly on the relative effectiveness of the precipitates as obstacles for dislocation motion and can be understood analogously to single-phase materials on the basis of the resulting dislocation glide character.

    Notes: In order to characterize the influence of hydrogen on the mechanical properties of β-titanium alloys, monotonic tensile and strain-controlled fatigue tests were performed on samples of the metastable alloy Ti-3Al-8V-6Cr-4Mo-4Zr in uncharged (0.5 at.% hydrogen) and hydrogen-charged (3-4 at.% hydrogen) conditions. The hydrogen was introduced into the material during the last 8 h of an ageing treatment (28 h at 482°C) from the gas phase, whereas the reference (uncharged) specimens were annealed completely in vacuum. The results of the mechanical tests indicate that hydrogen slightly increases the strength of the alloy in monotonic as well as in cyclic loading. Under tensile loading the fracture strain decreases as a result of hydrogen. Under cyclic loading both charged and uncharged conditions show initial softening followed by a saturation state. The cyclic lifetime at a constant total strain amplitude, however, is not reduced by the hydrogen charging. The effect of hydrogen on the mechanical behaviour can be interpreted and understood on the basis of microstructural observations that reveal a hydrogen-induced change in the precipitation state. This indirect influence of hydrogen on the microstructure, which leads to a reduction of the mean size of the α-precipitates, in combination with a slight decrease on the volume fraction of the α-phase, seems to dominate over any direct intrinsic hydrogen effect
