    Notes: . The antiproliferative effects and ultrastructural alterations induced in vitro by two antifungal compounds, the azole ketoconazole and the allylamine terbinafine on Leishmania amazonensis are reported. Promastigotes treatment with ketoconazole and terbinafine induced growth arrest and cell lysis in 72 hours. Combination of the two agents produced additive effects on promastigote axenic growth and synergistic effects on intracellular amastigote proliferation. The amastigotes, either axenically grown or infecting murine macrophages, were about 100-fold more sensitive to the drugs. These compounds induced the appearance of large multivesicular bodies, especially after ketoconazole treatment, increased amount of lipid inclusions as well as numerous, polymorphic volutin granules, particularly in terbinafine-treated cells. Multivesicular bodies were observed in close apposition with organelles such as mitochondria, which also showed alterations in the distribution and appearance of cristae, and the formation of paracrystalline arrays within the matrix. Some cells presented large portions of cytoplasm wrapped by endoplasmic reticulum and many parasites also presented myelin-like endoplasmic reticulum profiles. Such alterations together with the strong acid phosphatase activity observed in the multivesicular bodies and volutin granules may indicate the existence of an unusual autophagic process in cells treated with ergosterol biosynthesis inhibitors.

    Notes: . Tritrichomonas foetus is a flagellated protozoon found in urogenital tract of cattle. Its free movement in liquid medium is powered by the coordinated movement of three flagella projecting towards the anterior region of the cell, and one recurrent flagellum that forms a junction with the cell body and ends as a free projection in the posterior region of the cell. We have used video microscopy and digital image processing to analyze the relationships between the movements of these flagella. The anterior flagella beat in a ciliary type pattern displaying effective and recovery strokes, while the recurrent flagellum beats in a typical flagellar wave form. One of the three anterior flagella has a distinctive pattern of beating. It beats straight in its forward direction as opposed to the ample beats performed by the others. Frequency measurements obtained from cells swimming in a viscous medium shows that the beating frequency of the recurrent flagelium is approximate twice the frequency for the three anterior flagella. We also observed that the costa and the axostyle do not show any active motion. On the contrary, they form a cytoskeletal base for the anchoring and orientation of the flagella.

    Notes: The crustal structure of the south-eastern Brazilian continental shelf has been investigated through the dispersion curves of seismic surface waves obtained at Brazilian seismological station RDJ. A set of eight continental-shelf models has been tried, to obtain the best shear-wave velocity structure. The inversion results suggest a sedimentary structure divided into two layers with shear-wave velocities of 1.36 ± 0.10 km s−1 and 1.58 ± 0.10 km s−1, respectively. This is in agreement with preliminary results found by the Brazilian Petroleum Company Petrobras using seismic data in the area (O. R. Sá & L. P. A. Gamboa, private communication, 1994). The intermediate crustal layer has a shear-wave velocity (3.26 ± 0.10 km s−1 typical of basement. An anomalous shear-wave velocity of 3.98 ± 0.10 km s−1 has been determined for the lower crust. This suggests the existence of a transitional layer between lower crust and upper mantle, similar to that observed in both Mediterranean and Canadian passive continental margins. It was not possible to define precisely the crust-mantle transition, but an analysis of the inversion results has shown that the Moho boundary is at around 40 ± 5 km depth.

Growth by serial transfers of the trypanosomatid Crithidia deanei in culture medium containing 1 mg/ml of the β-lactam antibiotics ampicillin or cephalexin resulted in shape distortion of its endosymbiont. The endosymbiont first appeared as filamentous structures with restricted areas of membrane damage. An increase of electron lucid areas was also observed in the endosymbiont matrix. The continuous treatment with β-lactam antibiotics, resulted in endosymbiont membranes fragmentation; and later on the space previously occupied by the symbiont was identified as an electron lucid area in the host cytoplasm. The putative targets of β-lactam antibiotic were two membrane-bound penicillin-binding proteins (PBPs) detected in the Sarkosyl-soluble fraction of purified symbionts labeled with [3H]-benzylpenicillin. The apparent molecular weight of the proteins were 90 kDa (PBP1) and 45 kDa (PBP2). PBP2 represented 85% of the total PBP content in the membrane fraction of the endosymbionts. Competition experiments using the tested antibiotics and [3H]-benzylpenicillin showed that ampicillin and cephalexin have half saturating concentrations considerably higher than [3H]-benzylpenicillin and indicated that PBP1 is the probable lethal target of the antibiotics tested. These results suggest that a physiologically active PBP is present in the cell envelope of C. deanei endosymbionts and may play important roles in the control of processes such as cell division and shape determination.

    Notes: Mass seed production depends a great deal on the development of adequate artificial diets, which in turn depends on the development of methods to speedily and reliably assess the nutritional value of experimental diets. This study examined the sensitivity of light histological and ultrastructural criteria to detect differences in the nutritional value of diets for the Japanese spiny lobster, Panulirus japonicus (V. Siebold), phyllosoma larvae. The acceptability of diets, the apparent absorption of lipids and carbohydrates by the midgut gland cells, and the characteristics of various cell types were determined in 43-, 106- and 143-day-old intermoult phyllosoma fed diets of Mytilus edulis (L.) ovary alone (MO) or in combination with Anemia (MO+AR), which so far have given the best results in terms of growth and percentage of metamorphosis into puerulus, and two other diets considered less efficient, viz. Mytilus edulis testis (MT) and Artemia (AR).Acceptability of all diets attained 100% within 10 min of feeding as revealed by observation through the semitransparent cuticle of the larvae. Histological examination of larvae that ingested comparable amounts of food revealed pronounced variation in the absorption of lipids and carbohydrates by the midgut gland cells (AR〈MT≤MO+ARp〈MO), in the size and ultrastructure of the midgut gland epithelial cells (AR〈MT〈MO+AR〈MO), in the number, size, and electron-dense body content of the haemocytes (AR〈MT≤MO+ARp〈MO), and in the size of cells lining the midgut gland tubules (AR≤MT≤MO+AR〈MO). Growth and survival followed the order MT≤AR〈MO+AR≤MO with maximum survival rates around 25–35% on the more efficient diets. Thus, phyllosoma fed on each diet presented a distinctive set of histological characteristics. Moreover, histological variation between diets was detected earlier than, and yet correlated strongly with, the results on growth and survival. This suggests that histological criteria constitute a practical means for the preliminary assessment of the acceptability and nutritional value of diets for the Japanese spiny lobster.
