    Notes: . In vitro excystation of Spironucleus muris cysts, purified by sequential sucrose and Percoll gradients from mouse feces, was studied. Three in vitro excystation procedures, used for Giardia, were assessed to determine the most useful method. Excystation was monitored by light microscopy and subsequently characterized by transmission and scanning electron microscopy. Spironucleus muris excysted routinely at a level greater than 90% when induced in Hanks’ balanced salt solution containing sodium bicarbonate at pH 2.0 and transferred to Tyrodes’ salt solution as an excystation medium. Similarly, high rates of excystation were recorded after induction of S. muris cysts in 0.1 M potassium phosphate buffer (pH 7.0) with sodium bicarbonate and excystation in trypticase-yeast extract-iron medium (TYI medium) or phosphate-buffered saline. A lower rate and percentage of excystation were observed after induction of S. muris cysts in an aqueous hydrochloric acid solution (pH 2.0) followed by excystation in TYI medium. All excystation methods produced extremely active S. muris trophozoites with normal morphology. Nonexcysting S. muris cysts have a wall composed of an outer fibrous and an inner membranous portion. Following induction, numerous vesicles appeared in the peritrophic space. Excystation began by the cyst wall opening at one pole, and the anterior part of the trophozoite protruding from the cyst wall. The trophozoite emerged progressively from the cyst wall and the empty cyst wall appeared to collapse. Excysted trophozoites exhibited normal morphological features of S. muris trophozoites isolated from the mouse intestine.

Die Kristallstruktur (tetragonal; P4/mbm; Z = 2; a = 1347,5(1); c = 1212,5(1) pm) ist ähnlich wie jene von Na[Mo4]O6 bzw. [Sc4B]Cl6. Trans-kantenverknüpfte [Gd6]-Oktaeder verlaufen parallel [001]. Sie enthalten interstitielle C2-Einheiten, Jedes dritte Oktaeder enthält fehlgeordnete C2-Einheiten, senkrecht zu jenen in den benachbarten [Gd6(C2)]-Oktaedern. Diese sind daher entlang der (pseudo)-C4-Achse gestaucht. Rechnungen zur elektronischen Struktur zeigen, daß insgesamt 13 Elektronen zur Auffüllung aller Metall-Metall-bindenden Zustände für eine „leere“ [Gd4]Cl6-Struktur nötig wären. Die Einlagerung der C2-Dimeren verändert die Bindungsverhältnisse in [Gd4(C2)]X6 (X = Cl, I) erheblich. Die formale Ladung von -6 der C2-Einheit wird durch das Aufsplitten der πg-Zustände reduziert, Gd—Gd und Gd—C-bindende Zustände werden besetzt und bindende dx2-y2-Orbitale kombinieren zu den am niedrigsten liegenden nicht besetzten Zuständen.
    Notes: [Gd4(C2)](Cl, I)6 is obtained from CsI, Gd, GdCl3 and C2I4 in sealed niobium containers at 1000/800°C as black, shiny needles. The crystal structure (tetragonal, P4/mbm, Z = 2, a = 1347.5(1), c = 1212.5(1) pm) is similar to that of Na[Mo4]O6 and [Sc4B]Cl6. It may be regarded as being built from octahedra sharing common trans edges running in the [001] direction. The octahedra contain C2 units as interstitials. Every third octahedron contains a disordered C2 unit perpendicular to those in the two neighboring [Gd6(C2)] octahedra and is therefore compressed in the direction of the (pseudo) C4 axis. Calculations of the electronic structure of an “empty” [Gd4]Cl6 structure reveals a total of 13 electrons necessary to occupy all metal-metal bonding states. The incorporation of a carbon dimer substantially alters the bonding conditions for [Gd4(C2)]X6 (X = Cl, I). The formal charge of -6 of the C2 unit is significantly reduced as πg states split up, Gd—Gd and Gd—C bonding states are occupied and bonding dx2—y2 orbitals combine to form the lowest unoccupied energy states.

    Notes: Oyster cultch was added to the lower intertidal fringe of three created Spartina alterniflora marshes to examine its value in protecting the marsh from erosion. Twelve 5-m-wide plots were established at each site, with six randomly selected plots unaltered (non-cultched) and cultch added to the remaining (cultched) plots. Within each cultched plot, cultch was placed along the low tide fringe of the marsh during July 1992, in a band 1.5 m wide by 0.25 m deep. Marsh-edge vegetation stability and sediment erosion were measured for each plot from September 1992 to April 1994. Significant differences (p 〈 0.05) in marsh-edge vegetation change were detected at the only south-facing site after a major southwester storm. Significantly different rates of sediment erosion and accretion also were observed at this same site. Areas upland of the marsh edge in the cultched areas showed an average accretion of 6.3 cm, while noncultched treatment areas showed an average loss of 3.2 cm. A second site, with a northern orientation, also experienced differential sediment accretion and erosion between treatment type, caused instead by boat wakes that were magnified by the abutment of a dredge effluent pipe across the entire front fringe of the site. During this period we observed significant differences in sediment accumulation, with the areas upland of the marsh edge in the cultched treatment having an average accretion of 2.9 cm and the noncultched an average loss of 1.3 cm.

    Notes: In ciliates, development of the polyploid somatic macronucleus after sexual events involves extensive and reproducible rearrangements of the germ-line genome, including chromosome fragmentation and precise excision of numerous internal sequence elements. In Paramecium aurelia, alternative macronuclear versions of the same germ-line genome can be maternally inherited across sexual generations, showing that rearrangement patterns are not strictly determined by the germ-line sequence. Homology-dependent maternal effects can be evidenced by transformation of the vegetative macronucleus with cloned macronuclear sequences: new fragmentation patterns or internal deletions are specifically induced during differentiation of a new macronucleus, in sexual progeny of transformed clones. Furthermore, transformation of the maternal macronucleus with germ-line sequences containing internal eliminated sequences (short single-copy elements) can result in a specific inhibition of the excision of the same elements in the zygotic macronucleus. These experiments show that the processing of many germ-line sequences in the developing macronucleus is sensitive to the structure and copy number of homologous sequences in the maternal macronucleus. The generality and sequence specificity of this trans-nuclear, epigenetic regulation of rearrangements suggest that it is mediated by pairing interactions between germ-line sequences and sequences imported from the maternal macronucleus.

    Notes: An analysis was conducted of multidimensional subsurface moisture flow in a hypothetical low-level radioactive waste disposal facility. The key design feature examined in the analysis was a sloping sand/gravel capillary barrier designed to route natural infiltration around a concrete vault. Three barrier slopes (1:25, 1:10, and 1:5) and three sand/gravel property combinations were considered. The slopes and material property combinations were selected to represent a range in barrier effectiveness. The porous media flow code used for this analysis was VAM3DCG, a three-dimensional, finite-element code which was able to use nonorthogonal grid discretizations and employed robust, efficient numerical techniques. Three-dimensional modeling demonstrated that flow in the hypothetical design exhibited cross-slope flow because of the pressure gradient produced in the third, cross-slope dimension. Barrier effectiveness was shown to be highly sensitive to the sand/ gravel material properties. Barrier slope was less important, especially for the effective material combinations. The presence of three-dimensional flow could be important in a performance assessment if the quantity of water predicted to breach the capillary barrier by a two-dimensional model were different from that predicted by a three-dimensional model. Comparative modeling demonstrated that a two-dimensional analysis resulted in underestimation of barrier effectiveness. For the considered design, two-dimensional modeling is a conservative, yet reasonable, approach in a performance assessment application.

    Notes: : We review published analyses of the effects of climate change on goods and services provided by freshwater ecosystems in the United States. Climate-induced changes must be assessed in the context of massive anthropogenic changes in water quantity and quality resulting from altered patterns of land use, water withdrawal, and species invasions; these may dwarf or exacerbate climate-induced changes. Water to meet instream needs is competing with other uses of water, and that competition is likely to be increased by climate change. We review recent predictions of the impacts of climate change on aquatic ecosystems in eight regions of North America. Impacts include warmer temperatures that alter lake mixing regimes and availability of fish habitat; changed magnitude and seasonality of runoff regimes that alter nutrient loading and limit habitat availability at low flow; and loss of prairie pothole wetlands that reduces waterfowl populations. Many of the predicted changes in aquatic ecosystems are a consequence of climatic effects on terrestrial ecosystems; shifts in riparian vegetation and hydrology are particularly critical. We review models that could be used to explore potential effects of climate change on freshwater ecosystems; these include models of instream flow, bioenergetics models, nutrient spiraling models, and models relating riverine food webs to hydrologic regime. We discuss potential ecological risks, benefits, and costs of climate change and identify information needs and model improvements that are required to improve our ability to predict and identify climate change impacts and to evaluate management options.

Benthic macroinvertebrates were sampled for 1 year to assess functional and taxonomic differences in invertebrate biomass and production with respect to habitat types, reaches and catchments in Wine Spring Basin, western North Carolina. Quantitative samples were collected from depositional, cobble-riffle and bedrock outcrop habitats at four stream reaches (two headwater sites, one second order, and one third order). Other measures included physical parameters, periphyton and organic matter standing crops. Invertebrate data from the Wine Spring catchment were also compared with data from another catchment (Ball Creek) within the same region.2. The three habitat types had different current velocities and mean substratum particle sizes; both measures were greatest in bedrock outcrop habitats and lowest in depositional habitats. Organic matter standing crops, invertebrate functional group productivity and biomass also differed significantly with respect to habitat type. Cobble-riffle areas had the lowest standing crops of organic matter, invertebrate productivity and biomass.3. Both invertebrate communities and organic matter standing crops differed significantly between the two headwater reaches. First- to third-order reaches differed in taxonomic composition at the genus level, yet had similar relative functional group productivity and biomass.4. Annual mean invertebrate biomass and secondary production were greater in the Wine Spring Basin than in Ball Creek. Sites in both the Wine Spring and Ball Creek catchments, however, exhibited similar functional group distributions per habitat type.5. Local geomorphology and related physical parameters influenced the structure of invertebrate functional group composition, and the distribution of organic matter standing crops. Furthermore, comparison of community structure in Wine Spring with that in Ball Creek suggested that taxonomic composition was more related to catchment-specific parameters (e.g. thermal regime, evolutionary history) than stream size.
