    Notes: A viral disease of broad bean (Vicia faba) was observed in Jiangsu, Zhejiang and Sichuan Province, the People's Republic of China. Its typical symptom on infected leaves was a white mottle, and, rarely, red necrotic lesions on several broad bean cultivars. A non-aphid-transmissible strain of turnip mosaic virus (TuMV) was identified as the causal agent of the disease on the basis of biological properties, particle morphology, seed and aphid transmission and serological tests. The virus could not be transmitted by aphids in the non-persistent manner.

    Notes: Our previous studies showed that the nonapeptide fragment of human leukocyte antigen DQ with the TPQRGDVYT sequence strongly suppresses the immune response1 [Z. Szewczuk, I. Z. Siemion, and Z. Wieczorek (1996) Molecular Immunology, 33, 903-908]. The fragment contains the RGDVY sequence, which is very similar to thymopentin (pentapeptide RKDVY, an active fragment (32-36) of thymopoietin, an immune system activator produced in thymi), and at the same time contains the RGD sequence, known as an inhibitor of adhesion processes. In the present study we tested an influence of the nonapeptide and its shorter fragments on binding of activated platelets and K562 cells to fibrinogen and fibronectin, respectively. We also designed and synthesized a cyclic thymopentin-like peptide, C*RGDVYC* (where C* indicates Cys participating in disulfide bridge) to restrict its conformation. The cyclization product strongly suppresses the humoral and cellular immune response and selectively inhibits the adhesion of K562 cells to fibronectin. The results are discussed in the light of CD conformational studies. © 1997 John Wiley & Sons, Inc. Biopoly 40: 571-583, 1996

    Description / Table of Contents: Innere Chlorierung von Nickelbasislegierungen and ihre Beziehung zur Korrosion durch VerdampfungDie Kinetik der inneren Chlorierung von Nickelbasislegierungen im Temperaturbereich zwischen 400°C und 700°C wurde mittels optischer und Rasterelektronenmikroskopie untersucht. Die innere Chlorierung von Ni-4Cr und Ni-4Cr-O,5Ce erfolgte durch Bildung fester Ausscheidungen wie CrCl2 und CeCl3, die thermodynamisch stabil sind und bei den Temperaturen niedrige Dampfdrücke in der Legierungsmatrix haben. Im niedrigeren Temperaturbereich folgte die Kinetik der inneren Chlorierung annähernd einem parabolischen Geschwindigkeitsgesetz. Der Grad der inneren Chlorierung nahm mit steigender Temperatur zu. Im höheren Temperaturbereich wurde der innere Angriff durch Legierungsverbrauch infolge Verdampfung begleitet. Wenn die Geschwindigkeit der inneren Chlorierung gleich der Geschwindigkeit des Gesamtmetallverbrauches war, näherte sich die Tiefe der inneren Chlorierungszone einem stationären Wert. Bei hohen Temperaturen, bei denen die Verdampfung dominierte, war die stationäre Tiefe der inneren Chlorierung sogar kleiner als bei niedrigeren Temperaturen. Wenn eine Probe nur geringer Verdampfung ausgesetzt war, zeigte sie mehr inneren Angriff als bei stärkerer Verdampfung und umgekehrt.
    Notes: The kinetics of internal chlorination of Ni-based alloys between 400°C and 700°C were investigated by optical and scanning electron microscopy. Internal chlorination of Ni-4Cr and Ni-4Cr-0.5Ce alloy occurred by formation of solid precipitates such as CrCl2, and CeCl3 which are thermodynamically stable and have low vapor pressures at the temperatures within the alloy matrix. In the lower temperature range, the kinetics of internal chlorination nearly followed a parabolic rate law. The degree of internal chlorination increased with increasing temperature. In the higher temperature range, the internal attack was accompanied by the consumption of the alloy by volatilization. When the rate of internal chlorination equaled the rate of overall metal consumption, the depth of the internal chlorination zone would approach a steady state value. At high temperatures, where vaporization was predominant, the steady state depth of internal attack was even smaller than that at lower temperature. When an alloy specimen was subject to little volatilization, it showed more internal attack than when it was not, and vice versa.

    Notes: Further spectroscopic studies of gallstones are reviewed with an emphasis on the formation of black pigment gallstones. This type of gallstone appears mainly in Western countries, with only 3% of the cholelithiasis patients in China having black gallstones. Fourier transform infrared absorption and electron paramagnetic resonance are used as spectroscopic probes of gallstones and their metal bilirubinate components. Nonlinear phenomena in gallstone formation were investigated through the appearance of ring structure in gallstones and fractal patterns in the formation in the precipitates of bile salt systems. Although a complete understanding of gallstone formation has not yet been achieved, interesting progress toward this goal has been made recently. © 1997 John Wiley & Sons, Inc.     Notes: Random amplified polymorphic DNA (RAPD) markers were generated from 20 wheat, Triticum aestivum lines. Fifty-four fragments generated by six primers of a 10-mer arbitrary sequence were used to study their potential power in differentiating parents with different characteristics and predicting the yield performance of hybrids produced from these parents. Experimental results showed that the 20 wheat lines were divided into four groups. Group I was characterized by more grains per spike, group II by heavy grains and group III by more spikes per unit area and short plants; group IV was similar to group III but had a much higher biomass yield and grain yield. Hybrids from parents in different groups were generally superior to most hybrids from parents in the same group. Both yield performance and heterosis of hybrids from parents between group I and group III were much better than those of other intergroup hybrids. These results suggest that, based on RAPD markers, it is possible to differentiate wheat lines with different performances and that the classification of parents from these markers is of predictive value for developing superior hybrids. However, genetic distance (GD) based on RAPD markers was not significantly correlated with hybrid performance and heterosis. It appears to be impossible to predict hybrid performance from GD itself.

    Notes: Powdery mildew is an important disease in most of the wheat production areas of the world. The resistance gene Pm21 (6AL/6VS trans-location) derived from Haynaldia villosa confers resistance to all available isolates of Erysiphe (Blumeria) graminis f. sp. tritici in China and Europe. The objective of this study was to develop fast and reliable sequence characterized amplified region (SCAR) markers linked to the Pm21 gene. A random amplified polymorphic DNA (RAPD) marker for Pm21, OPH171400, was converted to SCAR markers after sequencing the two ends of the polymorphic DNA fragment. Two SCAR markers, SCAR1265 and SCAR1400, were developed to detect the Pm21 gene in different genetic backgrounds. The specific SCAR1265 marker enable large-scale accurate screening for the presence/absence of Pm21 allele.

    Notes: Adhesion of Candida albicans to host cells is believed to represent a fungal virulence factor and a significant step in the development of candidiasis. As C. albicans strains may differ in their in vitro adhesion ability we initiated a study to investigate whether mutant strains differ in this respect from their parent wild-type. We assessed the in vitro adhesion of C. albicans CBS562 and two mutants obtained by mutagenesis with N′-nitrosoguanidine: a histidine auxotroph, SAG5, derived from CBS562, and a respiratory-deficient strain (a petite mutant), SAR1, derived from SAG5. The adhesion was tested in vitro using two target cell systems: (1) exfoliated human buccal epithelial cells (BEC); and (2) human keratinocyte tissue line cells (HaCaT cells). Adhesion to BEC was evaluated microscopically and that to HaCaT cells by a direct ELISA technique. The results indicated a 54% reduction in adhesion to BEC for SAG5 and 30% for SAR1 as compared to the wild-type, and a 25% reduction in adhesion to HaCaT cells for SAG5 and 20% for SAR1. To verify whether the prototrophy restores the adhesion ability, we complemented the his-negative auxotroph by transforming the strain with the HIS4 gene. Then we assayed the adhesion to BEC of the complemented his-negative mutant in comparison to that of the wild-type, the his-negative mutant (SAG5) and the plasmid-cured transformant. The adhesion values of the complemented his-negative strain were similar to those of the wild-type, whereas the values of the plasmid-cured strain were similar to those of SAG5.
