    Notes: Abstract Root cultures have been established successfully for the Prunus rootstocks Adafuel and Adarcias (Prunus×amygdalopersica), A843 (P. armeniaca), Mariana 2624 (P. cerasifera×munsoniana) and Myrobalan 605AD (P. cerasifera), and for the apple rootstock Jork 9 (Malus×domestica). High percentages of root tips grew during the first 15 days and then decreased. Root growth was affected by culture conditions and the composition of the culture medium. Liquid medium was preferable as increasing agar concentrations reduced root growth. Darkness, instead of a 16-h photoperiod, was beneficial for Adafuel, but not the other genotypes. Sucrose at 3% was better than higher concentrations. Full Murashige and Skoog salts sustained better root growth than dilutions to 1/2 or 1/4. The addition of various organic supplements such as coconut water, casein hydrolysate or malt extract did not improve root growth, and sucrose was the best carbon source tested. Data presented here support the notion that excised root culture is an efficient experimental model to study the response to various factors, since controlled variations in the culture medium, such as those studied here, had a very noticeable effect on root length.

    Notes: Abstract Protein kinase CK2 is characterized by a number of features, including substrate specificity, inhibition by polyanionic compounds and intrasteric down-regulation by its β-subunit, which denote a special aptitude to interact with negatively charged ligands. This situation may reflect the presence in CK2 catalytic subunits of several basic residues that are not conserved in the majority of other protein kinases. Some of these residues, notably K49 in the ‘Gly rich loop’, K74, K75, K76, K77, K79, R80, K83 in the ‘Lys rich segment’ and R191, R195, K198 in the ‘p+1 loop’, have been shown by mutational studies to be implicated to various extents and with distinct roles in substrate recognition, inhibition by heparin and by pseudosubstrate and instrasteric regulation. Molecular modelization based on crystallographic data provide a rationale for the biochemical observations, showing that several of these basic residues are clustered around the active site where they make contact with individual acidic residues of the peptide substrate. They can also mediate the effect of polyanionic inhibitors (e.g. heparin) and of regulatory elements present in the b-subunit, in the N terminal segment of the catalytic subunit and possibly in other proteins interacting with CK2. Our data also disclose a unique mode of binding of the phosphoacceptor substrate which bridges across the catalytic cleft making contacts with both the lower and upper lobes of CK2.

    Notes: Summary Individual polystyrene (PS) and polyacrylonitrile (PAN) solutions in N,N-dimethylformamide (DMF) were studied by electronic absorption spectroscopy. Upon increasing the polymer concentration in the PS-DMF binary system a bathochromic shift of the band at about 260 nm was found. This spectral effect could be explained by an association of PS phenyl groups and/or by a complex formation between DMF and PS. The UV spectrum of PAN in DMF solution was interpreted in terms of the variety of possible donor-acceptor complex formations. The specific intermolecular interactions in the investigated binary liquid systems were discussed on the basis of the spectral data.

    Notes: Abstract The structure and ordering of nanoparticles of gold functionalized with n-alkyl thiol molecules are studied both experimentally and theoretically. Samples where produced using n=6 to n=16 alkyl thiol molecules. High Resolution Electron Microscopy coupled with image processing was used to study the gold particle structure. The details of the particle structure are discussed. We found that when the gold surface is saturated with thiol molecules there is some tendency to produce molecules with a disulphide structure. We also show that ordered arrays of particles can be produced using Langmuir-Blodgett techniques. The crystal structure of the films produced is studied and found to be 3D hcp. We also report that thiol covered gold particles with a size of ∼5 nm present a rounded shape suggesting that thiol molecules might induce an isotropic surface energy. It is found in the theoretical calculations that a strong bond between gold and sulphur is required to stabilize the complex metal-n-alkyl thiol. It is predicted that otherwise a heavily distorted nanocore will be formed. This is contrary to the observed structure of the particles.

    Notes: Abstract With a view to using bile acids as shuttles for delivering platinum-related cytostatic drugs to liver tumors, a chenodeoxycholylglycinato(CDCG)-derivative of platinum(II) has been synthesized. The complex - named Bamet-M2- was chemically characterized by elemental analysis, FT-IR, NMR, FAB-MS, and UV spectroscopy. The results indicate the following composition: C26H42N2O5Cl2NaPt(II), the metal Pt(II) being bound to two Cl− and one bidentate CDCG moiety, i.e., Na[Pt CDCG(N,O) Cl2]. The compound is highly soluble (up to 20 mM) in water and (up to 35 mM) in ethanol, methanol and DMSO. Hydrolysis was investigated because this is assumed to be an important step in intracellular activation and interaction with DNA of this type of compounds. The reaction kinetics of this complex in aqueous solution show unusual behaviour; the substitution process with the displacement of two Cl− was almost instantaneous, and the resulting species were found to be very stable. Kinetic studies carried out in the presence of different NaCl concentrations (up to 500 mM) revealed similar fast and nonreversible aquations of Bamet-M2. This contrasts with the slow aquation of cisplatin in extracellular-line solutions (i.e., at high NaCl concentrations) as compared with fast hydrolysis in cells. This difference may partly account for the low cytostatic activity observed here for Bamet-M2 against several tumor cell-lines.

    Notes: Abstract The partial oxidation of methane to synthesis gas has been studied in a continuous flow reactor using a Rh/α-Al2O3 catalyst under conditions as close as possible to those industrially relevant: pressures up to 800 kPa and temperatures higher than 1274 K in order to avoid the formation of carbon and to obtain high equilibrium selectivities to CO and H2. Intrinsic kinetic data were obtained when the feed was diluted with helium. Gas-phase reactions were found to occur at 500 kPa when the feed was not diluted. A reaction network has been derived from experimental results in which oxygen conversions range from 0 to 1. CO2, C2H6 and H2O are the primary products. C2H4 is formed by oxidative dehydrogenation of C2H6. CO and H2 are formed by reforming of CH4 by CO2 and H2O; an additional direct route to CO and H2 at low oxygen conversions cannot be excluded. The catalyst appears to be present in two states, the transition being at an oxygen conversion of 0.4 under the conditions used. The support probably enhances oxidation reactions by reverse spillover of oxygen or hydroxyl species onto rhodium. The support as such behaves similarly to the catalyst at low oxygen conversions, but shows no reforming activity.

    Notes: Abstract The effect of the platinum particle diameter and the catalyst support was investigated for the aqueous phase selective oxidation of methyl α-D-glucopyranoside, octyl α-D-glucopyranoside and α-cyclodextrin with molecular oxygen. No platinum particle size effect was observed in the platinum particle diameter range of 1.4 to 3.0 nm. The structure-insensitivity is attributed to the high degree of surface coverage by oxygen. The rate of deactivation due to over-oxidation of the catalyst increases to a small extent with decreasing platinum particle diameter. The effect of the catalyst support is much larger than the platinum particle size effect. The turnover frequency for platinum on activated carbon and carbon fibrils supported catalyst is 3-7 times higher than for graphite as the catalyst support. The support effect is attributed to the hydrophilic character of the support.
