Summary For the evaluation of paper chromatograms by measuring the reflected light some conditions must be fulfilled: 1. By use of modified “Matthias wedge-strips” band-shaped spots can be obtained. The filter paper shall be dense and the surface must be as smooth as possible. 2. The distance moved by the substances for calibration and that of the sample must be of the same length. The composition of the solvent mixture used is of minor importance. 3. The volume of the spotted substances must be the same for the samples and for the calibration. The concentration of the solutions should not be too low, so that the substance is equally spread over the whole spot. 4. Accompanying ions, which have similar Rf values, cause interferences also if they do not react with the developing reagents. 5. To diminish the errors only those strips should be used of which the rims have been subjected to the least possible mechanical stress on cutting.

Zusammenfassung Damit ein Papierchromatogramm nach dem Auflichtverfahren direkt ausgewertet werden kann, muß eine Reihe von Bedingungen erfüllt sein. 1. Durch Anwendung von modifizierten „Keilstreifen nach Matthias“ lassen sich bandenförmige Substanzflecken erhalten. Das hierzu benutzte Papier soll hart sein und eine möglichst glatte Oberfläche besitzen. 2. Die Substanzflecken auf den Chromatogrammen der Eichreihe und auf den zu untersuchenden Streifen müssen gleiche Strecken gelaufen sein. Dabei spielen die dazu verwendeten Fließmittel eine untergeordnete Rolle. 3. Das Volumen der aufgetragenen Substanzen muß bei Eich- und Meßreihe gleich groß sein. Eine Mindestkonzentration ist einzuhalten, damit die Startflecken gleichmäßig mit Substanz getränkt sind. 4. Begleitstoffe, die ähnliche Rf-Werte haben wie der zu prüfende Stoff, stören, auch wenn sie mit der Entwicklerlösung nicht reagieren. 5. Zur Verringerung der Meßfehler sollen Streifen verwendet werden, deren Ränder beim Zuschnitt möglichst wenig mechanisch beansprucht worden sind.
