    Notes: Metabolism is the conversion of available energy sources to those energy forms required for sustaining and propagating living organisms; this is simply biological energy conversion. Proteins are the machines of metabolism; they are the engines of motility and the other machines that interconvert energy forms not involving motion. Accordingly, metabolic engineering becomes the use of natural protein-based machines for the good of society. In addition, metabolic engineering can utilize the principles, whereby proteins function, to design new protein-based machines to fulfill roles for society that proteins have never been called upon throughout evolution to fulfill.This article presents arguments for a universal mechanism whereby proteins perform their diverse energy conversions; it begins with background information, and then asserts a set of five axioms for protein folding, assembly, and function and for protein engineering. The key process is the hydrophobic folding and assembly transition exhibited by properly balanced amphiphilic protein sequences. The fundamental molecular process is the competition for hydration between hydrophobic and polar, e.g., charged, residues. This competition determines Tt, the onset temperature for the hydrophobic folding and assembly transition, Nhh, the numbers of waters of hydrophobic hydration, and the pKa of ionizable functions.Reported acid-base titrations and pH dependence of microwave dielectric relaxation data simultaneously demonstrate the interdependence of Tt, Nhh and the pKa using a series of microbially prepared protein-based poly(30mers) with one glutamic acid residue per 30mer and with an increasing number of more hydrophobic phenylalanine residues replacing valine residues. Also, reduction of nicotinamides and flavins is shown to lower Tt, i.e., to increase hydrophobicity.Furthermore, the argument is presented, and related to an extended Henderson-Hasselbalch equation, wherein reduction of nicotinamides represents an increase in hydrophobicity and resulting hydrophobic-induced pKa shifts become the basis for understanding a primary energy conversion (proton transport) process of mitochondria. © 1998 John Wiley & Sons, Inc.     Notes: The suitability of hornblende as a support for immobilized β-fructofuranosidase (invertase) was studied, with regard to the physical stability of the support and the thermal and operational stability of the immobilized enzyme. Hornblende was more stable than Enzacryl-Alo or Enzacryl-TIO, and marginally more stable than porous glass. Invertase immobilized on hornblende was more stable during long-term operation than invertase immobilized on porous glass. An active preparation of immobilized invertase was obtained also on pyroxene particles.

    Notes: Results of pilot plant studies using a glass airlift fermentation device (55 liter fermentation volume) have proven the relative merits of such a system in the fermentation of a filamentous mold, Monascus purpureus, on 4% (w/w) starch media. The resultant overall yield of cell mass (Yx/s) of 0.38 was an appreciable increase over the 0.32 obtained with a pilot scale stirred tank fermentor previously studied. Power requirements of the airlift fermentor were approximately 50% of those for the mechanically agitated system. The lack of mechanical shear in the airlift system provides a more gentle environment or the cultivation of organisms than does the high degree of shear prevalent in the mechanically agitated vessels. Mass transfer of oxygen to the aqueous phase of the fermentation volume is improved significantly through use of the airlift device. Mass transfer coefficients in the range of 200 reciprocal hr were obtained to approximately 80 reciprocal hr in the stirred tank fermentor.

    Notes: The Membrane Bioreactor for Biotransformations (MBB) is based on the aqueous/organic two-phase system, and uses a tubular silicone rubber membrane to separate the two liquid phases. This avoids the key problem associated with direct contact two-phase processes, specifically, product emulsification. The baker's yeast mediated reduction of geraniol to citronellol was used as a model biotransformation to demonstrate MBB operation. Values for the overall mass transfer coefficient were determined for geraniol, (2.0 × 10-5 ms-1), and for citronellol, (2.1 × 10-5 ms-1) diffusion across the silicone rubber membrane. Using these values, and the specific activity of the biocatalyst (5 nmols-1g biomass-1), a suitable membrane surface area: biomass ratio was determined as 2.4 × 10-3 m2g biomass-1. The bioreactor was operated at this surface area: biomass ratio and achieved a product accumulation rate 90-95% that of a conventional direct contact two-phase system. The slight reduction in product accumulation rate was shown not to be due to mass transfer limitations with respect to reactant delivery or product extraction. © 1998 John Wiley & Sons, Inc.     Notes: A method for the control of dissolved oxygen tension in growing microbial cultures is described. The apparatus consists of a motor-driven air sparge pipe which may be lowered or raised to give a variable point of entry of the air stream into the culture liquid and hence a variable gas dispersion and gas-liquid contact time. Control of the sparge pipe position is by means of a feedback control loop consisting of a dissolved oxygen probe, an on/off controller, and a reversing electric motor which drives the sparge pipe. The difficulty presented by the relatively slow response of the oxygen probe has been overcome by incorporating an adjustable rate of control action.

    Notes: The intracellular levels of seven enzymes in mouse LS cells growing in suspension culture at controlled dissolved oxygen partial pressures (pO2) have been measured. During the growth of each culture large fluctuations were observed in the levels of some enzymes, particularly aldolase and cytochrome oxidase. Mean values for the concentration of each enzyme during the growth phase have been calculated. These results are discussed in relation to previous observations made on the growth of mouse LS cells.

    Notes: The presence of microorganisms have been shown to increase by over 40% the mass transfer rates from small oxygen bubbles at low Reynolds number flow. This increase was found to be due only to the microbe cells as inert particles disrupting the quasi-static liquid surface film surrounding the gas bubble and thus decreasing the gas-liquid interfacial resistance. The observed increase in oxygen mass transfer rates was not dependent on cell viability, no effect was noted due to cell-liquid interfacial resistance, nor was the phenomenon due to altering the physical properties of the liquid during cell propagation. These results were obtained in a unique plexiglass apparatus designed for observing under a microscope a small (0.4 mm dia.) stationary oxygen bubble collapsing into a flowing fluid. The oxygen bubble was injected by a small hypodermic needle and the fluid was suspensions of the yeast Candida intermedia, the bacterium Pseudomonas ovalis, 0.3μ alumina, as well as base points of cell free broth and pure water. Several well-known chemical inhibitors of oxidative phosphorylation were used to limit cell oxygen uptake. Calculations of oxygen mass transfer rates were compared with the semi-empirical model of Frössling, the circulating sphere model of Levich, and the rigid sphere concentration boundary layer model of Fried-lander, the latter two showing strong Reynolds number dependence that may be due to radial fluid motion.

    Notes: Batch cultures of Trichoderma viride have been carried out in a 10 liter stirred fermenter at controlled pH values of 2.5, 2.7, 3.0, and 4.0, and without pH control at a temperature of 28°C. Cell and glucose concentrations and dissolved oxygen values are reported. The yield coefficient was found to be constant at 0.40 kg cells/kg glucose and the maximum specific growth rate was linearly correlated with the hydrogen ion concentration.
