    Notes: Abstract The crystal and molecular structure of a series of 3,7-bis(arylazo)-1,3,5,7-tetraazabicyclo[3.3.1] nonanes (1a–d) have been determined by single crystal X-ray diffraction analysis. All four compounds have the same feature, that the tetraazabicyclononane unit assumes a cage-like structure with the aryltriazene moieties lying parallel to one another so that the aryl rings are held together by π – π stacking.     Notes: Summary The effects of endotoxin on mouse liver phospholipid fatty acid composition have been investigated. Administration of endotoxin fromSalmonella abortus equi led to a decrease in the polyunsaturated fatty acid content of livers from mice sensitized with Bacille Calmette Guérin (GCG). The content of arachidonic acid fell significantly in both the phosphatidylcholine and phosphatidylinositol fractions whereas in the phosphatidylethanolamine fraction the linoleic acid content was significantly reduced. The polyunsaturated fatty acids were replaced by increased amounts of oleic acid and palmitic acid, leading to a reduction in the polyunsaturated to saturated fatty acid ratio.

    Notes: Abstract We previously reported that the presence of the bacterial (Vitreoscilla) hemoglobin gene enhances alpha-amylase production in recombinant Escherichia coli strain MK79. Using the growth of MK79 on starch as a selective method we have produced a mutant strain (BSC9) that produces up to four times as much alpha-amylase as MK79. Both MK79 and BSC9 produce the most alpha-amylase (per cell and per milliliter) in the stationary phase; almost all of the enzyme is intracellular in both strains. Modification of the standard alpha-amylase assay increases the amount of amylase detected about sixfold. BSC9 has about five to nine times as many copies per cell as MK79 of the recombinant plasmid, which carries both the amylase and hemoglobin genes, but both strains produce about the same amount of hemoglobin. While MK79 respiration decreases upon going from log to stationary phase, BSC9 respiration increases during the same period. The two latter results may be of particular importance in determining the way in which hemoglobin enhances the production of cloned protein products in recombinant bacteria.

    Notes: Abstract. During recent years parathyroid hormone-related protein (PTHrP) research has been focused on the physiological functions of different fragments of the PTHrP molecule. Here we demonstrate that PTHrP (1–37) induced cyclic adenosine monophosphate (cAMP) response in primary human osteoblast-like cells, which were well characterized by the presence of alkaline phosphatase activity and osteocalcin production after stimulation with 1,25-dihydroxyvitamin D3. However, there was no cAMP response to PTHrP (58–77). Furthermore, the response to PTHrP (1–37) was dose dependent, with a significant increase at 1 nM. The presence of PTHrP (1–37)-induced cAMP response in human osteoblast-like cells implies that aminoterminal PTHrP fragments may exert important functions in the bone.

    Notes: Abstract Phylogenetic and physiological methods were used to study the evolution of the opsin gene family in Drosophila. A phylogeny based on DNA sequences from 13 opsin genes including representatives from the two major subgenera of Drosophila shows six major, well-supported clades: The “blue opsin” clade includes all of the Rhl and Rh2 genes and is separated into two distinct subclades of Rhl sequences and Rh2 sequences; the ultraviolet opsin clade includes all Rh3 and Rh4 genes and bifurcates into separate Rh3 and Rh4 clades. The duplications that generated this gene family most likely took place before the evolution of the subgenera Drosophila and Sophophora and their component species groups. Numerous changes have occurred in these genes since the duplications, including the loss and/or gain of introns in the different genes and even within the Rhl and Rh4 clades. Despite these changes, the spectral sensitivity of each of the opsins has remained remarkably fixed in a sample of four species representing two species groups in each of the two subgenera. All of the strains that were investigated had R1-6 (Rhl) spectral sensitivity curves that peaked at or near 480 nm, R7 (Rh3 and Rh4) peaks in the ultraviolet range, and ocellar (Rh2) peaks near 420 nm. Each of the four gene clades on the phylogeny exhibits very conservative patterns of amino acid replacement in domains of the protein thought to influence spectral sen sitivity, reflecting strong constraints on the spectrum of light visible to Drosophila.

    Notes: Summary Ten seedlings each of Eucalyptus kitsoniana Maiden and Eucalyptus globulus Labill. were subjected to two levels of water stress and two levels of nutrient stress (macro and micro-nutrients) in a greenhouse for 3 weeks. The objectives were to determine the degree to which seedlings show differences in sap chemistry, photosynthesis and transpiration that relate to the environments in which these two species live. Whole plants were then extracted for xylem sap using a pressure chamber and the sap was analyzed for 14 elements using an inductively coupled plasma spectrometer and a nitrometer. For E. kitsoniana water and nutrient stress, applied separately or in combination, significantly reduced leaf conductance, transpiration, photosynthesis and midday water potential. Nutrient stress alone had less effect than water stress on most functions measured. Water stress alone reduced the root/shoot ratio; the combination of water and nutrient stress increased the root/shoot ratio, primarily because of reduced shoot weight. In E. kitsoniana, water stress alone or in combination with nutrient stress increased the xylem sap concentrations of B and Si. Multi-nutrient stress alone, or in combination with water stress, significantly decreased sap Zn and K. For this species, sap N was decreased by nutrient stress, but increased by water stress. E. globulus had significantly lower transpiration rates and less root mass than E. kitsoniana. Slightly lower leaf conductance and photosynthesis were not significant in E. globulus compared to E. kitsoniana. Water and nutrient stress reduced conductance, transpiration (except for nutrient stress) and photosynthesis, and the effects of water stress on E. globulus were greater than the effects of nutrient stress. Midday water potential was reduced by water stress. Water or nutrient stress alone did not alter seedling root/shoot ratio, but the combination of water and nutrient stress significantly increased the root/shoot ratio for both species. For E. globulus, sap concentrations of Mn, Na, Si and K were increased by water stress (alone or in combination with nutrient stress). Sap N increased with water stress or combined stresses, but decreased under nutrient stress alone. When the two species were compared, E. globulus generally had lower or similar nutrient concentrations in the sap, with Ca, Mg, Mn and P significantly lower than in E. kitsoniana. Seedlings of these two species show strong site adaptations to water and nutrient availability.
