    Notes: Abstract: Internal ribosome entry sites (IRESs) allow ribosomal access to messenger RNA without a requirement for cap recognition and subsequent scanning to an initiator AUG. Hence, IRESs have been adapted into dicistronic vectors for the expression of more than one gene from a single mRNA. Dicistronic vectors have been used for many applications in mammalian tissue culture and transgenesis. However, whether the IRESs from mammalian viruses function without temporal or spatial restrictions in nonmammalian organisms like zebra fish (Danio rerio) is unknown. Therefore, we have examined the expression capabilities of the encephalomyocarditis virus (EMCV) IRES during zebrafish embryogenesis. We determined that the EMCV IRES was sufficient to permit detectable expression of several second cistron reporters during zebrafish embryogenesis, including luciferase and green fluorescent protein. This suggests that our dicistronic vectors are suitable for general use in any vertebrate system, from fish to humans.

    Notes: Abstract Absolute surface coverages (θ) of a series of tungsten-alumina catalysts prepared by the equilibrium adsorption method were determined by ion scattering spectroscopy (ISS) and low-temperature CO adsorption. The θCO values for the W catalysts were very similar to those obtained with the ISS method, for a given W loading. A linear increase in W coverage with increasing W loading, reaching ~55% coverage for the 13.1% W solid, was observed. The calculated W cross-sections (22.0- 24.5Å2/W) were in agreement with other reports in the literature. Due to the inherent limitations of each method, the combined use of ISS and low-temperature CO adsorption is recommended for estimating absolute surface coverages in tungsten-alumina catalysts.

    Notes: Abstract The acrobot is an underactuated two-link planar robot that mimics the human acrobat who hangs from a bar and tries to swing up to a perfectly balanced upside-down position with his/her hands still on the bar. In this paper we develop intelligent controllers for swing-up and balancing of the acrobot. In particular, we first develop classical, fuzzy, and adaptive fuzzy controllers to balance the acrobot in its inverted unstable equilibrium region. Next, a proportional-derivative (PD) controller with inner-loop partial feedback linearization, a state-feedback, and a fuzzy controller are developed to swing up the acrobot from its stable equilibrium position to the inverted region, where we use a balancing controller to ‘catch’ and balance it. At the same time, we develop two genetic algorithms for tuning the balancing and swing-up controllers, and show how these can be used to help optimize the performance of the controllers. Overall, this paper provides (i) a case study of the development of a variety of intelligent controllers for a challenging application, (ii) a comparative analysis of intelligent vs. conventional control methods (including the linear quadratic regulator and feedback linearization) for this application, and (iii) a case study of the development of genetic algorithms for off-line computer-aided-design of both conventional and intelligent control systems.

    Notes: Abstract We have evaluated the structure of the CGG repeat within the FMR1 gene of an Asian population and found the most common size of the repeat to be 29 and 30 with a minor population of 36 repeats. We have isolated and sequenced DNA containing the 36 repeats and found the basis sequence to be (CGG)9AGG(CGG)9AGG(CGG)6AGG(CGG)9; with a (CGG)6)AGG insertion, designated as 9A9A6A9. Of 144 Asian chromosomes, 11 (8%) had sequences with this insertion. Six different variations of the basic sequence were observed in the population: 9A9A6A2A9, 9A9A6A11, 9A9A16, 9A9A15, 8A9A6A6A9, and 11A6A6A9. All but one of the chromosomes with the insertion had the haplotype of DXS548/ FRAXAC1: 194/D suggesting that the sequences with the 6A insertion arose from a single ancestral allele. We have not observed the insertion in the FMR1 gene of Caucasians or Native Americans. The (CGG)6AGG insertion may be unique to Asians.

    Notes: Abstract Membrane diffusion potentials induced by amphotericin B (AmB), amphotericin B methyl ester (AmE), N-fructosyl AmB (N FruAmB) and vacidin, an aromatic polyene antibiotic, in ergosterol- or cholesterol-containing egg yolk phosphatidylcholine large unilamellar vesicles (LUV), were measured in various media, in order to determine the relative selectivity of Na+, K+, Cl− and other ions in these environments. Changes in the membrane potential were followed by fluorescence changes of 3,3′-dipropylthiadicarbocyanine (diS-C3-(5)). Subtle changes in intercationic selectivity were monitored by measuring biionic potentials, using the fluorescent pH sensitive probe pyranine. In all the cases studied, the intereationic selectivity of the permeability pathways induced by the four antibiotics was weak compared to that of specific biological channels, though distinct differences were noted. With AmB the selectivity appeared to be concentration dependent. Above 5 × 10−7 M, the sequence determined for sterol-free small unilamellar vesicles (SUV) and cholesterol-containing SUV and LUV, Na+ 〉 K+ 〉 Rb+ ≥ Cs+ 〉 Li+ (sulfate salts), corresponded closely to Eisenman selectivity sequence number VII. At 5 × 10−7 M and below the selectivity switched from Na+ 〉 K+ to K+ 〉 Na +. In contrast, Li+ was the most permeant ion for AmB channels in the presence of ergosterol. The selectivity between Na+ or K+ vs. Cl− varied with the antibiotic. It was very strong with vacidin at concentrations below 5 × 10−7 M, smaller with AmB, nil with AmE and N FruAmB. The selectivities observed were antibiotic, concentration and time de pendent, which confirms the existence of different types of channels.

    Notes: Summary Hemophilia B is due to multiple molecular defects in the factor IX gene. Over 80% of mutations are single base substitutions. By amplification and direct sequencing, 51 single base substitutions were found in the transcribed sequence of the factor IX genes of patients from 50 distinct families with hemophilia B. These include 30 mutations in 29 families not previously reported by us; of these, 12 are novel, i.e., not previously published in other series. Of the 51 substitutions in our overall series 23 (45%) occurred as C-to-T or G-to-A transitions at 11 sites within CG dinucleotides. It is estimated that CG transitions occur from one to two orders of magnitude more frequently than mutations in nucleotides that are not within a CG pair. More than one family had identical defects for 6 of the CG mutations. At 4 of these sites, most patients had different haplotypes compatible with distinct mutations. Non-CG-type mutations occurred thoughout the coding regions with only one mutation in more than one family. The latter included 7 families with a 397 Ile-to-Thr defect that all share a rare haplotype, suggesting a common ancestor.
