    Description: BACKGROUND: Lifestyle factors are known to affect skeletal development and integrity. Specifically, running has been reported to increase risk of fatigue fractures, whereas chronic alcohol consumption has been shown to reduce bone formation and bone mass. The combined effect of exercise and alcohol on the skeleton has yet to be explored, although alcohol consumption is common among certain physically active populations (e.g., military recruits, college athletes). It was hypothesized that chronic alcohol consumption would accentuate the inherent risk associated with endurance running exercise. METHODS: Six-month-old male Sprague Dawley rats were assigned to one of five groups: baseline, exercise-alcohol diet, exercise-normal diet, sham-alcohol diet, and sham-normal diet. Alcohol-fed rats (35% caloric intake) received a liquid diet ad libitum. Normal animals were pair-fed the identical diet with a maltose dextrin caloric substitute. Exercise was conducted on a motorized treadmill 5 days/wk for 16 weeks. Sham rats were placed on a stationary treadmill for matching time periods. Fluorochrome labels were administered 3 days before baseline and at 10 and 2 days before animals were killed. Heart, soleus, and rectus femoris muscles were wet weighed to assess the effects of training. Tibiae were collected for static and dynamic histomorphometric measurements on cancellous and cortical bone. RESULTS: Muscle weights were larger in the exercised rats versus the sham rats. Alcohol had no significant effect on skeletal muscle weight but did result in larger heart weights in both alcohol-treated groups. Cancellous and periosteal bone formation rates were significantly decreased in the alcohol-fed rats versus rats on the normal diet and were associated with a significant reduction in trabecular thickness in the tibial metaphysis. Cortical and cross-sectional areas were also significantly lower in the alcohol-fed groups compared with the non-alcohol-fed groups. Exercise had no significant effect on cancellous or cortical bone measurements. CONCLUSIONS: Chronic alcohol consumption significantly reduced bone formation. Exercise had no effect on the bone and did not attenuate any of the negative effects of alcohol. The results suggest that alcohol consumption weakens the skeleton and increases the incidence of endurance-exercise-related bone injuries. Thus, individuals who are participating in endurance exercise and consuming alcohol may be at greater risk for exercise-related skeletal injuries. Further investigation of the potential for alcohol to induce detrimental effects on the hearts of individuals participating in endurance exercise is indicated.

    Description: OBJECTIVE: To evaluate a new system, ISAID (Internet-based Semi-automated Indexing of Documents), and to generate textbook indexes that are more detailed and more useful to readers. DESIGN: Pilot evaluation: simple, nonrandomized trial comparing ISAID with manual indexing methods. Methods evaluation: randomized, cross-over trial comparing three versions of ISAID and usability survey. PARTICIPANTS: Pilot evaluation: two physicians. Methods evaluation: twelve physicians, each of whom used three different versions of the system for a total of 36 indexing sessions. MEASUREMENTS: Total index term tuples generated per document per minute (TPM), with and without adjustment for concordance with other subjects; inter-indexer consistency; ratings of the usability of the ISAID indexing system. RESULTS: Compared with manual methods, ISAID decreased indexing times greatly. Using three versions of ISAID, inter-indexer consistency ranged from 15% to 65% with a mean of 41%, 31%, and 40% for each of three documents. Subjects using the full version of ISAID were faster (average TPM: 5.6) and had higher rates of concordant index generation. There were substantial learning effects, despite our use of a training/run-in phase. Subjects using the full version of ISAID were much faster by the third indexing session (average TPM: 9.1). There was a statistically significant increase in three-subject concordant indexing rate using the full version of ISAID during the second indexing session (p 〈 0.05). SUMMARY: Users of the ISAID indexing system create complex, precise, and accurate indexing for full-text documents much faster than users of manual methods. Furthermore, the natural language processing methods that ISAID uses to suggest indexes contributes substantially to increased indexing speed and accuracy.

    Description: Neuromodulation denotes controlled electrical stimulation of the central or peripheral nervous system. The three forms of neuromodulation described in this paper-deep brain stimulation, vagus nerve stimulation, and transcranial magnetic stimulation-were chosen primarily for their demonstrated or potential clinical usefulness. Deep brain stimulation is a completely implanted technique for improving movement disorders, such as Parkinson's disease, by very focal electrical stimulation of the brain-a technique that employs well-established hardware (electrode and pulse generator/battery). Vagus nerve stimulation is similar to deep brain stimulation in being well-established (for the treatment of refractory epilepsy), completely implanted, and having hardware that can be considered standard at the present time. Vagus nerve stimulation differs from deep brain stimulation, however, in that afferent stimulation of the vagus nerve results in diffuse effects on many regions throughout the brain. Although use of deep brain stimulation for applications beyond movement disorders will no doubt involve placing the stimulating electrode(s) in regions other than the thalamus, subthalamus, or globus pallidus, the use of vagus nerve stimulation for applications beyond epilepsy-for example, depression and eating disorders-is unlikely to require altering the hardware significantly (although stimulation protocols may differ). Transcranial magnetic stimulation is an example of an external or non-implanted, intermittent (at least given the current state of the hardware) stimulation technique, the clinical value of which for neuromodulation and neuroprotection remains to be determined.

    Description: Three examples of neuroprotective applications of electrical stimulation-neuromodulation-are considered: (1) the diagnosis and treatment of epilepsy, (2) the augmentation of peripheral nerve regeneration after transection, and (3) the interaction between electrical stimulation and neurotrophins (notably brain derived neurotrophic factor [BDNF]) in various neuroprotective situations. The research cited demonstrates clear benefit from appropriate electrical stimulation in the treatment of (1) certain patients with medication-refractory epilepsy, and (2) the functional regeneration of peripheral nerves after transection and surgical repair. Furthermore, neuromodulation of peripheral nerve regeneration has been associated with an increase in the neurotrophin BDNF. The roles of BDNF and other neurotrophins in several disorders of the nervous system are discussed in the context of neuromodulation and its augmentation of neurotrophins. Neuromodulation-at least in part through its effect on BDNF and other neurotrophins-will likely play a major role in the treatment (and possibly prevention) of disorders of the nervous system for which neuroproteive pharmacologic agents have traditionally been sought.

    Description: During the ACE Asia intensive field campaign conducted in the spring of 2001 aerosol properties were measured onboard the R/V Ronald H. Brown to study the effects of the Asian aerosol on atmospheric chemistry and climate in downwind regions. Aerosol properties measured in the marine boundary layer included chemical composition; number size distribution; and light scattering, hemispheric backscattering, and absorption coefficients. In addition, optical depth and vertical profiles of aerosol 180 deg backscatter were measured. Aerosol within the ACE Asia study region was found to be a complex mixture resulting from marine, pollution, volcanic, and dust sources. Presented here as a function of air mass source region are the mass fractions of the dominant aerosol chemical components, the fraction of the scattering measured at the surface due to each component, mass scattering efficiencies of the individual components, aerosol scattering and absorption coefficients, single scattering albedo, Angstrom exponents, optical depth, and vertical profiles of aerosol extinction. All results except aerosol optical depth and the vertical profiles of aerosol extinction are reported at a relative humidity of 55 +/- 5%. An over-determined data set was collected so that measured and calculated aerosol properties could be compared, internal consistency in the data set could be assessed, and sources of uncertainty could be identified. By taking into account non-sphericity of the dust aerosol, calculated and measured aerosol mass and scattering coefficients agreed within overall experimental uncertainties. Differences between measured and calculated aerosol absorption coefficients were not within reasonable uncertainty limits, however, and may indicate the inability of Mie theory and the assumption of internally mixed homogeneous spheres to predict absorption by the ACE Asia aerosol. Mass scattering efficiencies of non-sea salt sulfate aerosol, sea salt, submicron particulate organic matter, and dust found for the ACE Asia aerosol are comparable to values estimated for ACE 1, Aerosols99, and INDOEX. Unique to the ACE Asia aerosol was the large mass fractions of dust, the dominance of dust in controlling the aerosol optical properties, and the interaction of dust with soot aerosol.

    Description: Abstract A 1999 study reports an advancement of spring in Europe by 0.2 days per year in the 30 years since 1960. Our analysis indicates that this trend results directly from a change in the late-winter surface winds over the eastern North Atlantic: the southwesterly direction became more dominant, and the speed of these southwesterlies increased slightly. Splitting the 52-year NCEP reanalysis dataset into the First Half, FH (1948-1973)), and the Second Half, SH (1974-1999), we analyze the wind direction for the February mean at three sites at 45N: site A at 30W, site B at 20W, and site C at 10W. The incidence (number of years) of the southwesterlies in SH Vs. (FH) at these sites respectively increased in SH as follows: 24(18), 19(12), 14(l 1); whereas the incidence of northeasterlies decreased: 0(2), 1(2), and 1(6). When the February mean wind is southwesterly, the monthly mean sensible heat flux from the ocean at these sites takes zero or slightly negative values, that is, the surface air is warmer than the ocean. Analyzing the scenario in the warm late winter 1990, we observe that the sensible heat flux from the ocean surface in February 1990 shows a "tongue" of negative values extending southwest from southern England to 7N. This indicates that the source of the maritime air advected into Europe lies to the south of the "tongue." Streamline analysis suggests that the Southwestern or southcentral North Atlantic is the source. For February 1990, we find strong, ascending motions over Europe at 700 mb, up to -0.4 Pa/s as monthly averages. Associated with the unstable low-levels of the troposphere are positive rain and cloud anomalies. Thus, positive in situ feedback over land in late winter (when shortwave absorption is not significant) apparently further enhances the surface temperature through an increase in the greenhouse effect due to increased water vapor and cloudiness.

    Description: Pollution is often considered a localized phenomenon, but it is now clear that it travels from region-to-region, country to country, and even continent to continent. In addition to urban pollution in developed countries, large emissions from developing nations and large-scale biomass fires add to the global pollution burden. Ozone and aerosols are two components of pollution that contribute to radiative forcing of the earth s climate. In turn, as climate changes, rates of chemical and microphysical reactions may be perturbed. Considering the earth as a coupled chemical-microphysical-climate system poses challenges for models and observations alike. These issues were the topic of a Workshop held in May 2002 at NASA GSFC s Laboratory for Atmospheres. Highlights of the Workshop are summarized in this article.

    Description: We examine a possible mechanism leading to late-winter warming and thus to an early spring in Europe. From the NCEP Reanalysis, we extract for the years 1948-1999 ocean-surface winds over the eastern North Atlantic, and air temperatures at the surface, T(sub s), and at the 500 mb level, T(sub 500), in late-winter and spring. T(sub s) is extracted at six European locations, all at 50.5 N, ranging in longitude from 1.9 E (northeastern France) to 26.2 E (Ukraine). To quantify the advection of maritime air into Europe, we evaluate for 3-pentad groups the Index I(sub na) of the southwesterlies at 45 N; 20 W: I(sub na) is the average wind speed at this point if the direction is from the quadrant 180-270 deg (when the direction is different, the contribution counts as zero). In late winter correlations C(sub it) between the Index I(sub na) and the temperature T(sub s) are substantial, up to the 0.6 level, in western Europe (but weaker correlations for Poland and Ukraine). C(sub it) drops sharply by mid-March, taking occasionally negative values subsequently. This drop in C(sub it) indicates that maritime air advection is no longer associated closely with the surface-air warming, the role of immolation becomes important, and thus the drop in C(sub it) marks the arrival of spring. Correlations C(sub i delta) between I(sub na) and our lapse-rate parameter delta, the difference between T(sub s) and T(sub 500), indicate that the flow of warm maritime-air from the North Atlantic into this 'corridor' at 50.5 N is predominantly at low tropospheric level. By computing the best linear fit to I(sub na) and T(sub s), the trends for the period 1948-1999 are evaluated. The trends are appreciable in the second half of February and the first half of March. Our 3-pentad analysis points to the interval from mid-February to mid-March as the end-of-winter period in which the southwesterlies over the eastern North Atlantic become stronger and the surface-air temperatures in Europe rise markedly, the lapse rate becomes steeper, and concurrently the longitudinal temperature gradient between the Somme (France) and the Oder (Germany/Poland border) is reduced by 0.8 C, that is, by 20% of its 1948 value. Our thesis, that the observed late-winter warming and the corollary advancement of spring in Europe resulted at least in part from stronger southwesterlies over the North Atlantic, merits further investigations.

    Description: Proteins are highly flexible molecules and often exhibit defined conformational changes in response to changes in the ambient temperature. Chicken egg white lysozyme has been previously shown to undergo an apparent structural change when warmed above the tetragonal/orthorhombic phase transition temperature. This is reflected by a change in the habit of the tetragonal and orthorhombic crystals so formed. In this study we show that possible conformational changes induced by heating are stable and apparently non- reversible by simple cooling. Exposure of protein solutions to temperatures above the phase change transition temperature, before combining with precipitant solution to begin crystallization, reduces final crystal numbers. Protein that is briefly warmed to 37 C, then cooled shows no sign of reversal to the unheated nucleation behavior even after storage for 4 weeks at 4 C. The change in nucleation behavior of tetragonal lysozyme crystals, attributed to a structural shift, occurs faster the greater the exposure to temperature above the equi-solubility point for the two phases. Heating for 2 h at 48 C reduces crystal numbers by 20 fold in comparison to the same solution heated for the same time at 30 C. Thermal treatment of solutions is therefore a possible tool to reduce crystal numbers and increase crystal size. The effects of a protein's previous thermal history are now shown to be a potentially critical factor in subsequent macromolecule crystal nucleation and growth studies.

    Description: In the fall of 1997 the Atmospheric Radiation Measurement (ARM program conducted an intensive Observation Period (IOP) to study water vapor at its Southern Great Plains (SGP) site. Among the large number of instruments, four sun-tracking radiometers were present to measure the columnar water vapor (CWV). All four solar radiometers retrieve CWV by measuring solar transmittance in the 0.94-micrometer water vapor absorption band. As one of the steps in the CWV retrievals the aerosol component is subtracted from the total transmittance, in the 0.94-micrometer band. The aerosol optical depth comparisons among the same four radiometers are presented elsewhere. We have used three different methods to retrieve CWV. Without attempting to standardize on the same radiative transfer model and its underlying water vapor spectroscopy we found the CWV to agree within 0.13 cm (rms) for CWV values ranging from 1 to 5 cm. Preliminary results obtained when using the same updated radiative transfer model with updated spectroscopy for all instruments will also be shown. Comparisons to the microwave radiometer results will be included in the comparisons.
