    Notes: Abstract The divalent cation requirements of NOS activity in bovine retina homogenate supernatant were investigated. Supernatants were assayed under standard conditions (in mM: EDTA 0.45, Ca2+ 0.25, Mg2+ 4.0). In order to investigate the enzyme's dependence on divalent cations, the tissue homogenate was depleted of di- and trivalent cations by passing it over a cation-exchange column (Chelex 100). Surprisingly, NOS activity was 50-100% higher in this preparation. However, addition of either EDTA (33 μM) or EGTA (1 mM) almost fully inhibited NOS activity, suggesting a requirement for residual divalent metal cation(s). Phenanthroline or iminodiacetic acid at low concentrations had little effect on activity, suggesting no requirement for Fe2+, Zn2+ or Cu2+. Ca2+ had a moderate stimulatory effect, with an optimum activity around 0.01 mM. Mg2+ or Mn2+ had little effect at concentrations 〈 0.25 mM. However, in the presence of EDTA, Mn2+ or Ca2+ markedly stimulated NOS activity with the optimum at 0.1 mM. At high concentrations (〉 0.1-0.2 mM), all divalent cations tested (Ba2+, Zn2+, Co2+, Mn2+, Mg2+, Ca2+), as well as La3+, dose-dependently inhibited NOS activity. We propose that retinal NOS requires low concentrations of naturally occurring divalent metal ions, most probably Ca2+, for optimal activity and is inhibited by high di- and trivalent metal concentrations, probably by competition with Ca2+.

    Description / Table of Contents: Zusammenfassung Es wird eine IR-spektralphotometrische Bestimmungsmethode für COS in technischen Gasen beschrieben. Bei Verwendung eines Zweistrahlphotometers und Küvettenschichtdicken von 10 cm bzw. 1 m können COS-Konzentrationen zwischen 0,3 und 1200 ppm bestimmt werden. CO2 und Kohlenwasserstoffe stören in den üblichen Konzentrationen die Bestimmung nicht. Der Einfluß des CO wird durch Absorption und Kompensation ausgeschaltet. Im Gebiet oberhalb der Nachweisgrenze werden die Differentialmethode und die Bandenfläche zur Auswertung herangezogen.
    Notes: Summary An IR-spectrophotometric determination of COS in technical gases is described. 0.3 to 1.200 ppm of COS can be determined by means of a double-beam photometer and 10 cm or 1 m-cuvettes. CO2 and hydrocarbons in usual concentrations do not cause interferences. The effect of CO is eliminated by absorption and compensation. In the range above the limit of detection the differential method is employed and the band area is used for evaluation.

    Description / Table of Contents: Zusammenfassung Der Einsatz einer Wärmeleitfähigkeitsmeßzelle zur Wasserbestimmung beim Wasserstoffreduktionsverfahren wird beschrieben. Es lassen sich bei 1 g Einwaage noch 10 ppm Sauerstoff nachweisen. Die Zeit fÜr eine Analyse beträgt nur 5–10 min. Ein weiterer Vorteil ergibt sich durch das Arbeiten in strömendem Wasserstoff unter Wegfall der Feinreinigung, da der Wasserstoff keinen Blindwert liefert. Es kann gleichzeitig neben dem Sauerstoff- auch der Wassergehalt der Metallpulver bestimmt werden. Ein Vergleich mit dem Heißextraktionsverfahren ergab fÜr Molybdän und Wolfram nahezu Übereinstimmung, fÜr Rhenium liefert die H eißextraktionsmethode wahrscheinlich zu niedrige Sauerstoffgehalte. Während das Wasserstoffreduktionsverfahren fÜr Molybdän- und Rheniumpulver gegenÜber der Heißextraktionsmethode zu besser reproduzierbaren Werten fÜhrte, konnte der entsprechende Unterschied bei Wolframpulver nicht gesichert werden. Die Versuche zeigten, daß technisches Rheniumpulver beträchtliche Mengen Wasser enthält.
    Notes: Summary The application of a thermal conductivity cell for the determination of water in the hydrogen-reduction process is described. With a sample weight of 1 g 10 ppm of oxygen can be detected. Only 5–10 min are required for one analysis. A further advantage arises from performing the operation in a current of hydrogen, where the fine purification may be omitted as hydrogen does not supply a blank. Furthermore, it is possible to determine simultaneously with the oxygen also the water content of the metal powders. A comparison with the hot-extracting process has shown a fairly close agreement in the case of molybdenum and tungsten; with rhenium, however, the hot-extracting method probably yields too small values for oxygen. Whereas in the case of molybdenum and rhenium powders the hydrogen-reduction process in comparison to the hot-extracting method was leading to more reproducible values, in the case of tungsten powder the relative differentiation could not be ascertained. The experiments revealed, that commercial rhenium powders are containing appreciable amounts of water.

Aus der Umsetzung des bei der Reduktion gebildeten Wassers mit Calciumhydrid resultiert obendrein eine hohe Empfindlichkeit des Wasserstoffreduktionsverfahrens. Dies bedeutet, da\ — im Gegensatz zu den anderen Varianten des Wasserstoff-reduktionsverfahrens — auch für Sauerstoffgehalte unter 0,01% Probenmengen von etwa 1 g ausreichend sind.
    Notes: Summary A method is described for the determination of oxygen in molybdenum, tungsten, and rhenium by reduction with hydrogen. The water formed is then reacted with calcium hydride to give hydrogen which is determined by gas volumetry. In contrast to the generally applicable hot-extraction the main advantage of the reduction procedure is the less expensive equipment. Moreover, the reaction of the water with calcium hydride results in a high sensitivity of the method. Sample weights of about 1 g are sufficient, also for oxygen contents below 0.01%.

    Notes: Synopsis Cytophotometry of Feulgen-Schiff stained mouse liver cell nuclei revealed DNA that is resistant to prolonged alkaline hydrolysis (5 hr or more at 37°C) with ethanolic barium hydroxide. Thus, in aldehyde-fixed tissue sections an interval of confidence exists between the complete degradation of RNA and the beginning of the extraction of DNA.

    Notes: Synopsis The histochemical demonstration of the carboxyl groups of mucosubstances was studied by two methods at the light and electron microscopic levels. Conditions for activating carbohydrate carboxyl groups were elucidated from which a modified carbodiimide reaction procedure was worked out. With this procedure several acid mucosubstances could be demonstrated; some were characterized as sialomucoproteins. The mechanism of the carbodiimide reaction is discussed. A method is also discussed for increasing the electron opacity of Alcian Blue-stained mucosubstances with a sulphide-silver reaction.

    Notes: Abstract In addition to β-haemolytic streptococci belonging to Lancefield group A (Streptococcus pyogenes, GAS), human isolates of group C (GCS) and group G (GGS) streptococci (S. dysgalactiae subsp. equisimilis) have been implicated as causative agents in outbreaks of purulent pharyngitis, of wound infections and recently also of streptococcal toxic shock-like syndrome. Very little is known about the organisation of the genomic region in which the emm gene of GCS and GGS is located. We have investigated the genome sequences flanking the emm gene in GCS by sequencing neighbouring fragments obtained by inverse PCR. Our sequence data for GCS strains 25287 and H46A revealed two types of arrangement in the emm region, which differ significantly from the known types of mga regulon in GAS. We named this segment of the genome mgrC (for multigene regulon-like segment in group C streptococci). In strains belonging to the first mgrC type (prototype strain 25287) the emm gene is flanked upstream by mgc, a gene that is 61% identical to the mga gene of GAS. A phylogenetic analysis of the deduced protein sequences showed that Mgc is related to Mga proteins of various types of GAS but forms a distinct cluster. Downstream of emm, the mgrC sequence region is bordered by rel. This gene encodes a protein that functions in the synthesis and degradation of guanosine 3′,5′ bipyrophosphate (ppGpp) during the stringent regulatory response to amino acid deprivation. In the second mgrC type (prototype strain H46A), the genes mgc and emm are arranged as in type 1. But an additional ORF (orf) is inserted in opposite orientation between emm and rel. This orf shows sequence homology to cpdB, which is present in various microorganisms and encodes 2′,3′ cyclo-nucleotide 2′-phosphodiesterase. PCR analysis showed that these two mgrC arrangements also exist in GGS. Our sequence and PCR data further showed that both types of mgrC region in GCS and GGS are linked via rel to the streptokinase region characterised recently in strain H46A. A gene encoding C5a peptidase, which is present at the 3′ end of the mga regulon in GAS, was not found in the mgrC region identified in the GCS and GGS strains investigated here.
