    Description: There is growing recognition that mammalian cells produce many thousands of large intergenic transcripts. However, the functional significance of these transcripts has been particularly controversial. Although there are some well-characterized examples, most (〉95%) show little evidence of evolutionary conservation and have been suggested to represent transcriptional noise. Here we report a new approach to identifying large non-coding RNAs using chromatin-state maps to discover discrete transcriptional units intervening known protein-coding loci. Our approach identified approximately 1,600 large multi-exonic RNAs across four mouse cell types. In sharp contrast to previous collections, these large intervening non-coding RNAs (lincRNAs) show strong purifying selection in their genomic loci, exonic sequences and promoter regions, with greater than 95% showing clear evolutionary conservation. We also developed a functional genomics approach that assigns putative functions to each lincRNA, demonstrating a diverse range of roles for lincRNAs in processes from embryonic stem cell pluripotency to cell proliferation. We obtained independent functional validation for the predictions for over 100 lincRNAs, using cell-based assays. In particular, we demonstrate that specific lincRNAs are transcriptionally regulated by key transcription factors in these processes such as p53, NFkappaB, Sox2, Oct4 (also known as Pou5f1) and Nanog.     Description: We report a high-quality draft sequence of the genome of the horse (Equus caballus). The genome is relatively repetitive but has little segmental duplication. Chromosomes appear to have undergone few historical rearrangements: 53% of equine chromosomes show conserved synteny to a single human chromosome. Equine chromosome 11 is shown to have an evolutionary new centromere devoid of centromeric satellite DNA, suggesting that centromeric function may arise before satellite repeat accumulation.     Description: Models of mammalian regulatory networks controlling gene expression have been inferred from genomic data but have largely not been validated. We present an unbiased strategy to systematically perturb candidate regulators and monitor cellular transcriptional responses. We applied this approach to derive regulatory networks that control the transcriptional response of mouse primary dendritic cells to pathogens. Our approach revealed the regulatory functions of 125 transcription factors, chromatin modifiers, and RNA binding proteins, which enabled the construction of a network model consisting of 24 core regulators and 76 fine-tuners that help to explain how pathogen-sensing pathways achieve specificity.     Notes: Underground detonations may produce observable effects in surrounding aquifers and wells. The nature and the duration of the effect at any observation point seem to depend on several factors such as the amount of energy released by the detonation, the geologic environment, the position of the buried explosive device in relation to the saturated zone, aquifer characteristics, and the distance from point of detonation. Precise measurement of these effects in wells presented numerous technical problems and resulted in the development of specialized techniques. Initially, these effects were observed by measuring the fluctuation of the free water surface in wells. The current technique employs high-resolution pressure transducers deep in the water column. Pneumatic packers may be used to restrict the movement of water into the well. Data are recorded on high-speed oscillographs.

    Notes: The role of auxin in the recovery of plant tissue from oxidant treatment was investigated. Treatment of oat coleoptile sections with concentrations of indoleacetic acid (IAA) or 2,4-dichlorophenoxyacetic acid (2,4-D) optimal for normal growth, following pretreatment with moderately inhibiting levels of peroxyacetyl nitrate (PAN) immediately accelerated recovery of growth rate. In some cases inhibition was also less at supraoptimal values of auxin. Treatment of ozonepretreated tissue with IAA or 2,4-D enhanced inhibition at high levels of auxin and produced an optimal growth concentration level which was lower than for sections not given ozone pretreatment. Auxin treatment also reduced the degree of inhibition in fluoride and iodoacetamide-pretreated sections. Mechanisms by which auxin-induced recovery from inhibition may occur are discussed.

    Notes: Conclusions 1. No direct relationship was found between the hardness and the wear resistance of white cast irons. 2. The wear resistance of cast iron increases with the hardness of the carbides. 3. The wear resistance of hypoeutectic chromium cast irons increases linearly with the quantity of carbides if the type of carbide does not change. An increase of the carbon concentration above the eutectic does not increase the wear resistance, since large brittle needles of hypereutectic carbides are formed. 4. The wear resistance decreases with increasing sizes of the carbides. 5. The wear resistance of white cast irons increases linearly with the hardness of the metallic base. The wear resistance is highest when the metallic base is martensitic. In some cases metastable austenite provides an equal wear resistance. Stable austenite reduces the wear resistance.

    Notes: Conclusions 1. Quenching makes it possible to obtain a high wear resistance of white cast irons at a relatively low concentration of alloying elements. 2. An increase of the molybdenum concentration raises the hardenability of white cast irons, while an increase of the carbon and silicon concentrations reduces it. The silicon concentration of cast irons quenched in air should not exceed 0.7%. 3. In castings with sections up to 50 mm an increase of the molybdenum concentration above that required to obtain a martensitic—austenitic structure does not increase the wear resistance of white cast irons.
