    Description: The temperature dependence of the Gilbert damping and magnetic anisotropy are investigated in L 2 1 Co 2 FeAl films epitaxially grown on GaAs (001) substrate by the time resolved magneto-optical Kerr effect. We found that the in-plane biaxial anisotropy increases by more than 90% with the temperature decreasing from 300 K to 80 K, which is mainly due to the strong variation of the magneto-elastic coefficients. In contrast, the intrinsic Gilbert damping rises only about 10%, which is mainly attributed to the reduction of the electron phonon scattering rate, independent of the strain-induced spin-orbit coupling energy.

    Description: T4 RNA ligases are commonly used to attach adapters to RNAs, but large differences in ligation efficiency make detection and quantitation problematic. We developed a ligation selection strategy using random RNAs in combination with high-throughput sequencing to gain insight into the differences in efficiency of ligating pre-adenylated DNA adapters to RNA 3'-ends. After analyzing biases in RNA sequence, secondary structure and RNA-adapter cofold structure, we conclude that T4 RNA ligases do not show significant primary sequence preference in RNA substrates, but are biased against structural features within RNAs and adapters. Specifically, RNAs with less than three unstructured nucleotides at the 3'-end and RNAs that are predicted to cofold with an adapter in unfavorable structures are likely to be poorly ligated. The effect of RNA-adapter cofold structures on ligation is supported by experiments where the ligation efficiency of specific miRNAs was changed by designing adapters to alter cofold structure. In addition, we show that using adapters with randomized regions results in higher ligation efficiency and reduced ligation bias. We propose that using randomized adapters may improve RNA representation in experiments that include a 3'-adapter ligation step.

    Description: Vitamin B12 (VitB12 or cobalamin) is an essential cofactor in several metabolic pathways. Clinically, VitB12 deficiency is associated with pernicious anemia, neurodegenerative disorder, cardiovascular disease and gastrointestinal disease. Although previous genome-wide association studies (GWAS) identified several genes, including FUT2 , CUBN , TCN1 and MUT , that may influence VitB12 levels in European populations, common genetic determinants of VitB12 remain largely unknown, especially in Asian populations. Here we performed a GWAS in 1999 healthy Chinese men and replicated the top findings in an independent Chinese sample with 1496 subjects. We identified four novel genomic loci that were significantly associated with serum level of VitB12 at a genome-wide significance level of 5.00 x 10 –8 . These four loci were MS4A3 (11q12.1; rs2298585; P = 2.64 x 10 –15 ), CLYBL (13q32; rs41281112; P = 9.23 x 10 –10 ), FUT6 (19p13.3; rs3760776; P = 3.68 x 10 –13 ) and 5q32 region (rs10515552; P = 3.94 x 10 –8 ). In addition, we also confirmed the association with the serum level of VitB12 for the previously reported FUT2 gene and identified one novel non-synonymous single-nucleotide polymorphism in FUT2 gene in this Chinese population (19q13.33; rs1047781; P = 3.62 x 10 –36 ). The new loci identified offer new insights into the biochemical pathways involved in determining the serum level of VitB12 and provide opportunities to better delineate the role of VitB12 in health and disease.

    Description: Pseudogenes have been shown to acquire unique regulatory roles from more and more organisms. We report the observation of a cluster of siRNAs derived from pseudogenes of African Trypanosoma brucei using high through-put analysis. We show that these pseudogene-derived siRNAs suppress gene expression through RNA interference. The discovery that siRNAs may originate from pseudogenes and regulate gene expression in a unicellular eukaryote provides insights into the functional roles of pseudogenes and into the origin of noncoding small RNAs.

    Description: A bstract Disc-like fossils from siltstones of the Taozichong Formation (Cambrian) in the Qingzhen area, Guizhou, South China are reported here. They are similar to some Ediacaran and Phanerozoic discoidal fossils, and assigned to Tirasiana ? disciformis ? Palij, 1976 . Based on the study of 43 specimens, dewatering or fluid escape structures, soft-sediment loading, scratch circles or other inorganic origins are ruled out, and the fossil is interpreted as a discoidal body fossil of unknown affinities rather than trace fossils. Energy-dispersive X-ray spectroscopy and elemental mapping analyses reveal that the discoid fossils contain higher concentrations of C, Fe, and P than the surrounding matrix, indicating the possible presence of pyrite, apatite, and organic carbon as a result of authigenic mineralization in association with decay and early diagenetic processes. The possible presence of extracellular polymeric substance suggests that the discs were surrounded by thin microbial mats composed primarily of extracellular polymeric substances, which facilitated their fossilization by promoting conditions that are favorable to secondary mineral precipitation. The new specimens provide useful information about the phylogenetic affinities of these early discoidal fossils and help us to better understand the taphonomic modes of non-biomineralizing organisms in Ediacara-type and Burgess Shale-type biotas.

    Description: Respiratory syncytial virus (RSV) is the leading cause of hospitalization for children under 5 years of age. We sought to engineer a viral antigen that provides greater protection than currently available vaccines and focused on antigenic site O, a metastable site specific to the prefusion state of the RSV fusion (F) glycoprotein, as this site is targeted by extremely potent RSV-neutralizing antibodies. Structure-based design yielded stabilized versions of RSV F that maintained antigenic site O when exposed to extremes of pH, osmolality, and temperature. Six RSV F crystal structures provided atomic-level data on how introduced cysteine residues and filled hydrophobic cavities improved stability.     Description: The emergence of the H7N9 influenza virus in humans in Eastern China has raised concerns that a new influenza pandemic could occur. Here, we used a ferret model to evaluate the infectivity and transmissibility of A/Shanghai/2/2013 (SH2), a human H7N9 virus isolate. This virus replicated in the upper and lower respiratory tracts of the ferrets and was shed at high titers for 6 to 7 days, with ferrets showing relatively mild clinical signs. SH2 was efficiently transmitted between ferrets via direct contact, but less efficiently by airborne exposure. Pigs were productively infected by SH2 and shed virus for 6 days but were unable to transmit the virus to naive pigs or ferrets.     Description: The nucleation process is crucial to many phase transitions, but its kinetics are difficult to predict and measure. We superheated and melted the interior of thermal-sensitive colloidal crystals and investigated by means of video microscopy the homogeneous melting at single-particle resolution. The observed nucleation precursor was local particle-exchange loops surrounded by particles with large displacement amplitudes rather than any defects. The critical size, incubation time, and shape and size evolutions of the nucleus were measured. They deviate from the classical nucleation theory under strong superheating, mainly because of the coalescence of nuclei.
