    Notes: An orthogonality theorem is obtained for square integrable representations on homogeneous spaces of a locally compact group. From this, a series of lemmas is derived showing the informational completeness of natural covariant localization operators, as well as of the generalized Wigner distributions (matrix elements of the group). Some of these results give explicit reconstruction formulas for the quantum state from its expectation values against these families of operators. The results are applied to special (phase space) representations of the Heisenberg, affine, and Galilei groups. © 1995 American Institute of Physics.

    Notes: Abstract There are many medical applications which benefit from the use of soluble biomaterials, including the sustained release of drugs over a precise period of time, or temporary conduits for controlling nerve regrowth. We have manufactured a series of phosphate-based controlled release glasses (CRGs) in which the solubility could be controlled by varying the concentration of CaO and Na2O. Fibres of the CRG containing iron and cerium were placed into direct contact with human neutrophils and macrophages in tissue culture for 2.5 and 24 h respectively and the responses analysed by scanning electron microscopy (SEM) and confocal microscopy. The supernatants were analysed for the cytokine IL-1β by enzyme-linked immunosorbent assay (ELISA). Disks of CRG of various compositions were placed in contact with whole blood for 30 min and platelet adhesion assessed by SEM. Activation of platelets, granulocytes and complement were quantified by ELISA for β-thromboglobulin, elastase and iC3b. Intrinsic coagulation activation was measured by timing the clotting of recalcified plasma. Only the cerium fibre inhibited IL-1β release from macrophages. No platelet adhesion was observed to any disk composition. Three compositions containing MgO inhibited plasma clotting and showed an insignificant level of complement activation. This study has demonstrated the development of a number of compositions of CRG, which have great potential in a wide variety of biomedical applications.

    Notes: Wound infections retard healing rates and can lengthen the time a patient is required to spend in hospital. Obviously it is desirable to minimize or remove pathogenic organisms to allow wound healing to progress optimally. At present there are a large range of topical applications available in the form of antibiotics or antiseptics, but the toxic effects of these agents may outweight the benefits. In this study two conventional antiseptic agents were examined together with a range of phosphate-based glasses which were designed to release metal ions into an aqueous environment. As a control a series of standard metal ion solutions were prepared and examined in the same test system. The metal ions examined are considered important in the wound healing environment either for their potential bacteriostatic effects or indeed their potential for increasing cellular activity. This part of the study examined the effects of these agents on mammalian cells in vitro, using a standard MTT assay to assess the cytotoxicity or activity of cultured mouse fibroblasts. It was found that chlorhexidene, a commonly used antiseptic agent, was toxic at low levels. The controlled release glasses, in most cases, increased the rate of cell activity before reaching toxic levels. This effect was also found with the standard metal solutions.

    Notes: Abstract Vesicoureteral reflux and urinary incontinence have previously been treated by various means including the endoscopic delivery of injectable bulking materials such as silicone micro-implants, PTFE implants, glass particles, fat and bovine collagen. These first three materials do not degrade and collagen requires frequently repeated injections in order to sustain the restored continence provided. Vesicoureteric reflux in children usually resolves independently before the age of five. Correction is required before this, because treatment by prophylactic antibiotics is frequently unsuccessful in preventing breakthrough infection. The ideal material for injection should have large particles to avoid migration, inject easily and controllably, be non-toxic and dissolve over the period of time by which time the kidney will be mature. Three different controlled-release glass (CRG) granule compositions have been prepared by Giltech Ltd, and suspended in a suitable carrier medium (in this case glycerol). The degradable glasses, which have two different size ranges of 200–300 and 〈53 μm, and three different solution rates, were injected intramuscularly into the dorso-lumbar region of rats. Histological analysis of cryostat cut section after time periods of 2 d, 4 and 9 wk, and 6 mon has been performed. Histology sections were stained for neutrophils and macrophages using enzyme histochemistry. ED1 (monocytes and immature macrophages), ED2 (mature tissue macrophages), CD4 (helper/inducer T-lymphocytes and macrophages), CD8 (suppresser/cytotoxic T-lymphocytes), Interleukin-1β, IL-2 (activated T-lymphocytes), Major Histocompatibility Complex (MHC) class II (activated macrophages and activated B-lymphocytes), α–β (T-lymphocytes) and CD45RA (B lymphocytes) antibodies have beed used to stain immunohistochemically each sample. This study demonstrates that particulate, degrading glass is stimulating an inflammatory response in soft tissue at time periods up to 6 mon. It should be noted that very small particulate, fast degrading glass is leading to tissue necrosis and should not be considered further for these applications. However, larger particulate, slower degrading materials are demonstrating effective potential for stress incontinence applications. © 1998 Kluwer Academic Publishers

    Description: Rapidly developing methods of digital acquisition, visualization and analysis allow highly detailed outcrop models to be constructed, and used as analogues to provide quantitative information about sedimentological and structural architectures from reservoir to subseismic scales of observation. Terrestrial laser-scanning (lidar) and high precision Real-Time Kinematic GPS are key survey technologies for data acquisition. 3D visualization facilities are used when analysing the outcrop data. Analysis of laser-scan data involves picking of the point-cloud to derive interpolated stratigraphic and structural surfaces. The resultant data can be used as input for object-based models, or can be cellularized and upscaled for use in grid-based reservoir modelling. Outcrop data can also be used to calibrate numerical models of geological processes such as the development and growth of folds, and the initiation and propagation of fractures.

    Description: The G2019S leucine rich repeat kinase 2 (LRRK2) mutation is the most common genetic cause of Parkinson's disease (PD), clinically and pathologically indistinguishable from idiopathic PD. Mitochondrial abnormalities are a common feature in PD pathogenesis and we have investigated the impact of G2019S mutant LRRK2 expression on mitochondrial bioenergetics. LRRK2 protein expression was detected in fibroblasts and lymphoblasts at levels higher than those observed in the mouse brain. The presence of G2019S LRRK2 mutation did not influence LRRK2 expression in fibroblasts. However, the expression of the G2019S LRRK2 mutation in both fibroblast and neuroblastoma cells was associated with mitochondrial uncoupling. This was characterized by decreased mitochondrial membrane potential and increased oxygen utilization under basal and oligomycin-inhibited conditions. This resulted in a decrease in cellular ATP levels consistent with compromised cellular function. This uncoupling of mitochondrial oxidative phosphorylation was associated with a cell-specific increase in uncoupling protein (UCP) 2 and 4 expression. Restoration of mitochondrial membrane potential by the UCP inhibitor genipin confirmed the role of UCPs in this mechanism. The G2019S LRRK2-induced mitochondrial uncoupling and UCP4 mRNA up-regulation were LRRK2 kinase-dependent, whereas endogenous LRRK2 levels were required for constitutive UCP expression. We propose that normal mitochondrial function was deregulated by the expression of G2019S LRRK2 in a kinase-dependent mechanism that is a modification of the normal LRRK2 function, and this leads to the vulnerability of selected neuronal populations in PD.

    Description: Porosity and permeability are notoriously difficult to predict in carbonates, especially prior to drilling when there is a lack of direct petrophysical data. The aim of this paper is to document the initial results of an integrated outcrop and laboratory study designed to investigate the relationships between pore systems and acoustic velocities in faulted Oligo-Miocene carbonates on the Mediterranean islands of Malta and Gozo. Depositional facies is shown to have a significant effect, with velocities in grain-dominated carbonates up to 1000 m s –1 higher than those in micrite-dominated carbonates. Based on outcrop structural data, the fault zones can be separated into three architectural components: a fault core; an intensely damaged zone; and a weakly damaged zone, with the last passing into undamaged protolith. Our data suggest that only the fault core component can be identified using porosity–velocity data, with P-wave velocity ( V p ) values of 5000–6500 m s –1 at helium porosities of less than 5%. Our study is novel in that the prediction of elastic properties and acoustic velocities across fault zones is anticipated by linking laboratory-scale measurements with seismic-scale predictions through quantitative rock physics modelling.
