    Notes: Abstract This paper describes the design and analysis of a research testbed developed to study the control of manipulator-coupled spacecraft with independent attitude control systems. This scenario could present itself in the assembly of Space Station Freedom (SSF). SSF assembly calls for a rendezvous of the Space Shuttle (SS) with SSF. Part of the assembly process requires that both spacecraft be coupled via the Space Shuttle Remote Manipulator System. An additional criterion that poses increased complexity is that the Space Shuttle controls and Space Station controls can not communicate. The technical issue involved is unwanted vibrations of the coupled-configuration that occur retraction and the complications due to non-interacting control systems. To understand these vibrations and possible complications, a research testbed has been built at the Marshall Space Flight Center in Huntsville, AL. To build the testbed, the manipulator links joints, and vehicles that represent the Space Shuttle and Space Station had to be designed. Pre-design simulation studies using ANSYS [1] (a Finite Element Computer Code) is used to size and design the manipulator links for the experimental facility. The ANSYS results were verified by the development of the Lagrangian Equations of motion. The Harmonic drives used as joints for the two link, three joint manipulator have been dissected into free body diagrams to ensure proper load paths in the ANSYS models. Accurate simulation of manipulator-coupled spacecraft is an important technology for NASA to understand. This paper outlines the methodology behind the preliminary design of a research testbed developed to help NASA gain knowledge in this area.

    Notes: Abstract The redundant robots analyzed here have large numbers of degrees of freedom. Such robots are analogous to snake or tentacles and are useful for operation in highly constrained environments and novel forms of locomotion — here it has been defined as critical manipulation. In this paper the end-point deflection of hyper-redundant robots have been studied by parameterization of the flexible links of the robot manipulator. A number of planar configurations of the redundant manipulator were taken into consideration for the present study and the configuration which gives minimum end-point deflection has been recommended for the critical manipulation. This work is significant for some of the research activities recently going on in NASA's Kennedy Space Center towards the application of hyper-redundant robots for manipulation inside the space Shuttle Cargo Bay.

    Notes: Abstract  The proteasome is a large multicatalytic proteinase that plays a role in the generation of peptides for presentation by major histocompatibility complex class I molecules. The 20S proteolytic core of mammalian proteasomes is assembled from a group of 17 protein subunits that generate a distinctive pattern of spots upon two-dimensional gel electrophoresis. The genes for most of these subunits have been cloned from humans and rats. We isolated cDNA clones for the mouse orthologues of ten of the subunits [PSMA1 (C2), PSMA2 (C3), PSMA3 (C8), PSMA4 (C9), PSMA5 (ZETA), PSMA6 (IOTA), PSMA7 (C6-I), PSMB2 (C7-I), PSMB3 (C10-II), and PSMB5 (X)] to complete the cloning of all of the mouse subunits. Using antisera raised against these subunits or their orthologues, we verified the identity of these proteins by two-dimensional NEPHGE-PAGE.

    Notes: Abstract The X-ray crystal structure of the trans-diaqua complex [VO(DPA)(H2O2)]·2H2O (1) (DPA=dipicolinate dianion) has been determined. Comparison with the known structure of [VO(DPA)(o-phen)]·3H2O (2), obtained from (1) by displacement of the two coordinated aqua molecules, shows that the coordination sphere around vanadium is reorganised during this reaction.

    Notes: Abstract A soluble protein phosphatase from the promastigote form of the parasitic protozoanLeishmania donovani was partially purified using Sephadex G-100, DEAE-cellulose and again Sephadex G-100 columns. The partially purified enzyme showed a native molecular weight of about 42, 000 in both Sephadex G-100 and sucrose density gradient centrifugation. The sedimentation constant, stokes radius and frictional ratio were found to be 3.43S, 2.8 nm and 1.20 respectively. The enzyme preferentially utilized phosphohistone as the best exogenous substrate. Mg2+ ions were essential for enzyme activity; among other metal ions Mn2+ can replace Mg2+ to a certain extent whereas Ca2+, Co2+ and Zn2+ could not substitute for Mg2+. The pH optimum of the enzyme was 6.5–7.5 and the temperature optimum 37°C. The apparent Km for phosphohistone was 7.14 μM. ATP, ADP, inorganic phosphate and pyrophosphate had inhibitory effect on the enzyme activity whereas no inhibition was observed with sodium tartrate and okadaic acid. These results suggest thatL. donovani promastigotes possess a protein phosphatase which has similar characteristics with the mammalian protein phosphatase 2C.

    Notes: Abstract Mullite, an extremely useful ceramic material, is doped with transition metal ions. The changes in the electronic properties of these doped materials have been studied by positron annihilation lifetime spectroscopy as well as Doppler broadened line shape analysis. The results on the positron annihilation parameters are characteristic of ionic size, oxidation state and the “d”-electron configuration of the respective transition metals doped in the parent lattice of the mullite. These results, along with the resistivity measurements are suggestive of transition of the parent mullite from an insulator to a semimetal in the modified structure.

    Notes: Abstract  To facilitate construction of physical map of the rice genome, a bacterial artificial chromosome (BAC) library of IR64 genomic DNA was constructed. It consists of 18 432 clones and contains 3.28 rice genomic equivalents. The insert size ranged from 37 to 364 kb with an average of 107 kb. We used 31 RFLP markers on chromosome 4 to screen the library by colony hybridization. Sixty eight positive clones were identified with 2.2 positive clones per RFLP marker. The positive clones were analyzed to generate 29 contigs whose sizes ranged from 50 to 384 kb with an average of 145.6 kb. Chromosome walking was initiated for ten contigs linked to resistance genes. Thirty eight BAC clones were obtained and two contigs were integrated. Altogether, they covered 5.65 Mb (15.1%) of chromosome 4. These contigs may be used as landmarks for physical mapping of chromosome 4, and as starting points for chromosome walking towards the map-based cloning of disease resistance genes which were located nearby.

    Notes: Abstract We exploited the newly developed amplified fragment length polymorphism (AFLP) technique to study the polymorphism, distribution and inheritance of AFLP markers with a doubled haploid rice population derived from ‘IR64’/‘Azucena’. Using only 20 pairs of primer combinations, we detected 945 AFLP bands of which 208 were polymorphic. All 208 AFLP markers were mapped and distributed over all 12 chromosomes. When these were compared with RFLP markers already mapped in the population, we found the AFLP markers to be highly polymorphic in rice and to follow Mendelian segregation. As linkage map of rice can be generated rapidly with AFLP markers they will be very useful for marker-assisted backcrossing.

    Notes: Abstract  The cytoplasmic genetic male-sterile (CMS) lines developed at the International Rice Research Institute are valuable in producing tropical rice hybrids. Efficient use of CMS lines in hybrid rice production will depend on their level of genetic diversity. Aside from morphological characterization, molecular analysis based on DNA markers can provide information on the genetic diversity of the germplasm. The Amplified Fragment Length Polymorphism (AFLP) technique was used to fingerprint 71 CMS lines and four rice cultivars, ‘IR64’, ‘Azucena’, ‘IR74’, and ‘FR13A’. Eleven primer pair combinations specific to the enzymes PstI and MseI were used to generate 530 AFLP markers, 176 of which were polymorphic. Each CMS line revealed a distinct fingerprint. The AFLP marker-based dendrogram depicted genetic variation among the CMS lines. The CMS lines developed in japonica background grouped with ‘Azucena’, a japonica cultivar. None of the CMS lines clustered with ‘FR13A’, a flood-tolerant traditional indica variety. ‘IR64’ was found to be distinct from the other indica CMS lines and clustered with lines developed in its background. The grouping of CMS lines into a few groups is useful for breeders in selecting genetically diverse CMS lines for hybrid rice production and in avoiding test crossing every CMS line empirically. This study demonstrated that AFLP is a powerful and reliable tool in determining the genetic relationships and in producing distinct fingerprints of rice cultivars.

    Notes: Abstract  The genus Oryza to which cultivated rice belongs has 22 wild species. Seventy-seven accessions of 23 Oryza species, five related genera, and three outgroup taxa were fingerprinted using amplified fragment length polymorphism (AFLP). A total of 1191 polymorphic markers were obtained using five AFLP primer combinations. AFLP data were analyzed to study species relationships using different clustering algorithms, and the resulting phenograms were tested for stability and robustness. The findings suggest a common ancestry to the genus Oryza. Moreover, the results demonstrate that: (1) evolution in Oryza has followed a polyphyletic path wherein multiple lineages underwent independent divergence after separation early in the evolution from a common ancestor/pool of related taxa; (2) newly assigned genomes, GG for O. meyeriana and HHJJ for O. ridleyi complexes, are among the most diverged in the genus; (3) CCDD tetraploids have a relatively ancient origin among the Officinalis complex; (4) O. malampuzhaensis, O. indandamanica, O. alta, and O. grandiglumis are diverged enough to deserve species status; (5) O. officinalis and O. eichingeri (CC) are putative progenitors of O. minuta * O. malampuzhaensis and tetraploid O. punctata, respectively, (6) O. brachyantha is most diverged species in the genus. AFLP is reliable molecular technique and provides one of the most informative approaches to ascertain genetic relationships in Oryza, which may also be true for other related species/organisms.
